Article 2,6-Bis((e) and Its Metal(Ii) Complexes: Synthesis, Spectroscopy, Biological Activity, and Photoluminescence Features(Wiley-v C H verlag Gmbh, 2013) Gulcan, Mehmet; Zengin, Huseyin; Celebi, Metin; Sonmez, Mehmet; Berber, IsmetThe preparation, characterization, magnetic measurements, antimicrobial activity, and photoluminescence properties of a number of metal complexes with a Schiff base ligand derived from 2, 6-diformyl-4-methylphenol and 1-amino-5-benzoyl-4-phenyl-1H-pyrimidine-2-thione are reported herein. UV/Vis spectra and magnetic measurements infer square planar stereochemistry for Pd-II and Pt-II complexes, and octahedral arrangement for the other metal complexes. The compounds results in intense emission (where (max) = 402 nm) upon irradiation by UV light. The photoluminescence quantum yields and long excited-state lifetimes of the ligand and its complexes were calculated as 42% and 3.94 ns, respectively. The photoluminescence intensities and quantum yields of metal complexes changed upon complexation with various metal ions. The ligand and its complexes are of interest as organic emitting materials for electroluminescent devices. The complexes were additionally evaluated for in vitro the antimicrobial activity against bacterial and fungal strains. While Cu-II, Co-II, and Pt-II complexes have good antifungal efficiency against yeast (MICs, 80 gmL(-1)), only the complex [Cu(L)(AcO)]4H(2)O had effective and selective antimicrobial activity against all tested microorganisms with MIC values in the range of 40-80 gmL(-1), except for E. coli ATCC 4230. Cultivars Grown in Turkey by Sds-Page of Seed Proteins(Pakistan Botanical Soc, 2011) Berber, Ismet; Yasar, FikretIn the present research, a total 28 bean cultivars grown in Turkey were characterized by the numerical analysis of seed protein profiles obtained by using sodium dodecyl sulphate polyacrylamide gel electrophoresis (SDS-PAGE) technique. The numerical analysis of seed protein profiles generated two major clusters at similarity levels of 82.52% or above. The first cluster included 5 reference bean cultivars at the intra-cluster similarity values of 91.25% or above. The second cluster comprised the all remaining genotypes at similarity values between 84.46% and 100%. Our results also indicated that Gina standard cultivar together with all eastern genotypes belonged to subcluster IIc, having the highest intra-cluster similarities between 95.54% and 100%. In this sense, it is suggested that probably there may be a very close genetic relationship between eastern genotypes and Gina cultivar. In conclusion, this study indicated that the numerical analysis of seed protein profiles offer no final answer to the identity of bean genotypes, however, the method can provide of extra banding pattern for the discrimination of bean genotypes cultivated in Turkey.Article Discrimination and Numerical Analysis of Human Pathogenic Candida Albicans Strains Based on Sds-Page Protein Profiles(Academic Journals, 2008) Berber, Ismet; Ekin, SuatIn the present study, 21 Candida albicans strains were investigated using the commercial kit API 20C AUX system and the numerical analysis of whole-cell protein profiles. The results of the commercial kit confirmed that the all the strains belonged to C. albicans species. However, the research indicated that SDS-PAGE of polypeptides of whole-cell extracts can provide more valuable taxonomic information than conventional yeast test kits at the subspecies level. Despite the fact that C. albicans subtypes isolated from different anatomical sites had similar protein profiles, there were some distinctive protein bands. Numerical analysis of whole-cell protein profiles of all strains revealed 2 major clusters at similarity degrees of between 46.26 and 100%. Moreover, the results of numerical analysis confirmed that each cluster had characteristic and distinctive protein profiles. The research showed that, the morphological examination of yeast isolates remains essential to obtaining a correct identification, both the commercial yeast kit system and the numerical analysis of whole-cell protein patterns can be useful for the more reliable identification of C. albicans strains.Article The Effect of Plantago Major Linnaeus on Serum Total Sialic Acid, Lipid-Bound Sialic Acid, Some Trace Elements and Minerals After Administration of 7,12-Dimethylbenz(a)anthracene in Rats(Sage Publications inc, 2012) Oto, Gokhan; Ekin, Suat; Ozdemir, Hulya; Levent, Abdulkadir; Berber, IsmetThe present study was designed to evaluate the effect of Plantago major Linnaeus (PM) extract on serum total sialic acid (TSA), lipid-bound sialic acid (LSA), some trace elements (copper (Cu), zinc (Zn) and iron) and mineral levels (magnesium, calcium and sodium) in Wistar albino rat administrated 7,12-dimethylbenz(a)anthracene (DMBA). Rats were divided into three equal groups (n = 6). Group I comprised the control group, group II was treated with DMBA (100 mg/kg, single dose) and group III was treated with DMBA (100 mg/kg single dose) and aqueous extract of PM 100 mg/kg/day for 60 days. After 60 days, statistical analyses showed that TSA and LSA levels in DMBA and DMBA + PM groups were significantly higher compared to the control group (TSA: p < 0.01, p < 0.05; LSA: p < 0.05, p < 0.05, respectively). Serum Zn levels were decreased in subjects treated with DMBA (p < 0.01) and DMBA + PM (p < 0.05) compared to the control group values. Serum Cu levels were increased in DMBA group and PM-treated group compared to the control group values. The results of this investigation showed that the levels of TSA and LSA changed significantly, which are sensitive markers for detecting the toxic effects of DMBA. On the other hand, observed decline in Zn levels in rats from DMBA + PM group might be due to decreased generation of free radicals and oxidative stress. Results from this study suggest that PM may be partially effective in preventing carcinogenesis initiated by environmental carcinogen DMBA.Article Effect of Urtica Dioica L. on Selected Trace Elements Levels and Serum Protein Patterns in the Rabbits Treated With 7,12-Dimethylbenz(a)anthracene(Asian Journal of Chemistry, 2008) Ekin, Suat; Oto, Gokhan; Berber, Ismet; Turel, Idris; Kusman, TubaIn the present study, the effect of Nettle (Urtica dioca L.) on trace elements and serum protein patterns in female rabbits administrated to 7,12-Dimethylbenz(a)anthracene (DMBA) was investigated. Rabbits were divided into equal four groups. Group I was control group (n = 8), group 11 (n = 8), treated with DMBA (1 mg/kg.d), group III treated with DMBA (I mg/kg.d) and methanol extract of U. dioica 35 mg/kg/d and group IV (n = 8) treated with DMBA (1 mg/kg.d) and aqueous extract of U. dioica 35 mg/kg/d. The results show that on 60th day DMBA-treated group were compared with the control group levels, a significant decrease (p < 0.05) was observed in the Zn. DMBA and methanol extract of U. dioica treated group were compared with the control group levels, a significant increase (p < 0.05) was observed in the Cu. Serum Fe and Mn levels among groups were determined not to be statistically significant (p > 0.05). It was found that U. dioica not affected on trace elements level and serum protein patterns in rabbits subjected to DMBA.Article Levels of Selected Trace Elements, Phytohormones, and Sugars in Pseudomonas-Infected Lycopersicum Esculantum Mill Plants(Humana Press inc, 2010) Berber, Ismet; Ekin, Suat; Battal, Peyami; Onlu, Harun; Erez, M. EmreThe present study investigated the levels of trace elements (Zn, Cu, Fe, Pb, and Cd), major elements (Ca and K), phytohormones (trans-Zeatin [t-Z] and gibberellic acid [GA]), and sugars (sucrose and glucose) following inoculation with Pseudomonas syringae pv. tomato strain. The results of the trace elements analysis showed that Fe (in the first, fourth, eighth, and tenth study days), Cd (in the fourth, eighth, and tenth study days), Cu (in the fourth and eighth study days), and Zn (in the eighth and tenth study days) increased in bacterium-infected tomato plants, compared to healthy plants. The levels of Pb, Ca, and K did not meaningfully determine a change after inoculation with pathogen. In this vein, the increase accumulation rates of Cu, Zn, and Fe in the injured plants can be an important indicator for the plant defense processes towards pathogen attack. Furthermore, in the first, fourth, eighth, and tenth study days, the glucose and sucrose contents crucially decreased in bacterium-infected plants compared to the control groups. The lowest level of sucrose in bacterium-infected plants was observed on the first day. The findings displayed that, when endogenous t-Z levels did not change after inoculation with virulent bacterium strain, there was a reduction in the first, fourth, eighth, and tenth days in the level of GA when compared with the control group levels. Therefore, there may be a link between lower GA level and enhancement in the expression of defense-related genes. The results of this study showed that there are complex relationships among levels of sugar, trace element, and endogenous phytohormone in the regulation of defense mechanisms against bacterial pathogen attacks.Article Method Validation and Simultaneous Determination of Retinol, Retinyl Palmitate, Β-Carotene, Α-Tocopherol and Vitamin C in Rat Serum Treated With 7,12 Dimethylbenz[A]anthracene and Plantago Major L. by High-Performance Liquid Chromatography Using Diode-Array Detection(Bentham Science Publ Ltd, 2013) Levent, Abdulkadi; Oto, Gokhan; Ekin, Suat; Berber, IsmetA new and simple high-performance liquid chromatography method was developed and validated for the simultaneous determination of retinol, retinyl palmitate, beta-carotene, alpha-tocopherol and vitamin C in rat serum treated with Plantago Major L. and 7,12 dimethylbenz[a] anthracene. High-performance liquid chromatography analysis was performed utilizing an Inertsil ODS3 reversed phase column with methanol-tetrahydrofuran-water as mobile phase under gradient conditions, at 1.5 mL min(-1) flow rate and 25 degrees C. Diode-array detection was at 325, 450, 290 and 270 nm (retinol and retinyl palmitate), beta-carotene, alpha-tocopherol and vitamin C, respectively and runnig time 18 min. The high-performance liquid chromatography assay and extraction procedure proposed are simple, rapid, sensitive and accurate. The method was then applied for the determination of retinol, retinyl palmitate, beta-carotene, alpha-tocopherol and vitamin C in rat serum. Results of this study demonstrated that; at 60th day DMBA-treated group, there was a significant decrease in vitamin levels compared to the levels of control group. A significant increase was observed in vitamin levels of 7,12 dimethylbenz[a]anthracene+Plantago Major L.-treated group compared to the DMBA-treated group. Additionally, the results obtained in the study are found to be in agreement with data reported in the literature.Article Microwave-Assisted Synthesis of Tetrahydropyrimidines Via Multicomponent Reactions and Evaluation of Biological Activities(Bentham Science Publ Ltd, 2011) Akbas, Esvet; Berber, Ismet; Akyazi, Inci; Anil, Baris; Yildiz, ElaVarious pyrimidine derivatives (1a-c) were synthesized using the Biginelli three component cyclocondensation reactions of a beta-diketone, arylaldehyde, and thiourea, under microwave irradiation and conventional conditions. The acetylation of compound 1a gave 3-acetyl thioxopyrimidin 2. Also, thiazolopyrimidine (3) and (4) derivatives were obtained by a simple one-pot condensation reaction of starting compounds 1a, and 1b with 2-bromopropionic acid. The prepared compounds were screened for their antimicrobial activities against Gram-positive, Gram-negative bacteria and fungi using micro dilution procedure. The results of the study showed that the compound 4 was effective and selective for antimicrobial activity against the tested bacteria and fungi. Thus, we suggested that the compound 4 might be a new potential antimicrobial substance for bacteria and fungi.Article Molecular and Physiological Changes in Maize (Zea Mays) Induced by Exogenous Naa, Aba and Meja During Cold Stress(Finnish Zoological Botanical Publishing Board, 2008) Battal, Peyami; Erez, Mehmet E.; Turker, Musa; Berber, IsmetDifferent concentrations of abscisic acid (ABA), naphthalene acetic acid (NAA) and methyl jasmonate (MeJa) were applied to maize (Zea mays) plants exposed to cold stress. ABA enhanced plant resistance against the damaging effects of freezing. ABA-treated plants produced some specific proteins when exposed to cold. Prominent changes were observed in the size and number of stomata with the application of ABA during cold stress. MeJa inhibited root growth, but increased root dry weight during cold acclimation. The chlorophyll content was lower at 100 mu g ml(-1) NAA and 50 mu g ml(-1) MeJa and higher in the residual applications when compared with that in the control plants. Carotenoid levels increased in all NAA and some ABA treatments, decreased however in MeJa treatment. Exogenous MeJa treatments increased endogenous trans-zeatin (t-Z) and indole-3-acetic acide (IAA) in contrast decreased ABA levels. Bis-chelate complexes of the ligand were prepared from acetate/chloride salts of Cu(II), Co(II), Mn(II), Ni(II), Zn(II), Cd(II), and Pd(II) in methanol. The structures of the ligand and its metal complexes were characterized by microanalyses, IR, AAS, NMR, API-ES, UV-Vis spectroscopy, magnetic susceptibility, and thermogravimetric analyses. An octahedral geometry has been suggested for all the complexes, except for Pd(II) complex, in which the metal center is square planar. Each ligand binds using C(2)=O, HN, and carboxylate. The cyclic voltammograms of the ligand and the complexes are also discussed. The new synthesized compounds were evaluated for antimicrobial activities against Gram-positive, Gram-negative bacteria and fungi using the microdilution procedure. The Cu(II) complex displayed selective and effective antibacterial activity against one Gram-positive spore-forming bacterium (Bacillus cereus ATCC 7064), two Gram-positive bacteria (Staphylococcus aureus ATCC 6538 and S. aureus ATCC 25923) at 40-80 mu g mL-1, but poor activity against Candida species. The Cu(II) complex might be a new antibacterial agent against Gram-positive bacteria.Article Numerical Analysis of Sds-Page Protein Patterns of Facultative Alkaliphilic Bacillus Species Isolated From Lake Van, Turkey(Parlar Scientific Publications (p S P), 2006) Berber, Ismet; Berber, SerifeIn the present study, seven reference Bacillus species and a total of eighteen new facultative alkaliphilic Bacillus strains, isolated from the water of Lake Van and its soil surroundings, were identified using phenotypic characteristics and the numerical analysis of whole-cell protein profiles. According to morphological, biochemical and physiological characteristics, it was found that the new isolated strains belonged to Bacillus genus. In addition, the research indicated that SDS-PAGE of polypeptides of whole-cell extracts can provide more valuable taxonomic information than conventional tests based on the phenotypic characteristics at the species and subspecies levels. Numerical analysis of whole-cell protein profiles of test strains revealed 4 basic clusters (I-IV) at dissimilarity values of 18.9 % or above. The results of numerical analysis confirmed that each cluster had characteristic and distinctive protein profiles. Indeed, this study showed that the application of numerical analysis, coupled with the utilization of a standardized identification system instead of simple quantitative comparison of protein patterns, greatly enhanced the utilization of whole-cell protein profiles for identification of the facultative alkaliphilic Bacillus species.Article Selected Trace Elements and Esterase Activity of Carbonic Anhydrase Levels in Lambs With Pneumonia(Humana Press inc, 2006) Ekin, Suat; Berber, Ismet; Kozat, Suleyman; Gunduz, HandanThe levels of, zinc, copper, Fe, Zn, Cu, Mn, Mg, K, Na, and Cl and the activity of carbonic anhydrase were determined in lambs with pneumonia. Asignificant decrease of p < 0.01 level in Zn concentration, in Cu level (p < 0.001) and significant increases in K and Na levels (p < 0.05) and of the Cu/Zn ratio (p < 0.001) were observed in the study group. The carbonic anhydrase activity was decreased in the study group, but the decrease was not statistically significant (p > 0.05). Also, nonsignificant decreases of Fe, Mg, and Cl and increase of the Mn concentration were also observed in the lambs with pneumonia (p > 0.05). Our results suggest that the significant element changes reported here and the Cu/ Zn ratio, but not the activity of carbonic anhydrase, can be used as indicators of pneumococcal infection.Article Synthesis, Characterization, and Antimicrobial Activity of a New Pyrimidine Schiff Base and Its Cu(Ii), Ni(Ii), Co(Ii), Pt(Ii), and Pd(Ii) Complexes(Tubitak Scientific & Technological Research Council Turkey, 2012) Gulcan, Mehmet; Sonmez, Mehmet; Berber, IsmetA new Schiff base, {1-[(5-bromo-2-hydroxy-benzylidene)-amino]-4-phenyl-2-thioxo-1,2-dihyro-pyrimidin-5-yl}-phenyl-methanone, was synthesized from N-amino pyrimidine-2-thione and 5-bromsalicylaldehyde. Metal complexes of the Schiff base were prepared from acetate salts of Cu(II), Ni(II), Co(II), Pd(II), and PtCl2 in methanol. The chemical structures of the Schiff base ligand and its metal complexes were confirmed by spectroscopic analysis. All of the compounds were evaluated for their antimicrobial against 4 gram-positive bacteria, I grain-negative bacterium, and 3 yeast strains. Bis-chelate complexes of the ligand were prepared from acetate/chloride salts of Cu(II), Co(II), Ni(II), Mn(II), Zn(II), Cd(II), Fe(III), Cr(III), and Ru(III) in methanol. The structures of the ligand and its metal complexes were characterized by microanalyses, IR, NMR, API-ES, UV-Vis spectroscopy, magnetic susceptibility, and conductometric analyses. Octahedral geometry was suggested for all the complexes, in which the metal center coordinates to ONO donors of the ligand. Each ligand binds the metal using C=O, HN, and carboxylate. The cyclic voltammograms of the ligand and the complexes were also discussed. The compounds were evaluated for their antimicrobial activities against Gram-positive and Gram-negative bacteria, and fungi using microdilution procedure. The antimicrobial studies showed that Cu(II), Fe(III), and Ru(III) complexes exhibited good antibacterial activity against Gram-positive bacteria with minimum inhibitory concentrations between 20 and 80 mu g mL-1. However, the ligand and the complexes possess weak efficacy against Gram-negative bacterium and Candida strains. The complexes were prepared by reacting ligand and the metal chlorides of Cu (II), Ni(II), Co(II) and Fe(II) in methanol to get a series of mononuclear complexes. The complexes were characterized by elemental analyses, conductivity measurements, magnetic susceptibility data, IR, UV-Vis, NMR and API-ES mass spectral data. The mononuclear structure of the complexes was confirmed on the basis of elemental analyses, magnetic susceptibility and API-ES mass spectral data. The ligand and all the metal complexes were evaluated for their antimicrobial activity against four Gram-positive bacteria (Staphylococcus aureus ATCC 6538, S. aureus ATCC 25923, Bacillus cereus ATCC 7064 and Micrococcus luteus ATCC 9345), one Gram-negative bacterium (Escherichia coli ATCC 4230), and three yeast (Candida albicans ATCC 14053, Candida krusei ATCC 6258 and Candida parapsilosis ATCC 22019) strains. Therefore, newly synthesized the ligand and two complexes [(Cu(II) and Co(II)] showed good biological activity against all tested bacteria and yeast strains. (C) 2010 Elsevier Masson SAS.
