    Effect of Selenium and N-(p Anthranilic Acid on Doxorubicin-Induced Kidney Injury in Rats

    Doxorubicin (DOXR) is one of the essential antitumor drugs. However, its serious adverse effects in many organs limit the clinical use of DOXR. This study aimed to investigate the effect of selenium (Se) and N-(p-Amilcinnamoyl) anthranilic acid (ACA) on kidney tissue in DOXR-administered rats. The rats in the study were divided into six groups (n=10); Control, DMSO, DOXR, DOXR+Se, DOXR+ACA and DOXR+Se+ACA. At the end of the study, intracardiac blood was drawn from rats, and kidney tissues were removed. Urea and creatine levels were measured in serum samples of rats. In addition, histopathological examination of kidney tissue was determined by H&E staining, and 8-OHdG expression was determined by immunohistochemical analysis. Urea and creatine levels increased with DOXR decreased in serum samples after Se and ACA treatments (p< 0.05). While glomerular atrophy, tubular and glomerular dilatation, vascular occlusion and degeneration of tubular epithelial cells were observed in the DOXR group, significant improvement was observed in the Se and ACA treatment groups. In addition, Se and ACA treatments reduced DOXR-induced 8-OHdG expression (p< 0.05). These findings indicated that Se and ACA could be used as critical therapeutic agents to suppress renal dysfunction and oxidative DNA damage that can occur after DOXR-induced kidney injury. © 2023, Inonu University.     Effects of Diclofenac Sodium Exposure During Pregnancy on Cox-2 Expression in the Ductus Epididymis and Ductus Deferens of the Offspring Rat

    The present study aimed to investigate the effects of diclofenac sodium application during pregnancy on COX-2 expression in the ductus epididymis (DE) and ductus deferens (DD) of the offspring rat. 8 Eight adult Wistar Albino non-pregnant female rats and two breeding male rats were utilized. Rats determined to be pregnant were separated into two categories as sham (S) and diclofenac sodium (DS). It was injected into the S group with 1 ml/kg/day saline for 10 days, and it was injected into the DS group with 6.1 mg/kg/day DS. 7 male offspring were used in each group. DS decreased the serum testosterone levels and incresed the number of mast. DS induced fibrotic thickening. Offspring of S group had moderate COX-2 in the caput epididymis (CPE) and severe COX-2 in the cauda epididymis (CE), while those in the DS group had weak COX-2 in the CPE and moderate COX-2 in the CE. There was no COX-2 expression in the vas deferens and corpus epididymis (COE) of the offspring rats in the S and DS groups. TNF-α expression was not observed in the S and DS groups. It was observed that moderate IL-10 expression in the CPE and CE, and weak IL-10 expression in the vas deferens and COE in the S and DS groups. The findings of our study reveal that DS administration during pregnancy can cause the downregulation of COX-2 expression in the DE and DD of offspring rats and therefore may have adverse effects on the male reproductive system. © 2024 Ondokuz Mayis Universitesi.     Effects of Epigallocatechin Gallate Administration on Diabetic Rat Myocardial Tissue

    This study aimed to investigate the actions of Epigallocatechin Gallate (EGCG) on heart tissue in diabetic rats. In the study, twenty-eight Wistar albino male rats were taken and divided into 4 groups as control group, EGCG, Diabetes, and Diabetes + EGCG. A one dose of 45 mg/kg streptozotocin (STZ) was administered intraperitoneally to the groups to be described with diabetes. EGCG was given to the animals at 20 mg/kg/day for 14 days using an intragastric tube. The heart tissue was stained with Masson trichrome and Hematoxylin-Eosin (H&E). Heart tissue was evaluated by histopathological and immunohistochemical (Collagen type III) methods. In the histological structure of the diabetic group, irregular heart muscle fibers, inflammatory cell infiltration, and necrotic myocytes in the myocardium an d increased collagen type III expression were observed, and these findings were found to decrease in the Diabetes + EGCG group. It was found that the density of collagen content increased in the Diabetes group. This finding decreased in the Diabetes + EGCG g roup. The findings of our study suggest that EGCG may have ameliorative effects on induced cardiotoxicity in diabetic rats. © 2025, Yuzuncu Yil Universitesi Tip Fakultesi. All rights reserved.
