The purpose of this study was to present findings of congenital colo-vesico-urethral fistula and atresia coli, rectal agenesis and atresia ani clinically, radiographically, macroscopically and histopathologically. According to the clinical examination; anus did not form, animal was dehydrated and not able to get to its feet and had abdominal tension. Besides, few amounts of unclear urine and meconium were present in preputium. By taking into consideration these findings, it was decided to make surgical operation on suspicion of fistula presence between gastrointestinal system and urinary bladder. The animal died during operation. Subsequent to necropsy, colon was swollen and observed at the cranial of pelvis ending bluntly. The blunt end of colon adhered to both urinary bladder and urethra. In order to diagnosis anomaly, samples were obtained from the region where fistulas were present. According to histopathological examination, fistula formation of colon separately to both urethra and urinary bladder was observed. Observing congenital of colo-vesico-urethral fistula, atresia coli, rectal agenesis and atresia ani all in one only in materials rare and may present a good sample for congenital anomaly cases. © 2017, Chartered Inst. of Building Services Engineers. The case was made clinical examination which brought to our clinics with complain of sudden onset of anorexia, a reduction of milk and failure to pass feces. After clinical examination of animal were decided in the operation. Determined the phytobezoar of bowel in the ceacum ileo-ceace-coloc area which accepted to cause of cecal dilatation and dislocation was removed. For a period of 20 days of postoperative follow-up in animal were observed any of the complications and getting better.Article Effect of Carboxymethylcellulose, Icodextrin and Hyaluronic Acid Solutions on Postoperative Intraabdominal Adhesion Formation in Rabbits and the Role of Cytokines in Intraabdominal Adhesion Formation(Polskie Towarzystwo Nauk Weterynaryjnych, 2024) Karasu, A.; Altug, N.; Bakir, B.; Yener, Z.In this study, the aim was to investigate the effects of 1% CMC, 4% ICO and 0.4% HA solutions in preventing postoperative IAs and their contribution to peritoneal wound healing, as well as the relationship of cytokines with the formation of postoperative IAs in rabbits. The material of the study consisted of 32 healthy rabbits. Rabbits underwent median laparotomy following general anesthesia. Serosal abraded was created at the antimesenteric border of the cecum. Next, a 3 × 2 cm peritoneum was excised on the right abdominal wall and the defect was closed using with a 2/0 silk suture. The rabbits were randomly assigned to either of the following four treatment groups: CNT (0.9% NaCl), ICO (4% icodextrin), HA (0.4% hyaluronic acid) and CMC (1% carboxymethyl cellulose). Both cecum and peritoneal surfaces were treated with 20 ml each of treatment solutions. On the 7th postoperative day, the rabbits were euthanized with a lethal dose of sodium pentobarbital and the degree of adhesion was evaluated. Samples taken from the peritoneal defect were examined histopathologically and tissue hydroxyproline levels were measured. Serum TNF-α, IL-1 and IL-6 levels were measured from blood samples taken before the surgery and at 1, 6, 24 and 48 hours after the surgery. It was observed that the adhesion grade in the HA group (p < 0.05) and CMC group (p < 0.01) was significantly lower than the control group. Although peritoneal tissue hydroxyproline levels were lower in the other groups compared to the control group, they were not statistically significant (p > 0.05). In rabbits with adhesion formation, it was determined that TNF-α levels increased at the 6th postoperative hour (p < 0.05), and IL-6 levels increased at the 6th, 24th and 48th postoperative hours (p < 0.001) compared to preoperative. In this study, treating tissues with 0.4% HA and 1% CMC solutions suppressed peritoneal inflammation, and this resulted in an increase in the levels of proinflammatory cytokines TNF-α and IL-6. It has been determined that the application of these solutions reduces postoperative adhesion formation. It was concluded that TNF-α and especially IL-6, which are proinflammatory cytokines, can be a non-invasive biomarker in determining postoperative IA formation and evaluating the adhesive process. © 2024 Polskie Towarzystwo Nauk Weterynaryjnych. All rights reserved.Article Effects of Desflurane and Isoflurane on Postanaesthetic Recovery Characteristics With Hepatic and Renal Functions in Dogs(Medwell online, 2009) Altug, M. E.; Gonenci, R.; Durgut, R.; Karasu, A.; Abdulhayoglu, B.The aim of the study, was to investigate the effects of desflurane and isoflurane on postanaesthetic recovery characteristics with hepatic and renal functions in dogs. Sixteen adult mongrel dogs of both sexes weighing between 16-21 kg were equally divided into 2 groups. Anaesthesia was induced with 0.3 mg kg(-1) midazolam and 10 mg kg(-1) thiopental intravenously. Maintenance of anaesthesia was continued with 7.2-8% desflurane or 1.3-1.5% isoflurane. Heart and respiration rates, arterial haemoglobin oxygen saturation and rectal temperatures were monitored before, during, 1 h and 1 day after anaesthesia periodically and postanaesthetic recovery score times were also observed. Serum alanine aminotransferase, aspartate aminotransferase, gamma-glutamyltransferase, blood urea nitrogen, total bilirubin and creatinine levels were measured in venous blood samples. Postanaesthetic recovery scores including time to standing, time to reaching sternal recumbency, eye opening and time to extubation were found shorter in desflurane group than isoflurane group (p<0.05). Alanine aminotransferase, aspartate aminotransferase and gamma-glutamyltransferase activities were non-significantly increased in the isoflurane group and there were no change between groups. Total bilirubin and creatinine levels were non-significantly decreased, blood urea nitrogen levels were non-significantly increased within normal range during and after the anaesthesia in both groups. The present study reveals 2 important results. Firstly, desflurane provided faster anaesthetic induction, recovery with less excitation and rapid recovery times (p<0.05) and the dogs stood up in shorter time than those of isoflurane. Secondly, this study indicates that both volatile agents have no harmful side effects on renal and hepatic functions in dogs.Article The Effects of Sildenafil Citrate and Vitamins A, C and E on Testicular Damage in Alloxan-Diabetic Rats(Medwell Journals, 2012) Uslu, B.A.; Ilhan, F.; Gulyuz, F.; Karasu, A.; Tas, A.; Sendag, S.; Wehrend, A.Sildenafil citrate is an active ingredient used successfully in the treatment of erection disorders caused by a variety of factors. The purpose of this study is to research the effects of Sildenafil citrate and vitamins A, C and E on testicular degeneration in rats with alloxan diabetes. The rats were divided into II groups; healthy control and diabetic groups. The diabetic rats were divided into X groups with ten animals in each group. Group I consisted of control diabetic rats that were given only distilled water, Group II was the diabetic group treated with glibenclamide; Group III was the diabetic group treated with insulin, Group IV was the diabetic group treated with Sildenafil citrate, Group V was the diabetic group treated with Vitamin A, Group VI was the diabetic group treated with vitamine, Group VII was the diabetic group treated with vitamin E, Group vni was the diabetic group treated with Sildenafil citrate and vitamin A, Group IX was the diabetic group treated with Sildenafil citrate and vitamin C and Group X was the diabetic group treated with Sildenafil citrate and vitamin E. Testis tissue samples were collected on the 3, 7 and 15th days of treatment in control and diabetic groups. In the histopathological examination of the testicles of diabetic rats, the seminiferous tubules and interstitium of testes in the control groups were normal and the complete spermatogenic cells in the seminiferous tubules were healthy and uniformly arranged. Severe degeneration and tubular atrophy was observed in the seminiferous tubules on the 3, 7 and 15th days in the group with induced alloxan diabetes given only distilled water (Group I). The lesions on day 3rd in the diabetic group given glibenclamide (Group II) were similar to those in the diabetic control group given only distilled water. All of the tubules had been affected by the 3rd day in group three which was given insulin. A small number of spermatogenic cells and giant cells were observed in some tubules. However, almost al of the degenerated tubules had healed on the 7 and 15th days. Even though, recovery on day 15th was very similar in Groups IV-X recovery was much more pronounced in Groups VIII-X beginning from day 3rd. © Medwell Journals, 2012.Article Effects of Sildenafil Citrate on the Hematological Parameters in the Early Phase of Wound Healing in Diabetic Rats(Academic Journals inc, 2011) Tas, A.; Karasu, A.; Comba, B.; Aksu, D. S.; Duz, E.; Tanritanir, P.In this study Investigation of effects of sildenafil citrate on some of the hematological parameters in the early phase of wound healing in diabetic rats was aimed in this study. Total 30 rats were used in the study. Rats were divided into three groups equal in number (control-non diabetic, diabetic and diabetic+Sc). While no treatment was given to the control-non diabetic and diabetic groups, sildenafil citrate was given to the diabetic+Sc group intraperitoneally with a dosage of 0.7 mg/kg/day dosage for 3 days after creating wounds: under ketamine anesthesia, a circular, full thickness, standard wound was created on the back of each rat by using a 0.8 cm sterile punch. Cardiac blood was drawn in tubes containing EDTA from each group before the creation of wounds and on days 1, 2 and 3 following the creation of wounds. Differential leukocyte counts were determined in blood samples containing anticoagulant. For this purpose, blood smears were prepared from blood samples and stained with Giemsa method. It was found that, while neutrophil counts increased in all the groups during the wound-healing periods with the increasing number of days, lymphocyte counts reduced and in control and Sc+diabetic groups, monocyte counts increased. As a result, it was concluded that Sc plays an important role in eliminating the pressure on the cellular activities in the early period of wound healing and continuance of the inflammatory process and wound healing within normal limits and also that this could be demonstrated with hematological analyses.Article Evaluation of the Anesthetic Effects of Xylazine-Ketamine, Xylazine-Tiletamine and Tiletamine-Zolazepam Using Clinical and Laboratory Parameters in Rabbits(Polskie Towarzystwo Nauk Weterynaryjnych, 2018) Karasu, A.; Altug, N.; Aslan, L.; Bakir, B.; Yuksek, N.The aim of this study was to evaluate the anesthetic effects of xylazine-ketamine (XK), xylazine-tiletamine-zolazepam (XTZ) and tiletamine-zolazepam (TZ) using hematological and biochemical parameters in rabbits. ' A total of 18 rabbits were divided into three equal treatment groups (n = 6). The rabbits in the XK, XTZ, and TZ groups were administered xylazine (5 mg/kg) and ketamine (50 mg/kg), xylazine (5 mg/kg) and TZ ; (15 mg/kg), and TZ (15 mg/kg), respectively, via the intramuscular route. Following the injection, their reflexes ; were tested every 5 minutes. Heart rate, respiratory rate, and body temperature were determined before ; the injection (0 min) and at 5, 15, 30, 45, 60, 75, 90, and 120 min after injecting the anesthetic combinations. Furthermore, hematological and biochemical (alanine transaminase [ALT], aspartate transaminase [AST], alkaline phosphatase [ALP], bilirubin, blood urea nitrogen [BUN], and urea) analyses were also performed before, during, and after anesthesia administration. The duration of anesthesia and loss of reflexes were significantly longer in the XTZ group than in the other groups. However, in the TZ group, reflexes were remained. Respiratory rate and body temperature decreased in all the groups. Moreover, heart rate reduced only in the XK and XTZ groups, and the hematological parameters of all groups were comparable. Serum AST and ALP levels increased in the XTZ group compared to that in the XK and TZ groups, respectively. However, these increases were within the reference limits. The post-anesthesia serum BUN and urea levels significantly increased in the XTZ group (p < 0.05) compared to that in the other groups. Thus, although the XTZ combination provided satisfactory anesthetic effect in rabbits, it may be nephrotoxic. Therefore, its use for anesthesia induction in invasive renal procedures and experimental nephrotoxicity studies is not advisable. © 2018 Polskie Towarzystwo Nauk Weterynaryjnych. All rights reserved.Article Extremity Problems in Ostrich Chicks and Their Treatment(2009) Asian, L.; Genccelep, M.; Karasu, A.; Duz, E.; Alkan, I.; Bakir, B.Extremity problems in 120 ostrich chicks were investigated in this study. Fledglings were followed for 1 year and extremity problems were evaluated as congenital or acquired. In the study, congenital lower extremity problems were found in 17.5% of the cases (both extremities extended sideways m 10 cases, 1 extremity extended sideways in 3 cases, one leg extended forward and the other back in 4 cases, big toe retrovert in 2 cases, 5th toe deviated towards underneath of the big toe, totaling to 21 cases) and acquired lower extremity problems in 19.64% of the cases (tibiotarsal luxation in 6 cases, tarsal bone fracture in 1 case, fractures in radius-ulna in 1 case, injuries of the extremities in 9 cases and arthritis in the tarsal joint in 2 cases and in proximal mterphalangeal joint in 3 cases, totaling to 22 cases). Extremity problems were found in 43 (35.83%) ostrich chicks out of 120 (congenital in 21 cases and acquired in 22). Of these 43 cases, it was observed that 29 (67.44%) were healed with the treatment applied, whereas, 14 (32.56%) did not heal. It was concluded in the study that the extremity problems causing great economic losses in ostrich husbandry can be minimized with prevention, early diagnosis and proper treatment. © Medwell Journals, 2009.Article Histopathological and Immunohistochemical Study of the Effect of Sildenafil Citrate, Vitamin A, Vitamin C and Vitamin E on Wound Healing in Alloxan-Induced Diabetic Rats(Univ West indies Faculty Medical Sciences, 2021) Tas, A.; Karasu, A.; Yener, Z.; Yildirim, S.; Atasoy, N.; Duz, E.; Yavuz, E.Objective: To examine the impact of sildenafil citrate, vitamin A (Vit A), vitamin C (Vt C) and vitamin E (Vit E) on wound healing in a rat model of diabetes (n = 100). Methods: Diabetes was induced in the rats by intraperitoneal (i.p.) administration of alloxan. After anaesthesia, a standard wound was created on the back of each rat using a 10-mm sterile punch. The rats were randomly divided into 10 groups (n = 10 in each), as follows: normal saline, glibenclamide, insulin, sildenafil, Vit A, Vit C, Vit E, Vit A + sildenafil, Vit C + sildenafil, Vit E + sildenafil daily for 15 days. The rats were sacrificed after being anaesthetized 3, 7 and 15 days later Wounded skin tissue samples were collected for histopathological and immunohistochemical analyses. Results: On the 7th day, epithelial regeneration was completed in groups 8 and 9. Angiogenesis was insufficient in group 2. In terms of connective tissue proliferation, partially matured connective tissue was observed in group 4. On the 15th day of the study, groups 8, 9 and 10 had mature connective tissue. However; group 1 still had exudate-containing neutrophils. Immunohistochemically, on the 3rd day, the level of inducible nitric oxide synthase (iNOS) reactivity was intense in macrophages and neutrophils surrounding the wound edges in groups 4, 8, 9 and 10. The level of iNOS reactivity was moderate in group 6 and less distinct in groups 1, 2, 3, 5 and 7. Conclusion: Sildenafil citrate, together with Vit A and Vit C, is beneficial in wound healing of diabetic rats.
