The purpose of this study was to investigate the effect of electroacupuncture on weight loss and whether acupuncture treatment can change circulating levels of irisin, nesfatin-l and preptin, which have been discovered in recent years and plays an important role in nutrition and energy metabolism. Irisin, a newly discovered myokine, is associated with increased energy consumption and has a potential role in obesity. Preptin, present in islet b-cells of pancreas and associated with the carbohydrate metabolism, nesfatin- 1 is an anorexogenic peptide composed of 82 amino acids and regulates food intake by modulating neuropeptides in feeding centers in the brain. These three new hormones regulate energy intake and expenditure and preserve energy homeostasis and composition. In this study 4 weeks old Sprague Dawley male rats (n=16) were randomly divided into 2 groups which were the control (C): normal diet and obese (O): high fat diet (HFD) groups. After 16 weeks which obesity was developed, electroacupuncture (EA) was performed to rats once in every second days for 3 times weekly for four consecutive weeks. EA was applied bilaterally to Zusanli (ST36), Bai Hui (Du20), He Gu (Li4), Tianshu (ST25) and Sanyinjiao (SP6) acupoints in animals to both groups. Blood was collected before and after EA and serum irisin, nesfatin-l and preptin levels were analyzed by ELISA (Enzyme-Induced- Immunosorbent Analysis) method. After four week EA treatment, there was sginificant decrease of the average body weight in both groups rats. There were increases in the serum nesfatin-l and preptin levels in the both groups after acupuncture. A decrease was observed in the irisin levels in obese group, but an increase in the control group. These findings suggest that electroacupuncture may help to weight loss due to its effects on some important metabolic hormones such as irisin, nesfatin-l and preptin only for a few weeks. It is thought that the changes of these hormones levels may playa role in beneficial effect of acupuncture used in the treatment of human obesity. The application of EA in the treatment of obesity is very effective, which may have led to weight loss by altering these hormone levels in obese people. A.; Ayar, A.; Ozcelik, O.The analysis of the relationships between minute ventilation (V-E) to CO2 output (VCO2), referred to as ventilatory efficiency, in response to incremental exercise testing, is considered a useful index for assessing the presence and severity of cardiopulmonary and metabolic diseases. The effects of constant load exercise testing performed at work intensity associated with anaerobic threshold (AT) and respiratory compensation points (RCP), on the accurate measurements of ventilatory efficiency are not well known. The aim of this present study was to investigate the reliability of the V-E/VCO2 ratio obtained from constant load exercise tests performed with two important metabolic rates (at the AT and RCP) and compare it to that of those of incremental exercise tests. A total of 20 young male (20.8 +/- 0.4 yr) subjects initially performed an incremental exercise test and then two constant load exercise tests, on different days. Respiratory and pulmonary gas exchange variables were used to estimate AT and RCP. A paired t-test was used to analyse data. AT and RCP (average) occurred the at 60% and at 71% of peak O-2 uptake, respectively. The lowest V-E/VCO2 ratio recorded within the first 2 minutes of constant load exercise tests with a work load of AT (26.4 +/- 0.3) and RCP (26.7 +/- 0.5) was not statistically different from the lowest ratio obtained from the incremental exercise tests (26.0 +/- 0.7). In the constant load exercise test, despite the different metabolic rates, the increase in ventilation corresponded closely with the increase in CO2 production, reflecting an optimal ventilation and perfusion ratio. Clinicians should consider the constant load exercise test work load associated with AT and RCP as it provides a meaningful lowest value for ventilatory efficiency.Article Comparative Evaluation of Mda Levels During Aerobic Exercise in Young Trained and Sedentary Male Subjects(Yuzuncu Yil Universitesi Tip Fakultesi, 2018) Algul, S.; Ugras, S.; Kara, M.The impacts of acute aerobic exercise on oxidative stress as determined altered MDA levels have been examined in young trained and sedentary male subjects. Total of 20 (10 trained and 10 sedentary) male subjects performed an aerobic cycling exercise at the anaerobic threshold for about 30 min. The venous blood samples (pre and post exercise) and analysed for MDA using HPLC methods. Wilcoxon and Mann Whitney U-tests were included in the statistical methods for data analysis. Acute exercise caused a systematic increase in MDA levels in trained (36.2%) and sedentary (55.4%) subjects during exercise (p<0.001). The increase in MDA levels were higher in sedentary subjects (0.79±0.08 µmol/L vs 1.02±0.05 µmol/L) compared to trained subjects (0.73±0.05 µmol/L vs 0.97±0.07 µmol/L) (p<0.001). Acute exercise may cause higher levels of increase in oxidative stress as determined alteration of MDA levels in sedentary subjects. All rights reserved.Article Comparatively Evaluation of the Methods Used in the Assessment of Health and Impairment of Body Composition(Yuzuncu Yil Universitesi Tip Fakultesi, 2025) Ayyildiz, S.A.; Yagiz, S.; Algul, S.; Ozcelik, O.Introduction: Assessment of body composition is an important way that reveal valuable information about the health status of individuals. To evaluate the effectiveness of the methods, the body compositions of young male and female students were compa ratively evaluated using the bioelectrical impedance method (BIA), body mass index (BMI), and waist-hip ratio (WHR). Materials and Methods: A total of 226 (n= 112 males and n= 114 females) young students participated in this study. Body composition analyses were performed using BIA, BMI, and WHR. Results: Considering the whole data, a statistically significant correlation between BMI and BIA was observed. However, there is a difference in risk groups with low and normal values that may result in the differences between the cut-off points determined by ethnicity and gender. Conclusion: In general, although anthropometric measurements have provided meaningful results for population evaluation, technological evaluations should be taken into account in measurements in decision-making situations related to the health status of individuals. © 2025, Yuzuncu Yil Universitesi Tip Fakultesi. Cardiac and skeletal muscle activity results in elevated NADPH oxidase levels in body. In the present study, we intended to evaluate NADPH oxidase levels in response to the aerobic exercise performed in the morning and at night in trained male subjects. Total of 10 healthy trained subjects (age: 19.3±0.7 years and height: 171.8±2.2cm) performed 2 aerobic running exercise in the morning and at night on different days. Venous blood samples were taken at onset and after exercise. NADPH oxidase levels were measured using ELISA method. NADPH oxidase levels were found as 355.17±48.65 nmol/L at basal and increased to 374.42±47.41 nmol/L (p<0.0001) in morning exercise. In addition, it was 353.42±56.07 nmol/L at basal and increased to 3 68.68±55.12 nmol/L after exercise at night (p<0.0001). During exercise, observation of increased percent of NADPH oxidase levels was 6.20±1.8 % in morning and 6.28±1.3 % in night exercise, and no statistical significance was found (p=0.2). As a result, acute aerobic exercise may cause an increase in NADPH oxidase levels in trained subjects but it was not dependent with the time of exercise. © 2018, Yuzuncu Yil Universitesi Tip Fakultesi. G.OBJECTIVE: We evaluated the efficacy of curcumin administration on blood glucose levels and its relationship with nesfatin-1 levels in blood brain and adipose tissue of streptozotocin-induced diabetic rats. MATERIALS AND METHODS: A total of 28 male rats were divided into four groups: control group, type 2 diabetes mellitus (DM) group, control plus curcumin group and type 2DM plus curcumin group. After fifteen days, blood samples were collected from sacrificed rats. Nesftain-1 levels were analysed from blood, brain, and fat tissues of rats in all groups. RESULTS: Nesfatin-1 level was found to be significantly lower in blood, brain and fat tissues of type 2 DM rats compared to the control group. A significant decrease in fasting blood glucose levels was observed in the curcumin administration group compared to type 2 DM group. Improvement of fasting blood glucose level was accompanied by improvement of nesfatin-1 levels in blood, brain, and fat tissues. CONCLUSIONS: As expected, curcumin administration caused significant improvement in fasting blood glucose levels. However, for the first time, we found marked improvements in nesfatin-1 levels in blood, brain, and fat tissues of type 2 DM rats. Thus, considering the crucial role of nesfatin-1 in regulation of glucose metabolism, it is logical to expect an interactive relationship between curcumin and nesfatin-1.Article The Effects of Curcumin Nanoparticles on an Experimental Diabetes Model: Reproductive Hormones, Lipid Profile and Immunohistochemical and Immunofluorescence Markers(B W K Publishing Solutions & verlag, 2024) Belhan, S.; Huyut, Z.; Yildirim, S.; Algul, S.Diabetes affects many organs and causes serious problems for reproductive health. There is still no effective treatment but a range of new strategies are being used to treat and to reduce the possible complications of diabetes. One of the most promising of them relies on the use of nanoparticles. We have evaluated the effects of curcumin and curcumin nanoparticles on blood glucose, lipid profile, reproductive hormones and histopathological, immunohistochemical and immunofluorescent parameters in experimental diabetic conditions. We created eight study groups (control group, diabetes group, curcumin group, Mg-curcumin nanoparticle group, diabetes+curcumin group, diabetes+Mg-curcumin nano- particle group, encapsulated Mg-curcumin nanoparticle group, diabetes+encapsulated Mg-curcumin nano- particle group), allocating rats to the groups at random. Triglyceride, cholesterol and LDL levels were high in the diabetes group, NOP10 and PI3K levels were intense and reproductive hormone levels were low. Curcumin and curcumin nanoformulations administered to the diabetes groups led to significant improvements in these parameters. The testosterone level, which was low in the diabetes group, increased significantly after the application of encapsulated Mg-curcumin. Curcumin nanoformulations were more effective than curcumin at countering the diabetes-impaired lipid profile and improving the levels of reproductive hormones, the histopathological findings and PI3K and NOP10 expression.Article Effects of Exercise on Ghrelin(Mattioli 1885, 2021) Algul, S.; Elcin, S.; Ozcelik, O.Ghrelin, a gastric derived acylated peptide, was discovered in 1999 as a mediator of growth hormone secretiation. Since then, ghrelin has been found to significant impacts on many physiological functions of body systems. Ghrelin has a potential on appetite stimulation that influence on regulation of energy metabolism. It is known that exercise has a marked impact on energy homeostasis and consequently many research focused on the interaction between exercise stimulus and ghrelin response. This review provides an overwiew of research relating to the acute and chronic effects of exercise on (total, acyl and desacyl) ghrelin levels and also impact of exercise intensity on ghrelin levels. Although there is uncertainty among the results of previous studies, our review suggests that acute exercise transiently enterferes with the acly and total ghrelin levels. However, chronic exercise may acause increase in ghrelin levels in contrast to acute exercise with related to change of body weight. In adition, inenstiy of exercise may also reveales various results concerning the relationship between exercise and circulation ghreline levels. The discovery of ghrelin has raised our knowledge of food intake regulation, energy homeostasis and metabolic function mechanism. Further studies should be focused betwen the impact of exercise (type, duration and intensity) on circulation ghreline levels. Exercise induced ghrelin levels will likely generate new pharmacological approaches to many metabolic diseases based on inbalance in body energy homeostasis.Article Evaluation of Relationship Between Aerobic Fitness Level and Range of Isocapnic Buffering Periods During Incremental Exercise Test(C M B Assoc, 2017) Algul, S.; Ozcelik, O.; Yilmaz, B.The purpose of this study is to examine the relationship between the amount of O-2 uptake (VO2) in the range of isocapnic buffering (ICB) periods and aerobic fitness levels of subjects with different exercise tolerance levels. A total of 50 young male subjects (20.8 +/- 0.4 years) performed an incremental exercise test using a cycle ergometer to determinetheir anaerobic threshold (AT), respiratory compensation point (RCP) and maximal exercise capacity (Wmax). The ICB period is defined as the region between AT and RCP. Pulmonary gas exchange parameters were measured breath-by-breath using a respiratory gas analyser. The subjects' fitness levels, as indicated by peak O-2 uptake to body weight ratios (VO(2)peak/BW), ranged from 28 ml/min/kg to 58 ml/min/kg, and Wmax capacity to body weight ratio (Wmax/BW) ranged from 1.94 W/min/kg to 3.96 W/min/kg. The VO2 in the range of ICB periodsranged from 101 ml to 793 ml (with an average of 295 +/- 157 ml). There was a positive linear correlation between VO(2)peak/BW, Wmax/BW and range of ICB: R= 0.76542 (p< 0.0001), and R= 0.92135 (p< 0.0001), respectively. The results of this study suggest that the range of ICB periods may be related to aerobic fitness. Also, it is aimed to investigate the relationships between other laboratory findings used in the diagnosis of brucellosis patients and the irisin hormone. 160 persons (80 brucellosis, 80 controls) were included in this study. Blood was drawn from the people participating in the study following their overnight fast. Irisin level was measured by ELISA method, CRP and sedimentation levels were measured using autoanalysers and commercial kits. It was determined that the irisin level of Brucella patients was statistically significantly higher than the control group (p <0.05). It was determined that there is a negative correlation between irisin level and CRP and sedimentation level in Brucella patients. In Brucella patients, the higher irisin level than the control group showed a negative correlation with CRP and sedimentation levels, which are infection diagnosis criteria; It suggests that the level of irisin can be considered as a supportive diagnostic marker in these patients. All rights reserved.Article Investigation of the Irisin, Preptin and Adropin Levels in the Blood Serum of Alburnus Tarichi(C M B Assoc, 2017) Caf, F.; Algul, S.; Koyun, M.Irisin, preptin and adropin are three newly discovered peptides that play critical roles in regulating energy homeostasis in various vertebrates. The purposes of this study were to measure the serum concentrations of preptin, adropin and irisin in the Alburnus tarichi and to investigate the relationship of these peptides to the weight, gender and length of this the fish, which will provide useful information for future biotechnology purposes aimed at improvements in aquaculture production. This study used 12 adult A. tarichi (6 female and 6 male) obtained from Van Lake (Van, Turkey). The serum irisin, preptin and adropin levels were measured using a commercially available enzyme-linked immunosorbent assay (ELISA) kit to determine correlations between the levels of these three hormones and fish body weight and length. No statistically significant correlations were detected between the serum irisin, adropin and preptin levels and A. tarichi body weight (p = 0.921, r = -0.031; p = 0.08, r = 0.519; p = 0.461, r = -0.235, respectively) or length (p = 0.901, r = -0.040; p = 0.105, r = 0.490; p = 0.236, r = -0.369, respectively). Thus, serum levels of these hormones are independent of fish gender, body weight and length.Article Nitric Oxide Levels in Response To the Patients With Different Stage of Diabetes(C M B Assoc, 2017) Ozcelik, O.; Algul, S.The association between hyperglycaemia and serum nitric oxide (NO) levels has not yet been fully clarified. Thus, it was aimed to evaluate the concentration of NO metabolites (nitrate, nitrite) in diabetic patients and to find the relationship between hyperglycaemia and serum NO levels in diabetic and prediabetic patients, and control groups. In this study, 100 subjects were divided into 3 groups: healthy control (n=20), prediabetic (n=40) and diabetic (n=40). Glycemic control was assessed using glycated hemoglobin (HbA1c). Nitrate and nitrite levels were measured using the Griess reagent. NO was obtained from the sum of nitrates and nitrites. The serum NO level was higher in diabetic (53.4 +/- 2.0 mu M) and prediabetic patients (51.6 +/- 1.6 mu M) as compared to that in the control (45.6 +/- 1.2 mu M), (p< 0.05). The NO level was not significantly different in diabetic and prediabetic patients. Higher levels of serum glucose, insulin and HOMA-IR may be responsible for the activation of endothelial cells to increase NO levels. This study supports the role of insulin resistance in increased NO levels in both diabetic and prediabetic patients.Article Nucleosome Assembly Protein 1-Like 1 (Nap1l1) in Colon Cancer Patients: a Potential Biomarker With Diagnostic and Prognostic Utility(verduci Publisher, 2020) Aydin, M. A.; Gul, G.; Kiziltan, R.; Algul, S.; Kemik, O.OBJECTIVE: To evaluate the diagnostic and prognostic utility of nucleosome assembly protein 1-like 1 (NAP1L1) in colon cancer patients. PATIENTS AND METHODS: A total of 95 patients with colon cancer [mean (SD) age: 61.0 (1.7) years, 58.9% were males] and 50 healthy individuals [mean (SD) age: 61.0 (2.3) years. 52.0% were males] were included in this prospective multicenter study. Data on patient demographics (age, gender) and serum NAP1L1 levels were recorded in both control and colon cancer groups. In colon cancer patients. serum NAP1L1 levels were further analyzed with respect to TNM stages and tumor size. RESULTS: Serum NAP1L1 levels were significantly higher in colon cancer patients as compared with control subjects [median (min-max) 14(12-16) vs. 2(1-2) ng/mL, p<0.001]. In colon cancer patients, serum NAP1L1 levels were significantly higher for tumor size of >4 cm vs. <4 cm [15(12-16) vs. 12(12-14) ng/mL), p<0.001] and for M1 vs. MO stage [15(12-16) vs. 12(12-14) ng/mL), p<0.001]. Serum NAP1L1 levels were significantly higher in T4 stage tumors vs. T1, T2 and T3 stage tumors (p<0.001 for each), in T3 stage tumors vs. T1 and T2 stage tumors (p<0.001 for each) and in N2 stage tumors vs. N0 and N1 stage tumors (p<0.001 for each). CONCLUSIONS: Our findings revealed for the first time the substantial rise in serum NAP1L1 levels among colon cancer patients as compared to controls and as correlated with the disease progression. Accordingly. NAP1L1 seems to be a potential biomarker for colon cancer, offering clinically important information on early diagnosis and risk stratification.Article Uncovering Nesfatin-1 and Irisin Hormones in Schizophrenia and Psychosis Patients: a Comparative Investigation(Elsevier B.V., 2024) Dinçer, E.; Algul, S.; Ozcelik, O.Objective: This study aims to elucidate the potential roles of the hormones nesfatin-1 and irisin in the pathophysiology of schizophrenia and psychosis. Methods: The study involved 80 participants, divided into three groups: schizophrenia (n=30), psychosis (n=20), and controls (n=30). Blood samples were collected from these participants, and the levels of irisin and nesfatin-1 were measured using the ELISA method. Results: The findings revealed that nesfatin-1 levels in schizophrenia patients were significantly higher compared to those in psychosis patients and controls. Additionally, nesfatin-1 levels in psychosis patients were significantly higher than in controls. In contrast, irisin levels were significantly lower in schizophrenia patients compared to both psychosis patients and controls. Discussion: These results suggest that nesfatin-1 and irisin may have important implications for understanding and treating schizophrenia and psychosis. Further research is needed to explore the roles of these hormones in the etiology, diagnosis, and treatment of these conditions. © 2024
