Article Effect of Intermittent Fasting on Sciatic Neurofilaments in Acrylamide Administered Rats(Yuzuncu Yil Universitesi Tip Fakultesi, 2018) Oğuz, E.K.Acrylamide is a widely used chemical in industrial manufacturing and laboratory studies. Long term use of this chemical causes axonal degeneration. In this study, 20 mg/kg of acrylamide was administered to Wistar-albino rats daily and effects on sciatic nerve and any potential protective activity of intermittent fasting were investigated. Effect of acrylamide and intermittent fasting was assessed by immunefluorescent method. As a result paralysis in hind legs and coordination problems during walking was observed in acrylamide administered group. These problems were less frequent in intermittent fasting group. In addition, a decrease in neurofilament number was observed in acrylamide group whereas neurofilament level was similar with control group in intermittent fasting group. Concomitant administration of acrylamide and intermittent fasting caused ameliorating effect of intermittent fasting. A potential protective effect of intermittent fasting against acrylamide can be considered according to such results. © 2018, Yuzuncu Yil Universitesi Tip Fakultesi. All rights reserved.Article Effects of Sublethal Concentrations of Cypermethrin on the Gills of Lake Van Fish (Alburnus Tarichi)(Taylor and Francis Ltd., 2022) Oğuz, E.K.Cypermethrin is one of the most commonly used pesticides in the Lake Van Basin of Turkey. In this study, the effects of a sublethal dose (0.05 µg/L) of cypermethrin for 96 h on the gills of Lake Van fish (Alburnus tarichi), the only vertebrate species living in Lake Van, were investigated. Levels of heat shock protein 70 (HSP70), Na,K-ATPase, and DNA damage (8-OHdG) were determined immunohistochemically in the gills. In addition, changes in gill mucous cells and histopathological changes in gill tissues after 96 h were investigated. At the end of the cypermethrin administration, an increase was observed in the numbers of mitochondria-rich cells, HSP70-positive cells, and 8-OHdG-positive cells in the gills. A decrease in both the number and volume of mucous cells was detected. The expansion of blood vessels, aneurysms, lamellar lifting, hypertrophy, lamellar fusion, and necrosis were observed in gill tissues. It was concluded that the 96-h sublethal application of cypermethrin caused stress, deterioration of osmoregulation, DNA damage, and histopathological damage in the gills of Lake Van fish. © 2022 Informa UK Limited, trading as Taylor & Francis Group.Article Histopathology of the Gills of Lake Van Fish Alburnus Tarichi (Güldenstädt, 1814) Infected With Dactylogyrus Spp. Parasites(Western Philippines University, 2023) Erdemir, A.N.; Alkan, Z.; Azizoğlu, B.E.; Sepil, A.; Oğuz, E.K.; Oğuz, A.R.The Lake Van fish is an anadromous carp endemic to the Lake Van basin. In this study, the histopathological effects of Dactylogyrus spp. parasites, which naturally infected fish, on the gills were determined during reproductive migration. Fish gills were stained with hematoxylin and eosin after routine histological procedures and apoptotic cells in the gills were determined immunohistochemically. Dactylogyrus spp. parasites were found in 8 of the 60 fish caught in the study. No parasites were found in the fish sampled from the lake. It was observed that the parasites caused hypertrophy, hyperplasia, edema, epithelial desquamation, hemorrhage, fusion of secondary lamellae, and necrosis in the gills. Immunohistochemically, no increase in the number of apoptotic cells was observed in the gills of the parasite-infected fish when compared with the non-infected fish gills. As a result, it can be inferred that the lake water has a restrictive effect in fish against parasites. Histopathologic lesions caused by the observed parasites in fish caught in fresh water could significantly affect gill functions. © Western Philippines University.Article Investigation of Changes in Goblet Cells in the Digestive Tract of Van Fish During Reproductive Migration(University of Zagreb, Facultty of Veterinary Medicine, 2025) Azizoğlu, B.E.; Alkan, Z.; Erdemir, A.N.; Örgi, E.; Oğuz, E.K.; Oğuz, A.R.Lake Van is among the largest soda lakes in the world. The Van fish is an endemic species of the carp family living in Lake Van. In order to breed, they migrate in flocks to the streams flowing into the lake between April and July, depending on the water temperature. After the fish lay their eggs in the fresh water, they return to Lake Van. In this study, goblet cells in the digestive tract of Van fish during reproductive migration were labeled with periodic acid–Schiff, Alcian Blue pH 2.5, Alcian Blue pH 1.0, Alcian Blue pH 0.5, and aldehyde fuchsin dyes. Changes in cell localization, density, and area were determined in different parts of the digestive tract in goblet cells containing neutral, acidic, O-sulfate ester, strongly sulfated, and sulfated acidic glycoconjugate. In the esophagus, goblet cells were only observed in the fish from Lake Van. Although goblet cell size and density varied in other parts of the digestive tract, all glycoconjugates were labeled. In the digestive tract of fish from the lake, the goblet cell areas were large, but the cell density was low, whereas the situation in the digestive tract of fish sampled from the streams was the opposite. It was concluded that the main reason for the goblet cell change in the digestive tract of Van fish is related to its feeding behavior and habitat. © 2025, University of Zagreb, Facultty of Veterinary Medicine.
