Article Comparison of Magenstrasse and Mill Gastroplasty and Sleeve Gastrectomy Techniques as an Experimental Study on Rabbits(Edizioni Luigi Pozzi, 2020) Sumer, Aziz; Celik, Sebahattin; Aktokmakyan, Talar Vartanoglu; Peksen, Caghan; Sancak, Tunahan; Kuscu, Yagmur; Eren, EryigitAIM: Bariatric surgery is an important option when lift-style modification, diet, and medical treatment are inadequate in lose weight. Bariatric surgical methods have gained popularity in recent years. In this paper, we compared the Magenstrasse and Mill(M&M) technique, with performing a simpler and more physiological type of gastroplasty without implanted foreign material such as band and reservoir, to the Sleeve Gastrectomy (SG) technique. This study aimed to determine the effects of the M&M for obesity on the rabbits in comparison with the SG, which is accepted as a standard bariatric technique with creating a gastric tube. MATERIAL AND METHODS: The study was approved by the University of Van Yuzuncu Yil Regional Committee of Ethics (Institutional Animal Care and Use Committee). 20New Zealand Rabbits underwent operations. After prestudy with 2 rabbits, the remaining 18 rabbits were divided into 2 groups; Group 1 (SG) and Group 2 (M&M). RESULTS: Group 1 rabbits were observed to lose weight in all, while Group 2 rabbits; 2 of them died 5 of them lost weight, 2 of them gained weight. When the pre and post-operative weight of the rabbits were compared; preoperative median weight values of 9 rabbits in Group 1 were significantly higher than postoperative values. On the other hand, there was no significant change in the mean weight of Group 2 of 7 rabbits (living up to 8weeks). The mean weight of rabbits undergoing standard SG was significantly lower than the MeM technique. CONCLUSION: We believe that this animal experimental study, which we conducted intending to compare MeM and SG techniques, will contribute to the literature as a pilot study and determine the survey of M&M technique as a pioneer in other studies.Article Congenital Extraneural Hemangioblastoma in a Lamb(Sage Publications inc, 2019) Yaman, Turan; Karasu, Abdullah; Uyar, Ahmet; Kuscu, Yagmur; Keles, Omer FarukA 1-mo-old Ivesi male lamb was presented with 2 large red masses on the skin of the left ear. The tumors were removed using gentle dissection and submitted for histologic evaluation. The tumors consisted of numerous thin-walled capillaries lined by endothelial cells and nests of stromal cells. Immunohistochemically, the endothelial cells were positive for CD45, and the stromal cells were positive for neuron-specific enolase. GFAP-positive cells were occasionally present within the tumor. Endothelial and stromal cells were negative for S100, CD34, CD31, and factor VIII-related antigen. The tumor had strong gross, microscopic, and immunohistochemical similarities with human extraneural hemangioblastoma.Article Effect of Low- and High-Dose Methotrexate on Wound Healing in Rats(Acta Cirurgica Brasileira, 2025) Karasu, Abdullah; Kuscu, Yagmur; Kayikci, Caner; Yildirim, Serkan; Kuscu, Oguzhan; Kiliclioglu, MetinPurpose: To investigate the effect of intraperitoneal treatment with low- and high-dose methotrexate (MTX) on wound healing in rats. Methods: The study sample consisted of 54 healthy rats. Under aseptic conditions, skin wounds were created with two circular fullthickness punch tools, 10 mm in diameter, one on the right and the other one on the left of the dorsal vertebral line. The rats were randomly assigned to one of three main treatment groups. On the 0th day (2 hours before wound creation), 7th day, and 14th day, the control group received 0.3-mL saline, the low-MTX group received 3 mg/kg MTX, and the high-MTX group received 30 mg/kg MTX, all administered intraperitoneally. The wounds were evaluated seven, 14, and 21 days after injury through morphometrical, biochemical, histopathological, and immunohistochemical analyses. Results: MTX dose-dependently decreased the degree of inflammation and angiogenesis, tissue hydroxyproline level, and HSP70 and tumor necrosis factor-alpha expression in the early phase of wound healing. It also suppressed epithelialization and collagen 1 expression throughout the wound-healing process. Conclusion: The wounds treated with high-dose of MTX had statistically delayed wound closure on days 7, 14 and 21 compared to the saline group, while wounds treated with low-dose of MTX only had statistically delayed wound closure on day 14. In addition, weight loss was observed in rats treated with high-dose MTX, which was thought to reflect its toxicity. The dose-dependent adverse effect of MTX on wound healing may be due to its antiproliferative, antifibrotic, anti-inflammatory, and antiangiogenic effects.Article Evaluation of Prolidase Activity, Oxidative Stress, and Antioxidant Enzyme Levels in Testicular and Penile Tissues After Human Chorionic Gonadotropin Treatment in Rats by Predicting Infertility and Erectile Dysfunction(Karger, 2018) Pirincci, Necip; Yildirim, Serkan; Tas, Abuzer; Kuscu, Yagmur; Ozan, Tunc; Firdolas, Fatih; Karakeci, AhmetObjectives: Prolidase plays a vital role in collagen turnover, matrix remodeling, and cell growth. We aimed to evaluate the association between treatment with chorionic gonadotropin and infertility and erectile dysfunction by investigating tissue prolidase activity, oxidative stress, and levels of antioxidant enzymes. Materials and Methods: The 16 male Wistar albino rats used in this study were randomly divided into 2 groups: rats treated with human chorionic gonadotropin (hCG) and control rats (n = 8 in each group). The rats in the hCG group were subcutaneously injected with 50 IU hCG daily for 15 days, while the rats in the control group were subcutaneously injected isotonic saline. All of the rats were sacrificed by a lethal overdose of sodium pentobarbital at the first month after hCG administration. Prolidase activity and levels of malonyl aldehyde, glutathione reductase, superoxide dismutase (SOD), glutathione peroxidase (GSH-Px), and catalase (CAT) were estimated in the testicular and penile tissue. The testicles and penis were transversely dissected and placed in formalin. Results: Levels of prolidase and malonyl aldehyde in the testicular and penile tissues were significantly higher in the hCG group than in the control group (p < 0.001), while levels of glutathione reductase, SOD, GSH-Px, and CAT were significantly lower in the hCG group than in the control group (p < 0.001). Conclusions: In this study, we observed that treatment with hCG increased prolidase activity and oxidative stress and decreased the antioxidant capacity of penile and testicular tissues; therefore, this may affect fertility and erectile function. Karger AG, BaselArticle Investigation of the Effect of Silymarin on Oxidative Dna Damage and Inflammatory Markers in Ischemia/Reperfusion Injury Following Experimental Testicular Torsion/Detorsion in Rats(Tubitak Scientific & Technological Research Council Turkey, 2021) Belhan, Saadet; Yildirim, Serkan; Kayikci, Caner; Komuroglu, Ahmet Ufuk; Ozdek, Ugur; Kuscu, YagmurThe aim of the present study was to clarify the effects of silymarin on experimental testicular ischemia / reperfusion injury. A total of 40 Wistar albino rats (10-12 weeks of age, weighing 280-300 g) were randomly divided into five groups. Control group: No surgical procedures were performed. Torsion 3 h / detorsion 3 h group; torsion 3 h / detorsion 24 h group; torsion 3 h / detorsion 3 h + silymarin (250 mg/kg) group; and torsion 3 h / detorsion 24 h + silymarin (250 mg/kg) group. In the study, 720 degrees torsion was applied to the left testicle. At the end of the study, blood was collected from the rats, and an orchiectomy was performed on the left testicles. It was found that tumor necrosis factor alpha (Tnf-alpha) and 8-hydroxy-2-deoxyguanosine (8-OHdG) expression levels in testicular tissue increased significantly in torsion/detorsion groups, and the expression levels decreased significantly with silymarin administration. In addition, in the testicular tissue of the torsion/detorsion groups, glutathione (GSH) levels, superoxide dismutase (SOD), and glutathione peroxidase (GPx) activities decreased, while malondialdehyde (MDA) levels increased. It was found that the parameters specified were reversed with the administration of silymarin. Based on our findings, we can say that silymarin reduces testicular injury by activating antioxidant mechanisms in ischemia/reperfusion injury and minimizing the inflammatory response.Article A New Approach in Bariatric Operations: Bridged Mini Gastric By-Pass. The increasing demand for bariatric surgery has led to the emergence of new techniques. We modified previously described Mini Gastric By-pass(MGB) technique via leaving a bridge at the most cranial 2 cm of the fundus of the human stomach to the follow-up and treatment of the remnant stomach and duodenum. We would like to entitle this new technique as Bridged MGB and aimed to apply on rabbits as an experimental study. Material and Methods: The study was performed in the experimental animal laboratory of university after ethical approval was taken from the local ethics committee. Described new technique was applied to 2.1 and 3.2 kg 2 New Zealand rabbits. Results: As a result of the operations, one of the rabbits died on the day of the operation; the other rabbit was exitus postoperatively on the third day. In autopsies, although no problem was detected at the anastomoses, necrosis was detected in the large curvature of both rabbits. Conclusion: Rabbit, one of the popular experimental animals, has been shown to be different from the human gastrointestinal system in both arterial and topographic aspects and it has been emphasized that it varies according to the species and even the diet and the climate. We believe that our study failed as a result of these differences and that animals more similar to humans should be used in gastrointestinal experimental studies.Article Serum Retinol, Alpha-Tocopherol, Cholecalciferol, and Some Mineral Levels in Ruminants With Congenital Digestive and Urogenital System Anomalies1(Revista Pesquisa veterinaria Brasileira, 2024) Karasu, Abdullah; Genccelep, Musa; Kayikci, Caner; Kuscu, YagmurThis study aimed to investigate the relationship between serum vitamin and mineral levels and congenital defects in digestive and urogenital system anomalies in calves, lambs, and kids. The study material consisted of 13 calves, 15 lambs, 10 kids clinically and radiologically diagnosed with congenital digestive and urogenital system anomalies and 10 newborn clinically healthy calves, 10 lambs, and 10 kids. Congenital defects were diagnosed by clinical and radiological examination. Blood samples were collected from all animals, and sera were extracted for biochemical analysis. Vitamins A, D, and E, calcium, phosphorus, sodium, potassium, chlorine, magnesium, copper, iron, zinc, selenium, and manganese levels were measured in serum samples. Penile urethral diverticulum in kids, atresia ani, atresia ani with vaginal fistula in lambs, and atresia ani and atresia coli defects in calves were determined. Copper levels were higher, and zinc levels were lower in kids with penile urethral diverticulum compared to the control group. Vitamin A levels were lower in lambs with digestive system anomalies compared to the control group. Meanwhile, copper levels were higher in lambs with digestive system anomalies. Vitamin A and D levels were lower in calves with digestive system anomalies compared to the control group. There was no difference in the levels of the other parameters compared to the control group. In conclusion, insufficient serum vitamin A levels may play a role in the etiopathogenesis of congenital intestinal atresia in calves and lambs. Therefore, we believe that parenteral vitamin A administration to the mother, especially in the last trimester of pregnancy in regions with continental climates and poor green vegetation, would be beneficial. Further research should be conducted to determine the role of vitamin A in the etiopathogenesis of congenital atresia ani and coli.
