Article Analysis of Phenolic Compounds by Lc-Hrms and Determination of Antioxidant and Enzyme Inhibitory Properties of Verbascum Speciousum Schrad(Acg Publications, 2023) Kiziltas, Hatice; Bingol, Zeynebe; Goren, Ahmet C.; Alwasel, Saleh H.; Gulcin, IlhamiStudies have shown an inverse correlation between age-related illnesses like coronary heart disease and cancer and fruit and vegetable intake. Given the probable health benefits of natural antioxidants from plants, research on them has increased. Verbascum L is a large genus of Scrophulariaceae family and 323 species distributed worldwide. Since Verbascum L. species are plants that are grown in many regions of Turkiye and are used in folk medicine, it is important to evaluate the biological activity of these species. In this study antioxidant properties of Verbascum speciosum Schrad. were investigated. The antioxidant capacities of water and ethanol-based extracts obtained from air parts were evaluated with Fe3+ reducing, CUPRAC, FRAP, DPPH center dot, and ABTS(center dot+) scavenging antioxidant methods. Extracts were also examined to determine their AChE, alpha-glycosidase and alpha-amylase enzyme inhibitions. (WEFO) several in vitro antioxidant methods such as ABTS(center dot+) scavenging activity, DPPH. scavenging activity, Fe(3+)reduction method, cupric ions (Cu2+) reduction capacity, and metal ion (Fe2+)-binding activities using ferrozine reagent were separately performed. Also, BHT, alpha-tocopherol and ascorbic acid were used as the standard antioxidant molecules. Moreover, some phenolic compounds that are responsible for antioxidant abilities of EEFO and WEFO were determined by LC-HRMS. EEFO and WEFO demonstrated effective antioxidant abilities when compared with the standards. Subs), some putative antioxidant methods such as DPPH. scavenging activity, ABTS(center dot)(-) scavenging effect, ferric ions (Fe3+) reduction method, cupric ions (Cu2+) reducing capacity, and ferrous ions (Fe2+) binding activities were separately performed. Also, ascorbic acid, alpha-tocopherol and BHT were used as the standard compounds. Additionally, some phenolic compounds that responsible for antioxidant abilities of EESB and WESB were screened by liquid chromatography-high resolution mass spectrometry (LC-HRMS). Co). In this study, vitamin C, antioxidant and antiradical properties were determined using spectrophotometer. The results are compared with the reference antioxidants such as trolox, alpha-tocopherol and BHT. Levels of vitamin (A, E) measured using HPLC method. Trace elements were carried out using method of dry ashing with ICP-MS. The results of this study showed that C. meyeri leaf has a high antioxidant capacity and vitamin levels. C. meyeri is thought to be used as additives for food products and pharmaceutical industries with appropriate antioxidant properties and an antioxidant in future studies of experimental animal models, against free radicals generated in response to oxidative stress.Article Antioxidant Activity of Lyophilized Water Extract of Aerial Parts of Italian Bugloss (Anchusa Azurea Mill.)(Kahramanmaras Sutcu Imam Univ Rektorlugu, 2022) Kiziltas, HaticeIn many studies, an increase in fruit and vegetable consumption has been shown to reduce the death rate from age-related diseases such as coronary heart disease and cancer. Italian bugloss (Anchusa azurea Mill.), in some regions of Turkey is a plant consumed as a vegetable and used in traditional medicine therefore it is important to evaluate the biological activity. Boiss. (Apiaceae) (FASB) is used to treat liver diseases and has been used both as food and therapeutics by many cultures for thousands of years because of the natural antioxidant compounds. Objective: This study determines antioxidant properties of FASB flowers, the levels of minerals and vitamins, and also, evaluates the hepatoprotective effect of flowers against N-nitrosodimethylamine (NDMA) induced on liver tissue by assessing antioxidant enzymes and histopathological parameters in Wistar albino rats. Materials and methods: In the study, the rats were divided into six groups of ten. Control, untreated animals were given 0.9% NaCl. Rats were intraperitoneally given NDMA (10 mg/kg) for the first 7 days. FASB methanol extract (150 and 300 mg/kg) was administered orally for 21 days. Results: a-Tocopherol, retinol, ascorbic acid, total antioxidant activity, phenolic and flavonoid contents of FASB were 0.70 +/- 0.13, 0.29 +/- 0.03 mu g/g, 139.32 +/- 7.06 mu g/100 g, 171.61 +/- 6.05 mM ascorbic acid/g, 90.47 +/- 4.11 mg GA/g and 37.39 +/- 2.85 mg QE/g. DPPH and hydroxyl radical scavenging activity was obtained IC 50 67.34 +/- 4.14 and 64.87 +/- 4.68 mu g/mL, respectively. Discussion and conclusion: The results of the study indicated that FASB flowers contain high levels of vitamins, minerals, total antioxidant activity, phenolics and flavonoids. Due to the positive effect on significant changes in antioxidant enzymes of liver tissue and histopathological examination, it is thought that the plant could be used as a hepatoprotective.Article Antioxidant Properties of Rosa Pisiformis and Its Protective Effect Against Isoproterenol-Induced Oxidative Stress in Rats(Walter de Gruyter Gmbh, 2016) Bayramoglu, Mahire; Ekin, Suat; Kiziltas, Hatice; Oto, Gokhan; Susen, Ebru Altindal; Ozgokce, FevziObjective: In this study, Rosa pisiformis was evaluated for its antioxidant activity, vitamin (A, E, C), trace element (Fe, Cu, Zn, Mn, Cr, Se) and mineral (Ca, K, Mg, Na) levels, in addition to its cardioprotective effects on cardiac tissue antioxidant enzymes (GSH-Px, SOD, CAT) during isoproterenol (ISO)-induced oxidative stress. Methods: In the study, vitamin and mineral analysis was carried out using HPLC and ICP-MS method, R. P. antioxidant and antiradical properties were determined using spectrophotometer. Forty wistar albino rats were divided into four groups: control (0.9% NaCl), ISO (100 mg/kg), ISO (100 mg/kg) + R. P. (300 mg/kg) and R. P. (300 mg/kg). ISO was injected subcutaneously into rats twice at an interval of 24 h for two consecutive days (on 28(th) and 29(th) day). R. P. fruit administered daily. The experimental period was continued for 30 days. Results: The results of the study indicated that R. P. fruit and leaves contain high levels of minerals and vitamins and possess antioxidant and antiradical activity and its fruits have positive effects on cardiac tissues. The results are supported with significant changes in antioxidant enzyme levels and histopathological examination. Conclusion: This study demonstrates that R. P. fruits in particular can be used to treat and prevent cardiac diseases.Article Antioxidant, Antidiabetic, Anticholinergic, and Antiglaucoma Effects of Magnofluorine(Mdpi, 2022) Durmaz, Lokman; Kiziltas, Hatice; Guven, Leyla; Karagecili, Hasan; Alwasel, Saleh; Gulcin, IlhamiMagnofluorine, a secondary metabolite commonly found in various plants, has pharmacological potential; however, its antioxidant and enzyme inhibition effects have not been investigated. We investigated the antioxidant potential of Magnofluorine using bioanalytical assays with 2,2-azinobis (3-ethylbenzothiazoline-6-sulfonic acid) (ABTS(center dot+)), N,N-dimethyl-p-phenylenediamine dihydrochloride (DMPD center dot+), and 1,1-diphenyl-2-picrylhydrazyl (DPPH center dot) scavenging abilities and K-3[Fe(CN)(6)] and Cu2+ reduction abilities. Further, we compared the effects of Magnofluorine and butylated hydroxytoluene (BHT), butylated hydroxyanisole (BHA), alpha-Tocopherol, and Trolox as positive antioxidant controls. According to the analysis results, Magnofluorine removed 1,1-diphenyl-2-picrylhydrazyl (DPPH) radicals with an IC50 value of 10.58 mu g/mL. The IC50 values of BHA, BHT, Trolox, and alpha-Tocopherol were 10.10 mu g/mL, 25.95 mu g/mL, 7.059 mu g/mL, and 11.31 mu g/mL, respectively. Our results indicated that the DPPH center dot scavenging effect of Magnofluorine was similar to that of BHA, close to that of Trolox, and better than that of BHT and alpha-tocopherol. The inhibition effect of Magnofluorine was examined against enzymes, such as acetylcholinesterase (AChE), alpha-glycosidase, butyrylcholinesterase (BChE), and human carbonic anhydrase II (hCA II), which are linked to global disorders, such as diabetes, Alzheimer's disease (AD), and glaucoma. Magnofluorine inhibited these metabolic enzymes with Ki values of 10.251.94, 5.991.79, 25.411.10, and 30.563.36 nM, respectively. Thus, Magnofluorine, which has been proven to be an antioxidant, antidiabetic, and anticholinergic in our study, can treat glaucoma. In addition, molecular docking was performed to understand the interactions between Magnofluorine and target enzymes BChE (D: 6T9P), hCA II (A:3HS4), AChE (B:4EY7), and alpha-glycosidase (C:5NN8). The results suggest that Magnofluorine may be an important compound in the transition from natural sources to industrial applications, especially new drugs.Article Chemical Composition and Antioxidant Activity of Two Wild Edible Mushrooms From Turkey(Begell House inc, 2015) Ekin, Suat; Uzun, Yusuf; Demirel, Kenan; Bayramoglu, Mahire; Kiziltas, HaticeThe purpose of this study was to determine the antioxidant and antiradical activities in methanol extracts of two edible mushrooms from Turkey, Agaricus urinascens (Jul. Schaff. & F. H. Moller) Singer and Volvopluteus gloiocephala (DC.) Vizzini, Contu & Justo, as well as to examine trace elements (Cu, Zn, Fe, Mn, Co, Cr, Cd, Pb), minerals (Mg, Ca, K, Na) and vitamin C content. The levels of trace elements and minerals were determined by inductively coupled plasma atomic emission spectroscopy. Vitamin C and antioxidant properties were measured with a spectrophotometer. According to the results, total antioxidant activity and phenolic and flavonoid contents of A. urinascens and V. gloiocephala were 67.53 +/- 2.54 mM ascorbic acid/g and 31.68 +/- 2.27 mM ascorbic acid/g, 6.88 +/- 0.31 mg GA/g and 5.22 +/- 0.53 mg GA/g, and 3.80 +/- 0.04 mg QE/g and 2.60 +/- 0.20 mg QE/g, respectively. The findings of this study indicated that methanol extract of A. urinascens has a significant antioxidant activity and contents of total phenolic and flavonoid compounds. However, V. gloiocephala contains high levels of Ca, Cr, Fe, K, Mg, Mn, and Na, whereas A. urinascens contains higher levels of Cd, Co, Cu, Pb, and Zn than that of V. gloiocephala. Some trace elements that are in the compound of macrofungi may contribute the antioxidant capacity of macrofungi. A. urinascens has the ability to accumulate some heavy metals that prevent environmental metal pollution and can be used as a biomonitor. Boiss. subsp. acerosum's (A. acerosum). The antioxidant activity of A. acerosum was assessed in a variety of bioanalytical methods. Strong antioxidant activity was shown for both water (WEAA) and ethanol (EEAA) extracts of the aerial parts of A. Acerosum. Also, it was shown that hyperoside is the most prevalent phenolic compound in EEAA (12,940.83 mg kg(-1)) and WEAA (5409.67 mg kg(-1)) by LCHRMS. The enzyme inhibitory abilities of the EEAA were then realised. The IC50 values of EEAA against acetylcholinesterase (AChE) (E.C.3.1.1.7), alpha-glycosidase (E.C.3.2.1.20), and alpha-amylase (E.C.3.2.1.1) were determined as 1.828 mu g mL(-1), 0.615 mu g mL(-1), and 1.081 mu g mL(-1), respectively. Subsequently, the alpha-amylase, alpha-glycosidase, and AChE enzymes were subjected to molecular docking and molecular dynamic interactions using major compounds of EEAA as ligands. High enzyme-inhibiting properties of EEAA were also determined in silico. It has been reported that numerous traditional and contemporary medical applications of A. acerosum. It can be said that the effective antioxidant results obtained in our study are the basis of previously performed traditional or contemporary medical practices.Article Comprehensive Evaluation of Reseda Lutea L. (Wild Mignonette) and 7 Isolated Flavonol Glycosides: Determination of Antioxidant Activity, Anti-Alzheimer, Antidiabetic and Cytotoxic Effects With in Vitro and in Silico Methods(Tubitak Scientific & Technological Research Council Turkey, 2022) Kiziltas, HaticeIn this study, anticholinergic, antidiabetic, antioxidant and cytotoxic activities of Reseda lutea L. (R. lutea) were determined. Ethanol extracts of R. lutea (EERL) and water extract of R. lutea (WERL) were prepared for biochemical analysis. The antioxidant capacities of EERL and WERE, were evaluated with 6 different methods. In addition, acetykholinesterase (AChE), alpha-amylase and alpha-glycosidase enzyme inhibition by EERL were measured. According to the results, EERL exhibited high inhibition effects against alpha-amylase, alpha-glycosidase and AChE enzymes. The IC50 values of EERL against AChE (2.21 mu g/mL), alpha-glycosidase (1.38 mu g/mL), and alpha-amylase (0.11 mu g/mL) were determined. Also, high cytotoxic effect of EERL was observed on human lung cancer cell lines (A549) with an IC50 value of 3.58 +/- 1.10 mu g/mL. The affinities of 7 kaempferol and isorhamnetin rhamnopyranoside molecules, previously isolated from R. lutea, for AChE, alpha-amylase and, alpha-glycosidase were determined by molecular docking studies. Molecular docking results supported the in vitro results of the study. The results showed that the aerial parts of R. lutea have effective antioxidant, anticholinergic, antidiabetic and cytotoxic activities. Extraction of various Acantholimon species are traditionally applied to treat liver disease and diabetes and roots are boiled used to treat wounds and skin irritation. Here, we quantified, the radical scavenging activities of ethanol (EEAC) and water (WEAC) extracts of Acantholimon caryophyllaceum Boiss (A. caryophyllaceum) against 1,1-diphenyl-2-picryl-hydrazyl (DPPHGreek ano teleia) and 2,2-azino-bis3-ethylbenzthiazoline-6-sulfonic acid (ABTS*+) as well as their abil-ity to reduce Fe3+, Fe3+-TPTZ, and Cu2+. The antioxidant activities of both extracts were similar to those of standards in all assays. For instance, IC50 of EEAC and WEAC for scavenging DPPH were 19.8 and 69.3 mg/mL, respectively. We also assessed the inhibitory effects of EEAC and WEAC against acetylcholinesterase (AChE), a-amylase and a-glycosidase, which are associated with Alzheimer's disease and diabetes. The ethanol extract inhibited these enzymes with IC50 of 1.137, 0.172 and 0.511 mg/mL, respectively. The results of liquid chromatography-high resolution mass spectrometry (LC-HRMS) revealed that (-)-epigallocatechin was the main phenolic compound in both EEAC and WEAC. It is rich in phenolic and flavonoid compounds, which contain antioxidant, antimicrobial, and anticancer properties. In this study, the chemical composition and antioxidant activities of propolis were investigated; ABTS(center dot+), DPPH center dot and DMPD center dot+ were prepared using radical scavenging antioxidant methods. The phenolic and flavonoid contents of propolis were 53 mg of gallic acid equivalent (GAE)/g and 170.164 mg of quercetin equivalent (QE)/g, respectively. The ferric ion (Fe3+) reduction, CUPRAC and FRAP reduction capacities were also studied. The antioxidant and reducing capacities of propolis were compared with those of butylated hydroxyanisole (BHA), butylated hydroxytoluene (BHT), alpha-tocopherol and Trolox reference standards. The half maximal inhibition concentration (IC50) values of propolis for ABTS(center dot+), DPPH center dot and DMPD center dot+ scavenging activities were found to be 8.15, 20.55 and 86.64 mu g/mL, respectively. Propolis extract demonstrated IC50 values of 3.7, 3.4 and 19.6 mu g/mL against alpha-glycosidase, acetylcholinesterase (AChE) and carbonic anhydrase II (hCA II) enzyme, respectively. These enzymes' inhibition was associated with diabetes, Alzheimer's disease (AD) and glaucoma. The reducing power, antioxidant activity and enzyme inhibition capacity of propolis extract were comparable to those demonstrated by the standards. Twenty-eight phenolic compounds, including acacetin, caffeic acid, p-coumaric acid, naringenin, chrysin, quinic acid, quercetin, and ferulic acid, were determined by LC-MS/MS to be major organic compounds in propolis. The polyphenolic antioxidant-rich content of the ethanol extract of propolis appears to be a natural product that can be used in the treatment of diabetes, AD, glaucoma, epilepsy, and cancerous diseases.Article Determination of Lc-Hrms Profiling, Antioxidant Activity, Cytotoxic Effect and Enzyme Inhibitory Properties of Satureja Avromanica Using in Vitro and in Silico Methods(Elsevier Sci Ltd, 2022) Kiziltas, HaticeIn this study, some biochemical properties of Satureja avromanica such as phenolic content, anticholinergic, antidiabetic and antioxidant activities were determined. Ethanol extract of Satureja avromanica (EESA) and water extract of Satureja avromanica (WESA) were prepared and used for all biochemical analyses. The antioxidant capacities of EESA and WESA were evaluated by six different antioxidant methods. In addition, acetylcholinesterase (AChE), alpha-amylase and alpha-glycosidase enzyme inhibition of EESA were measured. EESA exhibited high inhibition effects against alpha-amylase, alpha-glycosidase and AChE enzymes. The IC50 values of EESA against AChE (1.909 mu g/mL), alpha-glycosidase (0.701 mu g/mL), and alpha-amylase (0.517 mu g/mL) were determined. The phenolic composition of Satureja avromanica was evaluated by LC-HRMS. Rosmarinic acid (19,961.31 mg/kg), hederagenin (18,895.43 mg/kg) and hesperidin (11,409.60 mg/kg) were identified as major compounds in EESA. Rosmarinic acid (9209.82 mg/kg), hesperidin (2384.73 mg/kg) and hispidulin (1530.03 mg/kg) were identified as major compounds in WESA. Molecular docking analysis showed possible roles of hesperidin, hederagenin, and rutin in AChE, alpha-amylase and alpha-glycosidase enzymes inhibition. Quinic acid, fumaric acid, gallic acid, chlorogenic acid, vanillic acid, quercetin, apigenin, and isorhamnetin were determined by LC-MS/MS in all of the species. Metabolic enzymes are essential catalysts regulating biochemical reactions within living organisms, facilitating energy production, detoxification, and biosynthesis. These enzymes play a crucial role in maintaining cellular homeostasis and are tightly regulated to ensure optimal metabolic function. High docking scores were also obtained for butyrylcholinesterase (BChE), alpha-glycosidase, alpha-amylase, and human carbonic anhydrase I and II enzymes (hCA I and hCA II). Among the extracts, Anthemis pseudocotula was concluded to be the best inhibitor for the enzymes, which was further determined by in vitro enzyme inhibition tests. Besides, it was concluded that all extracts showed anti-cholinergic, anti-diabetic, and anti-glaucoma properties. Also, it is used to treat diabetes mellitus, leukemia, nephritis and uterine cancer. It is known that in Anatolia, Astragalus roots are traditionally used against leukemia and wound healing. For the purpose, the measuring of antioxidant activity of evaporated ethanol extract of Asfragalus brachycalyx FISCHER (EEAB), some bioanalytic methods including DPPH center dot and ABTS(center dot+) scavenging effects, ferric ions (Fe3+) and cupric ions (Cu2+) reducing abilities, and metal (Fe') chelating activity were realized. alpha-Tocopherol, ascorbic acid, and BHT were used as the standard antioxidants. On the other hand, some phenolic compounds, which responsible for antioxidant activities of EEAB was determined by liquid chromatography-high resolution mass spectrometry (LC-HRMS). At the similar concentration, EEAB exhibited efficient antioxidant effects when compared to standard compounds. In the study, Wistar albino rats were assigned to six groups: Control (0.9% NaCl), (DMN 10 mg/kg), (FA 150 mg/kg), (DMN + FA 150 mg/kg), (FA 300 mg/kg), and (DMN + FA 300 mg/kg). Rats were intraperitoneally given DMN for the first 7 days. Renal injury caused by DMN was proved by the histopathological alterations. The FA (300 mg/kg) treatment significantly normalized Se, Cr, Ca levels in liver and Co level in kidney tissue samples. These observed positive effects are due to the phytochemical content of the plant. The flower extract of FA (300 mg/kg) can be used for the prevention of kidney damage. Practical applications Ferulago angulata flowers are used in traditional medicine for treat kidney and liver digestive system diseases. This species is endemic taxa of the family Apiaceae, which has been used both as food and therapeutics because of their phytochemical composition. In this study, the phenolic characterization of FA flower was used to a new RP-HPLC method, as well as the biological activity of FA flower and possible in vivo nephroprotective effect of FA flowers on trace elements, minerals, MDA and GSH in kidney and liver tissue samples and vitamins, TSA, and LSA in serum samples a rat model of DMN-induced nephrotoxicity. In the study, 40 Wistar albino rats were divided into four groups of ten each: control, isoproterenol 100 mg/kg bodyweight; isoproterenol 100 mg/kg bodyweight; then R. pisiformis 300 mg/kg bodyweight, and R. pisiformis 300 mg/kg bodyweight. Rats were given isoproterenol twice at an interval of 24 h for two days (on days 28 and 29) subcutaneously. The experimental period was maintained at 30 days. According to analysis results, caffeic acid and p-coumaric acid were found to be the high contents of the fruit extracts at 6.01 +/- 0.0006 and 3.93 +/- 0.007 mg/100 g dry weight. It showed that R. pisiformis (300 mg/kg bodyweight) aqueous extract had a potent action on oxidative damage. The R. pisiformis (300 mg/kg bodyweight) treatment significantly alleviated toward normalcy on the zinc, manganese, cobalt, magnesium, and sodium values in the heart, and zinc and magnesium values in liver tissue samples. Several antioxidant techniques were used in this investigation to assess the antioxidant capabilities of spiraeoside, including 1,1diphenyl-2-picrylhydrazyl radical (DPPHGreek ano teleia) scavenging, N,N-dimethyl-p-phenylenediamine radical (DMPD'') scavenging, 2,2'-azinobis-(3-ethylbenzothiazoline-6-sulphonate) radical (ABTS'') scavenging activities, cupric ions (Cu2') reducing and potassium ferric cyanide reduction abilities. In contrast, the water-soluble a-tocopherol analogue trolox and the conventional antioxidants butylated hydroxytoluene (BHT), butylated hydroxyanisole (BHA), and a-tocopherol were utilized as the standards for evaluation. Spiraeoside scavenged the DPPH radicals an IC50 of 28.51 mu g/mL (r2: 0.9705) meanwhile BHA, BHT, trolox, and a-tocopherol displayed IC50 of 10.10 mu g/mL (r2: 0.9015), 25.95 mu g/mL (r2: 0.9221), 7.059 mu g/mL (r2: 0.9614) and 11.31 mu g/mL (r2: 0.9642), accordingly. The results exhibited that spiraeoside had effects similar to BHT, but less potent than a-tocopherol, trolox and BHA. Also, inhibitory effects of spiraeoside were evaluated toward some metabolic enzymes including acetylcholinesterase (AChE), butyrylcholinesterase (BChE), carbonic anhydrase II (CA II) and a-glycosidase, which are related to a number of illnesses, such as Alzheimer's disease (AD), diabetes mellitus and glaucoma disorder. Spiraeoside exhibited IC50 values of 4.44 nM (r2: 0.9610), 7.88 nM (r2: 0.9784), 19.42 nM (r2: 0.9673) and 29.17 mM (r2: 0.9209), respectively against these enzymes. Enzyme inhibition abilities were compared to clinical used inhibitors including acetazolamide (for CA II), tacrine (for AChE and BChE) and acarbose (for a-glycosidase). Spiraeoside demonstrated effective antioxidant, anticholinergic, antidiabetic and antiglaucoma activities.
