Article Aspiculuris Tetraptera ile Doğal Enfekte Farelerde Yerli ve Kontrolsuz Levamizolun Karşılaştırılmalı Etkisi(2007) Keles, Ihsan; Akkan, Hasan Altan; Özdal, Nalan; Türel, İdris; Özbek, Hanefi; Ayaz, ErolBu çalışma, Aspiculuris tetraptera ile doğal enfekte farelerde iki farklı ülkede (İran ve Türkiye) üretilen levamizol (Levamisole) preparatlarının etkinliklerini karşılaştırmak amacıyla yapıldı. Bu amaçla, Yüzüncü Yıl Üniversitesi Tıp Fakültesi Deney Hayvanları ünitesinden temin edilen farelerin santrifüj flotasyon yöntemi ve selofan bant tekniği ile parazitolojik muayeneleri yapıldı. Yukarıdaki muayenelerle A.tetraptera ile enfekte oldukları tespit edilen fareler 3 gruba ayrıldı. Birinci gruptaki hayvanlara (8 fare) İran orijinli levamizol, ikinci gruptaki hayvanlara (8 fare) Türkiye orijinli levamizol uygulanırken, üçüncü grup fareler (6 fare) tedavi edilmeyen grup olarak kullanıldı. Her iki levamizol preparasyonu da 10 mg/kg dozunda verildi. İlaç uygulamasından sonra bütün gruplardaki farelerin dışkı örnekleri 7 gün süre ile alınarak incelendi. Sekizinci gün bütün fareler insancıl bir şekilde inhalasyon anestezisi kullanılarak öldürüldü. Ötenazi sonrası bağırsaklardaki parazit sayımları da yapıldı. Sonuç olarak, kontrol edilemeyen sınır ticareti ile İran’dan getirilen levamizol %69,3 oranında, Türkiye’de üretilen levamizol ise %91,7 oranında doğal olarak A. tetraptera ile enfekte farelerde etkili oldukları belirlendi. Elde edilen bulgular istatistiksel olarak karşılaştırıldı ve farklılık önemli (P<0.001) bulundu.Article Atropine Sulphate Test Can Be an Aetiologic Indicator of Vagal Bradycardia Developed in a Megaoesophagus Case(Natl veterinary Research inst, 2007) Keles, Ihsan; Altug, Nuri; Kaya, Abdullah; Ozkan, Cumali; Yuksek, Nazmi; Aslan, LogmanAn acquired megaoesophagus in a calf has been described. The diagnosis of megaoesophagus was made based on clinical and laboratory examinations, using contrast radiography, and necropsy findings. Atropine sulphate was used to enlighten the relationship between damage in the N. vagus and bradycardia, which developed in the megaoesophagus case.Article Biotyping and Serotyping of Mannheimia (Pasteurella) Haemolytica Isolated From Lung Samples of Slaughtered Sheep in the Van Region(Tubitak Scientific & Technological Research Council Turkey, 2007) Ilhan, Ziya; Keles, IhsanA total of 584 lung samples of slaughtered sheep having clinical symptoms of pneumonia were investigated microbiologically. Out of the 584 samples, 66 (11.3%) Mannheimia (Pasteurella) haemolytica strains were isolated on the basis of cultural, morphological and biochemical characteristics. Sixty- six isolates of M. haemolytica were biotyped by carbohydrate fermentation tests and serotyped by coagglutination and indirect haemagglutination (IHA) tests. Out of the 66 M. haemolytica strains, 57 (86.3%) were detected as biotype A, and 9 (13.6%) were biotype T. The 66 M. haemolytica isolates were serotyped as follows by coagglutination test: 13 (19.6%) A2, 11 (16.6%) A1, 9 (13.6%) A6, 7 (10.6%) A9, 4 (6.0%) A5, 4 (6.0%) A7, 4 (6.0%) A12, 3 (4.5%) A8, 1 (1.5%) T4, 1 (1.5%) A13, and 1 (1.5%) T15. Of the M. haemolytica strains, 14 (21.2%) belonged to serotype A2, 11 (16.6%) to serotype A1, 10 (15.1%) to serotype A6, 7 (10.6%) to serotype A9, 4 (6.0%) to serotype A5, 4 (6.0%) to serotype A7, 4 (6.0%) to serotype A12, 4 (6.0%) to serotype A8, 1 (1.5%) to serotype T4, 1 (1.5%) to serotype A13 and 1 (1.5%) to serotype T15 by IHA test. Eight (12.1%) and 5 (7.5%) isolates were unserotyped by coagglutination and IHA tests, respectively. T3, T10, A11, A14 and A16 serotypes were not determined.Other Bir Van Kedisinde Hepatosellüler Karsinom(2001) Keles, Ihsan; Karaca, Mehmet; Yener, ZabidBu raporda, 5 yaşlı dişi bir Van kedisinde belirlenen generalize metastazlı hepatosellüler karsinom olgusu tanımlandı. Makroskobik olarak, karaciğer, dalak, akciğer, böbrekler, karın boşluğu ve deri altında 1mm-6 cm çapına varan, grimtırak-beyaz renkte tümöral nodüller belirlendi. Mikroskobik olarak, tümör hücreleri solid, kordon ve trabeküler yapılar oluşturmuştu. Neoplastik hücreler genellikle poligonal ve veziküler çekirdekli olup çok sayıda mitotik figürlere sahipti. İmmunohistokimyasal olarak tümör markerlerinden alfa-fetoprotein (AFP), sitokeratin, düz kas aktin (SMA) ve faktör VIII'e ilişkin antijen (FVIII-Rag) araştırıldı. Bunlardan AFP ve SMA pozitif bulundu. The disease occurs especially on the unpigmented areas such as tip of the ears, nose, body and extremities as a result of phototoxic reactions when subjected to ultraviolet lights. The disease has been reported to come into view as chronic dermatitis in cats having white ear and blue eyes. (Haziroglu and Milli, 1998). The pathogenesis of feline dermatitis is thought to occur as a result of free radicals released after phototoxic reactions (Soter, 1990).Article Determination of Adenosine Deaminase Activity in Cattle Naturally Infected With Theileria Annulata(Springer, 2008) Altug, Nuri; Yuksek, Nazmi; Agaoglu, Zahid Tevfik; Keles, IhsanThe purpose of this study was to determine serum ADA activity in cattle naturally infected with Theileria annulata. In this study, a total of 37 cross-bred cattle which 27 of it showing clinical signs of theileriosis constituted infected group and 10 healthy cattle as control group were used as animal materials. Infected group divided into three groups according to their PCV values. Cattle with PCV >= 25 were put on group I (n=9), those with PCV 13-24 were put on group II (n=11) and those with PCV >= 12 were put on group III (n=7). Microscopical diagnosis of the disease was also made. Hematological parameters, serum enzyme activities (ADA, AST, ALT and ALP) were determined in all cattle. Hematological results revealed that significant progressive decreases in HGB, PLT, PBML counts and ratios from group I onwards to group III, whereas the WBC, PBPL counts and ratios showed an increase from group I onwards to group III. The serum ADA, AST, ALT and ALP activity increased significantly in all infected groups compared to control group. However, these parameters were also observed to decrease progressively from group I to group III. Furthermore, the highest increase in enzyme activities observed in the infected group I. But, these enzyme's activities started to decrease in infected group II and III in parallel with PBML and PLT counts. Eventhough, this decrease did not reach to the values obtained from control group. On the contrary, PBPL counts and ratios increased in infected group II and III in contrast to decrease in PCV. As a result, increased serum ADA activity in tropical theileriosis may reflect the involvement of the cellular immune responses.Article Effeciency of Various Supportive Treatments as a Cure for Anaemia in Cattle With Theileriosis(Chulalongkorn Univ, 2014) Altug, Nun; Yuksek, Nazmi; Keles, Ihsan; Ozkan, Cumali; Yoruk, Ibrahim Hakki; Arslan, SezaiThe purpose of this study was to investigate the efficiency of various supportive treatments on the restoration of anaemia in theileriosis. A total of 20 cattle infected with theileriosis received intramuscularly a single dose of buparvaquone (2.5 mg/kg(-1) bw) and were divided into 4 equal groups according to the different therapy options: whereas no supportive treatment was applied in group 1, administrations of vitamin B complex, antioxidant vitamins (A, D, E and C), minerals and trace elements were performed in groups 2, 3 and 4, respectively. Haematological parameters, serum concentrations of antioxidant vitamins, minerals and trace elements as well as serum glutathion peroxidase (GSH-Px) and superoxide dismutase (SOD) activities were determined before treatment and 7 days after in infected animals and compared to control values from 10 healthy cattle. In the theileriosis affected cattle, remarkable decreases in haematocrit values, haemoglobinaemia and platelet counts were associated to decreased serum alpha- / gamma-tocopherol, vitamin C, vitamin B-12, Ca, Mg, Zn, Na, GSH-Px and SOD levels and to increased sideraemia. The iron concentrations remained elevated and some antioxidants (alpha- / gamma-tocopherol and vitamin C concentrations and GSH-Px and SOD activities) decreased in all treated cattle. Haematocrit values, haemoglobinaemia and vitamin B-12 concentrations also remained depressed but maximal increases were recorded in cattle receiving vitamins B as supportive therapy (group 2). In parallel, platelet numeration was significantly restored in all groups, but specifically in group 2. On the other hand, the maximal increases in alpha-tocopherol and Zn concentrations recorded in groups 3 and 4, respectively, were not associated with cure of anaemia. These results show that the most efficient supportive therapy in the cure of anaemia during theileriosis was vitamin B administration.Article The Efficacy of Ricobendazole and Ivermectin on Naturally Infected Sheep With Trichostrongylidae Sp in the Region of Van(Medwell online, 2009) Sahin, Ali; Gul, Abdurrahma; Karaca, Mehmet; Akkan, Hasan Altan; Keles, IhsanThe efficacy of two broad spectrum antelmentics (Ricobendazole and Ivermectin) against gastrointestinal nematode infection was compared in sheep. Naturally infected thirty sheep with Trichostrongylidae sp., which were diagnosed by McMaster technique and their coprocultures were performed. Sheep were randomly assigned to two treatment groups (Ricobendazole and Ivermectin) and one control group of 10. Fecal samples for determining nematode egg counts were collected pre- and post-treatment. All treatments resulted in decrease in the number of Eggs per Gram (EPG) in the post-treatment samples compared to the pre-treatment samples. Mean egg counts reduced 100% for both ricobendazole and ivermectin on trial 14 days. Furthermore, no significant differences were found among treatment groups. The data demonstrate a similar efficacy obtained from both ricobendazole and ivermectin. They were effective and convenient to apply. Either ricobendazole or ivermectin believed to be suggestible to treat gastrointestinal nematodes in sheep.Article Investigation of the Seroprevalance of Maedi-Visna in the Region of Van Using Elisa and Histopathological Findings(Medwell online, 2009) Akkan, Hasan Altan; Karaca, Mehmet; Tutuncu, Mehmet; Keles, Ihsan; Ilhan, Fatma; Cetin, Yunus; Tasal, IbrahimIn the present study, seroprevalance of maedi-visna in sheep in the region of Van was aimed to investigate. A total of 465 serum samples obtained from sheep raised in Van and its towns were investigated with regard to maedi-visna specific antibody using ELISA procedure. Out of 465 samples 30 of them were seropositive (6.45%) and 20 of them were doubtful (4.30%) according to test procedure. Histopathology was also applied to some of these sheep which slaughtered in an abattoir after blood sampling. According to histopathological analysis 9 doubtful samples were also found to be positive. But, due to mild lymphoproliferation in there samples ELISA gave doubtful results. As a result with this study, seroprevalance of maedi-visna was determined in the region of Van and in the doubtful cases histopathological assessments believed to strengthen serological findings.Article Investigation on the Effect of P. Haemolytica A1 Leukotoxin on Some Cellular Functions(2001) Keles, IhsanPasteurella haemolytica ile ilgili asıl virulens faktörün ruminantlarm lökositlerini tahrip eden bir toksin (leukotoxin) olduğuna inanılmaktadır. Bu nedenle, bu toksinin kuzu ve buzağıların monositlerinden elde edilen monositik hücre hatlarının (MCL) latex beads'lerini fagosite etmeleri üzerine olan etkilerini, toksine karşı bu hücre hatlarının ve mononükleer hücrelerin (MNC) duyarlılıklarım ve öldürücü dozun altındaki leukotoxin 'in kuzuların mononükleer hücrelerinin bir mitojen olan phytohaemagglutinin (PHA) 'e cevabı üzerindeki etkileri araştırıldı. Sonuç olarak, mononükleer hücreler P. haemolytica A1' den elde edilen leukotoxinin 1/2 sulandırmasına karşı bile yaklaşık % 56 oranında lize olurken, monositik hücre hatları üzerine herhangi bir sitolitik etkisi görülmedi. Aynı toksin bu hücre hatlarının fagositik aktivitelerini de deprese edemediği gözlendi. Öte yandan, leukotoxinin öldürücü dilüsyonlarımn altındaki dilüsyonları, mononükleer hücrelerin bir mitojen olan PHA 'ya cevabını deprese ettiği ve bu depresyonun rat growth faktörü (RGF) tarafından tamamen, inteleukin l ve 2 tarafından ise kısmen ortadan kaldırıldığı saptandı.Article Investigation on the Seroprevalence of Toxoplasmosis, Listeriosis and Brucellosis in Goats Living in the Region of Van,turkey(2007) Çelebi, Bekir; Kılıç, Selçuk; Akkan, Hasan Altan; Keles, Ihsan; Uslu, Barış Atalay; Karaca, Mehmet; Babür, CahitBu çalışma Van yöresi keçilerinde toxoplasmosis, listeriosis ve brucellosis seroprevalansını belirlemek için yapıldı. Toxoplasma gondii, Listeria monocytogenes ve Brucella melitensis’e karşı oluşan antikorları belirlemek için sırası ile Sabin Feldman Dye Testi (SFDT), Osebold Agglutination Testi (OAT) ve Micro Agglutination Testi (MAT) uygulandı. 98 serum örneğinin, %80.61 toxoplasmosis, %34.69 listeriosis and %6.12 brucellosis yönünden seropozitif olduğu tesbit edildi. Bu sonuçlar ile Van yöresi keçilerinde Toxoplasmosisin, listeria ve brucella’ya göre çok daha yaygın olduğu görüldü.Other Kan Parazitli Hayvanlarda Sialik Asit ve Lipid-bağlı Sialik Asit Düzeylerinin Araştırılması(2000) Ekin, Suat; Ertekin, Ali; Akkan, Hasan Altan; Keles, Ihsan; Karaca, MehmetBu çalışmada 6 ay - 5 yaş arası, farklı ırk ve cinsiyette toplam 42 adet .sığır kullanıldı. Şunlardan 12 tanesi theileriozisli, 10 anaplazmozisli, 10 tanesi theileriozis ve anaplazmosisli, 10 tanesi ise klinikman sağlıklı hayvanlardan oluştu. Yapılan analizlerde kontrol grubundan elde edilen değerlere kıyasla, sialik asit miktarlarındaki artış tüm enfekte gruplarda önemli (p<0.001), lipid-bağlı sialik asit düzeylerinde ise theileriozis grubu hariç diğer gruplarda anlamlı (p<0.01) bulundu. Aneminin düzeyi theileriozis grubunda fazla olmasına rağmen, anaplazmanın bulunduğu hayvanlarda sialik asit ve lipid-bağlı sialik asit düzeyleri daha yüksek bulundu.Article The Levels of Trace Elements and Selected Vitamins in Goats With Chronic Fluorosis(Tubitak Scientific & Technological Research Council Turkey, 2013) Altug, Nuri; Arslan, Sezai; Yuksek, Nazmi; Keles, Ihsan; Yoruk, Ibrahim Hakki; Basbugan, Yildiray; Aytekin, IsmailThis study was conducted to investigate the effects of chronic fluorosis on trace elements and vitamin levels in goats. Thirty-three goats exhibiting clinical signs of chronic fluorosis and 10 healthy goats from the same region were used. Animals with chronic fluorosis were divided into 2 groups according to their age as 1-3 years old and 4-6 years old. Blood samples were collected from all goats. Fluoride concentrations were determined with an ionometer. Na, K, and Cl levels were measured with ion-selective equipment and serum mineral substance levels were obtained by atomic absorption spectrophotometer. Ca, Mg, and P levels were determined by biochemistry analyzer. Serum vitamin concentrations were determined by liquid chromatography. The levels of serum copper, iron, manganese, and nickel decreased in goats with chronic fluorosis, whereas other trace elements did not change. Among vitamins, levels of serum a-tocopherol were increased, whereas retinol and vitamin D3 levels were not changed significantly differently between the fluorosis and control groups. In evaluation of goats with fluorosis according to age group, copper levels in the age group of 1-3 years and iron levels in the age group of 4-6 years were found to be lower than in the other age groups. Other minerals and all vitamin parameters were not different according to age. The results of the present study clearly showed that serum copper, iron, manganese, and nickel levels were reduced in goats with chronic fluorosis, and so goats with fluorosis should receive support in terms of these concerned trace elements.Article Relationship Between Packed Cell Volume Levels and Serum Nitric Oxide Concentrations in Cattle With Tropical Theileriosis(Univ Agriculture, Fac veterinary Science, 2015) Ozkan, Cumali; Altug, Nuri; Akgul, Yakup; Keles, Ihsan; Kaya, Abdullah; Yuksek, Nazmi; Yur, FatmagulThis study was performed to determine changes in serum NO (nitric oxide) levels in cattle with theileriosis in accordance with PCV levels. For this purpose, 42 cattle with theileriosis and 10 healthy cattle have been used as material. Infected animals have been divided into 3 equal groups. 1st group had <12%, 2nd group had 13-24% and 3rd group had <25% PCV level. Blood samples have been used to determine NO concentrations. Serum NO concentrations prior to treatment increased in all infected groups compared to control group however its levels were significant only in the third group. NO concentrations decreased to almost normal values after treatment especially in 2nd and 3rd group, however this decrease was not significant in the first group. As a result, NO levels have been found increased in cattle with natural theileriosis. Therefore, its concluded that NO level and PCV levels in theileriosis should be studied with more detail in the future to enlighten the role of NO release and strategies for controlling its' release should be made especially in severe cases. A tentative diagnosis of complete rupture of the muscle was based on information obtained from both anamnesis and clinical signs. The definitive diagnosis was made on necropsy. In this study, blood samples randomly collected from 465 sheep were analysed for the presence of antibodies to pestiviruses (bovine viral diarrhea virus, border disease virus) using an enzyme-linked immunosorbent assay in the province of Van and their towns. The seroprevalance were estimated as 75.9% and 60.0-82.5% in the sampled animals and sampled towns, respectively. The results revealed that pestiviruses are important abort pathogens in the province of Van and their towns.Article Seroprevalence of Q Fever in Cattle and Sheep in the East of Turkey(Academic Journals inc, 2009) Ceylan, Ebubekir; Berktas, Mustafa; Keles, Ihsan; Agaoglu, ZahitThe present study was carried out to determine the seroprevalence of antibodies to C. burnetii in cattle and sheep in the east of Turkey. Serum samples collected randomly from 92 cattle and 92 sheep were examined by ELISA (Vircell-SL, Spain) to detect IgG antibodies against C. burnetii phase II antigen. Seropositivity was observed in 16.3% of the cattle and in 5.4% of the sheep. Coxiellosis has an important seropositivity in both cattle and sheep and it can cause serious health problem in humans living in Eastern Turkey.Article Serum Adenosine Deaminase Activity in Domestic Animals: Reference Values(Tubitak Scientific & Technological Research Council Turkey, 2009) Altug, Nuri; Yuksek, Nazmi; Agaoglu, Zahid; Keles, IhsanThis study was aimed to determine normal serum adenosine deaminase (ADA) activity in domestic animals in both sexes and that were considered healthy on the basis of clinical and laboratory examinations. In this study, 262 non-pregnant animals of both sexes, belonging to 5 species [cattle (n = 50), sheep (n = 65), goat (n = 52), dog (n = 55), and cat (n = 40)] were used. Serum total ADA and ADA1 isoenzyme activities were found to be 6.34 +/- 0.31 IU/l and 6.34 +/- 0.31 IU/l in cattle, 7.57 +/- 0.30 IU/l and 7.31 +/- 0.47 IU/l in sheep, 7.52 +/- 0.36 IU/l and 6.44 +/- 0.72 IU/l in goat, 2.56 +/- 0.13 IU/l and 2.03 +/- 0.30 IU/l in dogs, 23.36 +/- 1.59 IU/l and 21.71 +/- 2.13 IU/l in cats, respectively. The serum levels of ADA2 were low in sheep, goats, dogs, and cats, whereas in these species ADA1 was high. Furthermore, there was no serum ADA2 activity in cattle. Additionally, there were no significant differences between genders in ADA activities in all species. As a result, determination of serum normal ADA and its isoenzyme activities in domestic animals will be useful for scientists who want to investigate ADA activity in future in different diseases.Article Serum Iron, Total Iron-Binding Capacity, Unbound Iron-Binding Capacity, Transferrin Saturation, Serum Copper, and Hematological Parameters in Pregnant Akkaraman Ewes Infected With Gastro-Intestinal Parasites(Tubitak Scientific & Technological Research Council Turkey, 2006) Kozat, Suleyman; Yuksek, Nazmi; Goz, Yasar; Keles, IhsanThe aim of this study was to investigate serum iron parameters, and copper and iron deficiency anemia in ewes infected with gastro- intestinal parasites. This study was conducted with 60 pregnant Akkaraman ewes. In the statistical analyses, values obtained from both parasite infected and uninfected ewes were compared. Significant decreases were observed in PCV (P < 0.05), Hb (P < 0.05), RBC (P < 0.05), total protein (P < 0.05), albumin (P < 0.05), globulin (P < 0.05), and MCHC (P < 0.01), and a significant increase was seen in the WBC value (P < 0.05) in the parasite infected group. Furthermore, the parasite infected group had lower serum Fe (P < 0.001), TS % (P < 0.001), and serum Cu (P < 0.01) levels, and had higher UIBC values (P < 0.05) compared to the values obtain from the uninfected group. Therefore, in addition to antiparasite drug treatment in parasite infected pregnant ewes, iron preparations should also be administered in order to treat iron deficiency.Article Serum Zinc, Copper and Thyroid Hormone Concentrations in Heifers With Retarded Growth(2006) Altuğ, Nuri; Ceylan, Ebubekir; Keles, Ihsan; Dönmez, NurcanBu çalışmada gelisme geriligi problemleri olan düvelerde Zn, Cu ve tiroid hormon konsantrasyonlarının rolünü arastırmak amaçlandı. Çalısmada Yüzüncü Yıl Üniversitesi Veteriner Fakültesi İç Hastalıkları kilinigine getirilen 6-12 aylık yaşlarda 35-50 kg canlı agırlıgında 11 adet (Test Grubu) ve normal canlı ağırlığında (80-100 kg) 6 adet (Kontrol Grubu) Dogu Anadolu Kırmızısı düve materyal olarak kullanıldı. Klinik olarak enfeksiyon ve paraziter enfestasyon teshis edilemedi. Bu nedenle, büyüme çagındaki düvelerde gelisme geriligine neden oldugu bilinen iz element ve troid hormon durumları arastırıldı. Kontrol grubundan elde edilen degerlerle karsılastırıldıgında test grubunda Zn, Cu, T3, FT3, T4 ve FT4 degerlerinin daha düsük olmasına ragmen, sadece Zn (p<0.05), T4 ve FT4 (p<0.01) degerlerindeki farklılıklar istatistiksel olarak önemli bulundu. Bu nedenle, özellikle Zn ve Cu içeriginden yetersiz ve dengesiz beslenmenin, test grubunda absorbsiyon bozukluklarına yol açarak growth hormonunun stimülatörü olan tiroid hormon konsantrasyonları ve gıda degerlendirilmesindeki azalmaya baglı olarak gelisme geriligine neden oldugu sonucuna varıldı.
