Article The Effects of Streptozotocin on Spinal Cord Motor Neurons Count in Rat: a Stereological Study(Yuzuncu Yil Universitesi Tip Fakultesi, 2019) Rahım, O.O.; Çölçimen, N.; Rağbetli, M.Ç.In this study we investigated effects of Streptozotocin (STZ) administered at different doses on the total number of neurons in segment Thoracic 11 (T11) of the spinal cord of adult rats by using stereological methods. Twenty-eight adult male Wistar albino rats were used. The rats were divided into four groups; control group, saline group (0.5 ml of 0.9% NaCl i.p.), 45mg/kg of STZ (i.p.) group and 65 mg/kg of STZ (i.p) exposed group. At the seventh day, the rats were perfused with 10% neutral buffered formaldehyde. T11, which is one of the transverse spinal cord segments, was isolated and processed for routine histologic examination. Semithin sections (0.5µm thick) were obtained using a ultramicrotome according to systematic random sampling strategies. The sections were stained with Toluidine Blue with Borax. The motor neurons were counted unbiased stereological disector/Cavalieri combination method. The groups were compared statistically. In terms of motor neuron number, no significant difference among the groups was found (p>0.05). Our results indicated that exposure to different doses of Streptozotocin has no effect on the T11 segment of the spinal cord total motor neuron number of the adult rats. © 2019, Yuzuncu Yil Universitesi Tip Fakultesi. All rights reserved.Article Evaluation of the Effects of Alcohol on the Heart by Stereological Methods(Yuzuncu Yil Universitesi Tip Fakultesi, 2020) Çölçimen, N.Alcohol is an agent with known toxic effects on body organs and tissues. It injures heart tissue, causing arrhythmia, left ventricular hypertrophy, and sudden death. In the present study we aimed to determine ethanol’s detrimental cardiac effects using the stereological technique. We enrolled 13 male Wistar albino rats. They were randomized into two groups as the Control (n=6) and Ethanol (n=7) groups. The ethanol group was orally administered 6.4% ethanol mixed with drinking water. The control group was subjected to no application. These procedures were continued for 1 8 days. At the end of the experiment the cardiac tissue was excised under anesthesia and the left ventricle was dissected. It was then subjected to the routine light microscopic histological examination procedure and embedded in paraffin. Sections of 5µm t hick were taken and stained with Hematoxylin & Eosin. In the stereological measurement the modified method of the Cavalieri principle was used. We detected the volume of left ventricular wall increase by the stereological technique, supporting the view tha t alcohol forms left ventricular hypertrophy. © 2020, Yuzuncu Yil Universitesi Tip Fakultesi. All rights reserved.Article Inter-Observer Variability of Radiologists and Gynecologists in Hysterosalpingogram Evaluation(Ortadog u Reklam Tanitim Yayincilik Turizm Egitim Insaat Sanayi ve Ticaret A.S., 2016) Güler Okyay, A.; Öner, G.; Çölçimen, N.; Turan, A.; Beyazal Çeliker, F.Objective: Reading of hysterosalpingography (HSG) films is important for management of patients. The aim of this study was to evaluate and compare HSG interpretation of radiologists and clinicians. Material and Methods: Two clinicians and 2 radiologists, who were 35-43 years old and were fulfilled 5 years in their speciality, evaluated 116 hysterosalpingograms. HSG pictures were viewed at computer monitor and the observers were asked to evaluate them within a standard framework consisted of several questions for diagnosis of uterine and tubal disease. The consistency of each individual reader, the reliability of detecting specific abnormalities, and the consistency of clinicians compared with radiologists was measured. Results: There were statistically significant differences in the consistency of interpretations for contour of uterine cavity, uterine deviation, and uterine filling defect (p<0.05). Evaluation of uterine anomalies with HSG were similar between clinicians and radiologists (p>0.05). Although ratios of hydrosalphinx were similar between clinicians and radiologists (p>0.05), there were differences for the evaluation of bilateral tubal contrast falling (p<0.05). Conclusion: Gynecologists have more inter-observer variability than radiologists in hysterosalpingography evaluation. However, both clinicians and radiologists were compatible within themselves. Compatibility of radiologists was higher than that of clinicians. Better designed studies are needed in order to confirm the variability of HSG reports and to answer the question of "who should read HSGs of infertile women?. © Copyright 2016 by Türkiye Klinikleri.Article Investigation of the Effects of Diclofenac Sodium in Rat Ovary on the Number of Preantral Follicles by Stereological Methods in Prenatal Period(Yuzuncu Yil Universitesi Tip Fakultesi, 2017) Çölçimen, N.; Rağbetli, M.Ç.; Kara, M.; Arıhan, O.; Akyol, V.For healthy future generations pregnancy is a period in which maximum amount of attention should be given. In this period, choice, duration and dosage of medication as well as the changing physiology of the mother should be taken into account. Non-steroidal anti-inflammatory drugs (NSAIDs) are a group of drugs that have analgesic, anti-pyretic and antiinflammatory effects. This group of drugs can be used in the treatment of the ongoing rheumatic diseases or which occur during the pregnancy period and the complications that occur due to pregnancy. We aimed to show how it affects the number of ovarium preantral follicles of newborn fetus on rats by using Diclofenac Sodium which belongs to this drug family group. Diclofenac Sodium (1 mg/ kg, IM) was injected for 15 days from the 5th day of pregnancy to pregnant rats in Diclofenac Sodium group and saline (1 ml/ kg, IM) was injected to the pregnant rats of the sham group at the same time period. In the 4th week after birth, ovarian tissue preparates of rat pups were stained with hematoxylin-eosin and ovarian total tissue volumes and preantral follicle counts were evaluated under light microscope. There was no statistically significant difference between groups for ovarium total tissue volumes and preantral follicle numbers (p>0.05). Lowest value of follicular intensity was observed in Diclofenac sodium group among study groups however no statistically important difference was found (p>0.05). Results obtained in our study may be related with administration of drug in not a high dosage (1mg/kg). All rights reserved.Article The Stereological Ratio of Stromal/Parenchymal Components in Postnatal Prostate Tissue of Rats Prenatally Treated With Diclofenac Sodium(2013) Kara, M.; Raǧbetli, C.; Koyun, N.; Çölçimen, N.; Raǧbetli, M.Ç.Since diclofenac sodium extends the pregnancy and affects the fetus, our study aimed to stereologically investigate the components of the rat prostate with prenatally treated with diclofenac sodium. The rats were left overnight to mate. Pregnant animals were kept in a room tempreture with normal light and dark cycles and fed with normal diet. After injections to sham and treated groups for 15 days, 18 male offsprings were housed in three groups (6 placebo; 6 control; 6 treated). The 20 weeks-old animals were perfused and the prostates removed and embedded in paraffin. Tissue sections at 5μm thickness were taken at regular intervals and stained with Hematoxylin-eosin and Masson trichrome. Cavalieri principle was used for volume fractions analysis. Stereological examination revealed 55% acinar epitelium and 45% connective tissue in both control and placebo groups, as well as 60% acinar epitelium and 40% connective tissue in the treated group. Prenatally applied diclofenac sodium did not cause a statistically significant change in the prostate stroma-parenchymal ratio in 20-week old rats compared to the control animals (p>0.05). © 2013 OMU.
