Article Changes in Sperm Parameters Following Administration of Theophylline, a Competitive Antagonist of Adenosine in Rats Exposed To Bleomycin, a Chemotherapeutic Agent(Ataturk Universitesi, 2019) Belhan, S.; Oto, G.; Arihan, O.; Koşal, V.The aim of the present study is to determine the effects of theophylline (THP), a methylxanthine derivative, on the sperm parameters of rats exposed to bleomycin (BLM) as a chemotherapeutic agent. The study was performed on 32 male rats. The rats in Group 1 were administered only with sterile saline intratracheally. The rats in Group 2 were administered a single dose of 7.5 mg/kg of BLM intratracheally. The rats in Group 3 received a single dose of 7.5 mg/kg of BLM intratracheally. In addition, THP was administered to rats in Group 3 intraperitoneally at a dose of 75 mg/kg/ day for 14 days. The rats in group 4 were administered an intraperitoneal injection of THP at a dose of 75 mg/kg/day for 14 days. Motility, sperm density, abnormal sperm and dead sperm rates were examined. Significant deteriorations were observed in sperm parameters of rats that were administered BLM. In sperm parameters of rats that were administered THP, it was determined that it only increased motility, but did not significantly change sperm density, abnormal sperm and dead sperm count. In this study, THP only increased motility. There was no positive effect on sperm parameters except motility. © 2019 Ataturk Universitesi. All rights reserved.Article Effect of Heliotropium Hirsutissimum, Heliotropium Dolosum and Heliotropium Lasiocarpum Methanolic Extracts in Pentylentetrazol Induced Convulsions on Mice(University of Ankara, 2018) Arihan, O.; Karakaya, S.; Kilic, C.S.; Ergul Erkec, O.; Kara, M.; Duman, H.Objective: Epilepsy is a common neurological problem known since ancient times. Modern treatment of epilepsy includes treatment with drugs to avoid seizures. Search for new antiepileptic drugs continues due to the presence of resistant patients to drug treatment. Plants are important sources of new drug discovery for epilepsy. We aimed to evaluate anticonvulsive activity of methanolic extracts of Heliotropium (Scorpion herb, Herb for warts in Turkish) species namely Heliotropium hirsutissimum Grauer, H. dolosum De Not and H. lasiocarpum Fisch. et Mey. on mice. Material and Method: PTZ (pentylentetrazol) is used for the induction of convulsions. Experimental groups are PTZ and 3 groups of extracts of aforementioned Heliotropium species+PTZ. Result and Discussion: In H. hirsutissimum and H. lasiocarpum groups, number of animals having tonic-clonic convulsions was decreased compared to PTZ and H. dolosum groups however difference was found insignificant. Latency time for first myoclonic convulsion and latency time for first tonic-clonic convulsion were prolonged in all Heliotropium groups compared to PTZ group (p>0.05). Number of ex animals following tonic-clonic convolsions decreased in Heliotropium groups (p>0.05). Tonic-clonic convulsion period decreased in H. hirsutissimum and H. lasiocarpum (p>0.05). When results are evaluated together Heliotropium species may have an ethnopharmacological relevance for its anticonvulsive usage. © 2018 University of Ankara. All rights reserved.Article Effect of Intermittent Fasting During Acrylamide Exposure on Blood Copper, Zinc and Lead Levels in Rats(Yuzuncu Yil Universitesi Tip Fakultesi, 2018) Huyut, Z.; Arihan, O.Zinc and copper are responsible for certain physiological functions in the body. On the other hand lead is a toxic trace element. In this study, effect of intermittent fasting on blood zinc, copper and lead levels during acrylamide exposure was studied. Forty female adult Wistar albino rats were divided into 4 groups (n=10 in each group). First group: control group without any administration, second group: acrylamide group in which 20 mg/kg/day acrylamide was administered for 30 days, third group: intermittent fasting group in which 12 hours of fasting were administered overnight for 30 days, fourth group: intermittent fasting and acrylamide group in which 20 mg/kg/day acrylamide and 12 hours of intermittent fasting was administered for 30 days. At the last day of experimental procedure blood was withdrawn under anesthesia. Zinc as well as copper levels were measured from plasma samples in flame unit and lead was measured from erythrocyte packages in combustion unit of Atomic Absorption Spectrophotometer. In acrylamide group, zinc was lower (p<0.05) whereas copper (p>0.05) and lead levels (p<0.05) were increased. In intermittent fasting group zinc level was higher whereas copper and lead levels were lower compared to acrylamide group (p<0.05). In acrylamide+intermittent fasting group lead level was significantly lower (p<0.05). Conclusion: Results indicate an augmentation of toxic copper and lead elements due to acrylamide administration whereas a decrease in zinc which is an important element for growth and immune system. On the contrary, intermittent fasting decreased such untoward effect of acrylamide and increased potential detoxification of toxic chemicals. © 2018, Yuzuncu Yil Universitesi Tip Fakultesi. All rights reserved.Article Effect of Safran, Safranal and Crocin Which Are Active Ingredients of Saffron (Crocus) on Erythrocyte Fragility and Hematological Parameters in Carbon Tetrachloride Intoxicated Rats(Yuzuncu Yil Universitesi Tip Fakultesi, 2016) Arihan, O.; Oto, G.; Bayram, I.; Aras, I.Carbon tetrachloride is a toxic chemical and well known for its carcinogenic property. Intoxication with carbon tetrachloride causes serious liver injury and it is used as an experimental model for triggering liver related diseases. Saff ron (Crocus) species are well known bulbous ornamental and aromatic plants. Many of the saffron species are used in ethnomedicinal practices. In addition both saffron species and their ingredients are also subject to scientific research. In this study 72 rats were divided into 9 groups (n=8 in each group). Groups were set as control, olive oil, carbon tetrachloride, safran, safranal, crocin, carbon tetrachloride + safranal, carbon tetrachloride + crocin. Administrations were conducted for 7 days. Erythrocyte osmotic fragility and some selected hematological parameters were assessed. Results state significant increase in erythrocyte fragility due to carbon tetrachloride and amelioration due to active ingredients of saffron. Among hematocrit, hemoglobin and red blood cell count parameters only significant alteration was observed in safran with an increase in red blood cell count. Different administration schemes can be evaluated in future studies in order to assess detailed impact of saffron constituents on hematological parame ters and erythrocyte integrity. © 2017 Yuzuncu Yil Universitesi Tip Fakultesi. All rights reserved.Article Effects of Acute Aerobic Exercise on Nadph Oxidase Levels in Trained Male Subjects(Yuzuncu Yil Universitesi Tip Fakultesi, 2018) Algul, S.; Arihan, O.; Ozcelik, O.Exercise induced muscle activity causes increase in oxidative stress and free radical levels. Cardiac and skeletal muscle activity results in elevated NADPH oxidase levels in body. In the present study, we intended to evaluate NADPH oxidase levels in response to the aerobic exercise performed in the morning and at night in trained male subjects. Total of 10 healthy trained subjects (age: 19.3±0.7 years and height: 171.8±2.2cm) performed 2 aerobic running exercise in the morning and at night on different days. Venous blood samples were taken at onset and after exercise. NADPH oxidase levels were measured using ELISA method. NADPH oxidase levels were found as 355.17±48.65 nmol/L at basal and increased to 374.42±47.41 nmol/L (p<0.0001) in morning exercise. In addition, it was 353.42±56.07 nmol/L at basal and increased to 3 68.68±55.12 nmol/L after exercise at night (p<0.0001). During exercise, observation of increased percent of NADPH oxidase levels was 6.20±1.8 % in morning and 6.28±1.3 % in night exercise, and no statistical significance was found (p=0.2). As a result, acute aerobic exercise may cause an increase in NADPH oxidase levels in trained subjects but it was not dependent with the time of exercise. © 2018, Yuzuncu Yil Universitesi Tip Fakultesi. All rights reserved.Article How Long-Term Intake of Sodium Fluoride (Naf) in Different Doses and 7,12 Dimethylbenz(A)anhtracene (Dmba) Affect the Erythrocyte Parameters in Rats(Pakistan Agricultural Scientists Forum, 2019) Comba, B.; Oto, G.; Arihan, O.; Comba, A.; Uyar, H.This study was aimed to search the effect of Sodium Fluoride (NaF) and 7,12 dimethylbenz(a)anthracene (DMBA) to erythrocyte fragility and parameters in rats. The nine groups were formed and each group contained 8 animals. Group 1: Control group (without any treatment). Group 2: Sesame oil (vehicle for DMBA). Group 3: 1 ppm NaF. Group 4: 15 ppm NaF. Group 5: 30 ppm NaF. Group 6: DMBA.Group 7: 1 ppm NaF + DMBA. Group 8: 15 ppm NaF + DMBA. Group 9: 30 ppm NaF + DMBA. Fluoride was added into the animals' drinking water in the form of NaF once a day, for 12 weeks. DMBA (10 mg/kg) was administered once a week and in a total of 12 weeks with oral gavage. Erythrocyte fragility was analyzed with osmotic hemolysis method and erythrocyte parameters with blood cell counter in whole blood. At 0.4% NaCl concentration groups 4, 5, 6, 8, and 9 showed significantly higher erythrocyte fragility values than control group (p <= 0.05). At 0.5% NaCl concentration, groups 6, 8 and 9 showed significant increase in erythrocyte fragility compared to other groups (p <= 0.05). The erythrocyte and hematocrit values were found significantly high in group 5 (p<0.001) and group 4 (p<0.01) while it was found low in all groups with DMBA (6, 7, 8, 9) (p <= 0.05) compared to control group. Hemoglobin amount in group 5(p <= 0.01) and group 4 (p <= 0.05) were significantly higher than other groups. MCV and MCH in group 5 were significantly lower and these values in all groups with DMBA (6, 7, 8, 9) were determined significantly high compared to other groups. RDWC in group 5 (p <= 0.001) and group 4 (p <= 0.01) and in all groups with DMBA (6, 7, 8, 9) (p <= 0.05) was significantly increased compared to other groups. As a result, exposure to high doses of floride and DMBA may cause augmented erythrocyte fragility, abnormal erythrocyte parameters and anemia. Therefore, measures must be taken to protect the health of all living organisms in area exposed to high levels of fluoride and DMBA.Article Protective Effect of Astaxanthin in the Lung Injury Caused by Ischemia Reperfusion of the Lower Extremities(Pakistan Agricultural Scientists Forum, 2019) Akkoyun, H. F.Pathological and biochemical alterations due to lower extremity (I/R) damage and protective effects of astaxanthine (AST) were investigated. Rats were divided into four groups. GI-Sham group (n=7):Anesthesia without (I/R)(2hours);GII-I/R (n=7) : 2 hours of ischemia and 2 hours of reperfusion under anesthesia; Group III-AST(n=7): Rats were subchronically orally administered for 7 days at 125 mg/kg astaxanthin (AST) and then anesthetized (2hours) without ischemia; GIV-I/R+AST (n=7) : 7 days prior to ischemia rats were subchronically orally administered 125 mg/kg astaxanthin (AST) and then 2 hours of ischemia and reperfusion under anesthesia; Then lung tissues were investigated for MDA,GSH and histopathology. An increase in MDA and a decrease in GSH was observed I/R administered group compared to control. Histopathological evaluations showed intense congestion in pulmonary veins and alveolar septum and partial alveolar macrophage and erythrocyte accumulation and edema was observed in lumens of some bronchioles and alveoli in the second and fourth group compared control. Second group (3.41) damage score had high significance compared to control (p <= 0.001). Fourth group damage score (0.92) was indifferent from control but significantly different from I/R group (p <= 0.001). As a result; The protective effect of AST has been demonstrated by biochemical, histopathological and immunohistochemical effects.
