Article Determination of Adenosine Deaminase Activity in Cattle Naturally Infected With Theileria Annulata(Springer, 2008) Altug, Nuri; Yuksek, Nazmi; Agaoglu, Zahid Tevfik; Keles, IhsanThe purpose of this study was to determine serum ADA activity in cattle naturally infected with Theileria annulata. In this study, a total of 37 cross-bred cattle which 27 of it showing clinical signs of theileriosis constituted infected group and 10 healthy cattle as control group were used as animal materials. Infected group divided into three groups according to their PCV values. Cattle with PCV >= 25 were put on group I (n=9), those with PCV 13-24 were put on group II (n=11) and those with PCV >= 12 were put on group III (n=7). Microscopical diagnosis of the disease was also made. Hematological parameters, serum enzyme activities (ADA, AST, ALT and ALP) were determined in all cattle. Hematological results revealed that significant progressive decreases in HGB, PLT, PBML counts and ratios from group I onwards to group III, whereas the WBC, PBPL counts and ratios showed an increase from group I onwards to group III. The serum ADA, AST, ALT and ALP activity increased significantly in all infected groups compared to control group. However, these parameters were also observed to decrease progressively from group I to group III. Furthermore, the highest increase in enzyme activities observed in the infected group I. But, these enzyme's activities started to decrease in infected group II and III in parallel with PBML and PLT counts. Eventhough, this decrease did not reach to the values obtained from control group. On the contrary, PBPL counts and ratios increased in infected group II and III in contrast to decrease in PCV. As a result, increased serum ADA activity in tropical theileriosis may reflect the involvement of the cellular immune responses.Article Genetic Structure and Variation of Van Cats(Springer/plenum Publishers, 2011) Altunok, Vahdettin; Yuksek, Nazmi; Berkman, Ceren C.; Agaoglu, Zahid Tevfik; Togan, InciTo determine the genetic structure and variation of Van cats and some other cats, seven enzyme loci were examined using horizontal starch gel electrophoresis. ME bands were observed for the first time in cats. For the enzyme loci CA (1) , SOD, GPI, and GOT, neither the individual Van cats nor the specimens of other cat species exhibited any variation. These enzymes presented identical bands, all of which were homozygous. With respect to the PGD, ME, and ESD loci, however, genetic variation was observed in all of the cats. Hence, three of the seven gene-enzyme systems (43%) were polymorphic with two alleles, contributing to an estimate of average heterozygosity of 0.33-0.49 for the Van cats. PGD was the most discriminatory among the three polymorphic loci. The phylogenetic tree indicated that the Van, Persian, Turkish Angora, and Turkish Tekir cats are distinct from Siamese and Bombay cats.Article Immunostimulatory Effects of Aloe Vera and Β-Glucan on Cellular and Humoral Immune Responses Following Vaccination With Polyvalent Vaccines in Dogs(Kafkas Univ, veteriner Fakultesi dergisi, 2010) Altug, Nuri; Yuksek, Nazmi; Agaoglu, Zahid TevfikThe aim of this study was to evaluate the effects of Aloe vera and beta-Glucan on lymphocyte subsets, haematological parameters and immunglobulin concentrations following vaccination in dogs. For this purpose, totally 20 street dogs were used. The animals were allocated into five groups. Group 1 consisted of only polyvalent vaccine applied dogs. Group 2 and 3 were only Aloe vera and beta-Glucan applied dogs, respectively. Group 4 and 5 were consisted of polyvalent vaccine in addition to Aloe vera and beta-Glucan applied dogs, respectively. Blood samples were collected before vaccination and after vaccination (AV). Although platelet (PLT) counts in group 1 decreased AV, in all other groups (group 2, 3, 4, 5) it increased. White blood cell (WBC) and peripheral blood mononuclear lymphocyte (PBML) counts (in all groups), peripheral blood polymorphonuclear lymphocyte (PBPL) counts (group 1, 2, 4, 5), neutrophil (group 1, 4, 5), monocyte (group 2, 3, 5), packed cell volume (PCV) ratios (group 2 and 3) and haemoglobin (HGB) concentrations (group 3) increased AV. But, lymphocyte ratios in all groups did not change AV. CD8 T lymphocyte ratios in all groups increased at 7(th) and 14(th) days AV. Although CD3 T lymphocyte ratios in group 1 increased at 7(th) day AV, CD4 T and B lymphocytes ratios did not change AV period. But, both CD3 and CD4 T lymphocytes ratios in all other groups increased at 7th and 14th days AV. Moreover, B lymphocytes ratios in all groups except for group 1 increased AV. On the other hand, B lymphocytes ratios in group 2 and 3 increased only 7(th) day AV, while this increase in group 4 and 5 were present on both 7(th) and 14th days AV. Serum IgM concentrations in group 1 increased only 7(th) day AV, whereas its levels did not change in group 2 and 3. Serum IgG concentrations in group 1, 2 and 3 increased only 14(th) day AV. But, serum IgM and IgG concentrations in group 4 and 5 increased at 7(th) and 14(th) days AV. Administration of Aloe vera and beta-Glucan after vaccination in dogs may stimulate both cellular and humoral immun responses. Additionally, it might have restorative effect on PLT counts.Article Investigation on the Relationship Between Lymphocyte Subsets, Immunoglobulin Levels and Adenosine Deaminase Activities in Immunosuppressive Dose Methylprednisolone Treated Dogs(Natl veterinary Research inst, 2007) Altug, Nuri; Agaoglu, Zahid TevfikSeven healthy dogs were administered with methylprednisolone orally (2 mg/kg body weight/d) at the beginning of the study (between 0-10 d), then the dosing was decreased at 10 d intervals throughout 10 to 60 d in an alternate-day therapy programme. Drug administration to the dogs was stopped on the 60 d. Total leukocyte count; polymorphonuclear leukocytes, and neutrophil ratios increased and eosinophil, mononuclear leukocytes, lymphocyte, and monocyte ratios were decrease when these values were compared to changes between days. Furthermore, in lymphocyte subsets, B lymphocytes and CD 8 T lymphocytes ratios were found to decrease. In addition, CD 3 T lymphocyte ratios decreased on the 61 d and CD 4 T lymphocyte ratios increased on the 21, 41, 51 and 61 d. IgM and IgG levels decreased between the days. Serum total ADA and ADA 1 activities decreased during the application, but ADA 2 activity did not change significantly. As a result, after immunosuppressive dose of methylprednisolone application, reduction in the ADA and ADA 1 activity can be used as determinant of the suppression of immune system components.Article Rupture of the Gastrocnemius Muscle in a Cow Two Months After Twin Birth(Natl veterinary Research inst, 2007) Altug, Nuri; Ozkan, Cumali; Yuksek, Nazmi; Karasu, Abdullah; Keles, Ihsan; Agaoglu, Zahid Tevfik; Ilhan, FatmaThe rupture of the gastrocnemius muscle was described in a cow who gave a twin birth. A tentative diagnosis of complete rupture of the muscle was based on information obtained from both anamnesis and clinical signs. The definitive diagnosis was made on necropsy. The role of serum minerals, especially Ca, changes in the muscle originated enzymes, and other laboratory parameters together with clinical examination findings was discussed.
