Article Anti-Mutagenic and Anti-Oxidant Potencies of Cetraria Aculeata (Schreb.) Fr., Cladonia Chlorophaea (Florke Ex Sommerf.) Spreng. and Cetrelia Olivetorum (Nyl.) W.l. Culb. & C.f. Culb.)(Shaheed Beheshti Univ, Sch Pharmacy, 2018) Ceker, Selcuk; Orhan, Furkan; Sezen, Selma; Gulluce, Medine; Ozkan, Hakan; Aslan, Ali; Agar, GulerayIn this study, the mutagenic and anti-mutagenic effects of methanol extract of three lichen species (Cetraria aculeata, Cladonia chlorophaea and Cetrelia olivetorum) were investigated by using E. coli-WP2, Ames-Salmonella (TA1535 and TA1537) and sister chromatid exchange (SCE) test systems. The results obtained from bacterial test systems demonstrated that methanol extracts of three lichen species have strong anti-mutagenic potencies on TA1535, TA1537 strains and to a lesser extent on E. coli-WP2 strain. The anti-oxidant level of human lymphocytes cells was determined in order to clarify the mechanism underlying the anti-mutagenic effects of these lichen species. Co-treatments of 5, 10 and 20 mu g/mL concentrations of these three lichen species with AFB decreased the frequencies of SCE and the level of MDA and increased the amount of SOD, GSH and GPx which decreased by aflatoxin. The findings of this work have clearly demonstrated that Cetraria aculeata, Cladonia chlorophaea and Cetrelia olivetorum have significant anti-mutagenic effects which are thought to be partly due to the anti-oxidant activities and the interaction capability of lichen extracts with mutagen agents (Sodium azide, acridin, N-methyl-N'-nitro-N-nitrosoguanidine and aflatoxin B-1).Article Phytohormone Levels in Germinating Seeds of Zea Mays L. Exposed To Selenium and Aflatoxines(Springer, 2006) Agar, Guleray; Turker, Musa; Battal, Peyami; Erez, M. EmreSeeds of Zea mays L. were exposed to allatoxine B1 (AFB1), aflatoxine G1 (AFG1) and selenium (Se) alone and in combination and allowed to germinate. Phytohormone levels of GA-like substances (GAs), trans-Zeatin (t-Z) and Indole-3-acetic acid (IAA) were determined by High Performance Liquid Chromatography (HPLC) when the roots of the germinating seeds reach 1.5-3.0 cm in length. The levels of endogenous hormones decreased in seeds treated with AFB1 and AFG1 compared to control; however an increase was noted in seeds exposed to AFG1 and Se together. AFB1 and Se treatment caused reduced hormone levels in most of the treatments. When plants were exposed to Se alone, the highest levels of GAs, t-Z and IAA were observed in the application of 800 ppm Se. The highest levels of GAs, t-Z and IAA were observed when seeds were treated with 0.2 ppm AFG1 + 8 ppm Se, 0.2 ppm AFG1 + 8 ppm Se and 0.2 ppm AFG1 + 0.08 ppm Se, respectively, whereas the lowest levels of the hormones were observed in 0.2 ppm AFB1 + 8 ppm Se, 0.2 ppm AFB1 + 0.08 ppm Se and 0.1 ppm AFB1, respectively. In conclusion, the levels of phytohormones were reduced by the treatment of AFB1 and AFG1 alone. However Se removed the negative effect of AFB1 on phytohormones, but not AFB1.Article Phytotoxical Effect of Lepidium Draba L. Extracts on the Germination and Growth of Monocot (Zea Mays L.) and Dicot (Amaranthus Retroflexus L.) Seeds(Sage Publications inc, 2015) Kaya, Yusuf; Aksakal, Ozkan; Sunar, Serap; Erturk, Filiz Aygun; Bozari, Sedat; Agar, Guleray; Battal, PeyamiLaboratory experiments were performed to determine phytotoxic potentials of white top (Lepidium draba) methanol extracts (root, stem and leaf) on germination and early growth of corn (Zea mays) and redroot pigweed (Amaranthus retroflexus). Furthermore, the effects of different methanol extracts of L. draba on the phytohormone (indole-3-acetic acid (IAA), gibberellic acid (GA), abscisic acid (ABA) and zeatin) levels of corn and redroot pigweed were investigated. It was observed that all concentrations of methanol extracts of root, stem and leaf of L draba inhibited germination, radicle and plumule elongation when compared with the respective controls. Besides this, the degree of inhibition was increased in concert with increasing concentrations of extracts used. On the other hand, phytohormone levels changed with the application of different extract concentrations. Comparing with the control, the GA levels significantly decreased while the ABA levels increased in all the application groups. Zeatin and IAA levels showed changes depending upon the applied extracts and concentrations.
