Article Activity, Acute and Sub-Acute Toxicity and Safety Assesment of the Hydroalcholic Root Extract of Diplotaenia Turcica(Agricultural Research Communication Centre, 2018) Ozdek, U.; Basbugan, Y.; Yildirim, S.; Boga, M.; Firat, M.; Deger, Y.Diplotaenia turcica is widely utilized in conventional treatment in the east of Turkey. Due to the insufficient data on the safety profile, the acute and sub acute toxicity of Diplotaenia turcica was determined. Furthermore, total phenolic content, flavonoid content and antioxidant activity of the extract were determined. The acute toxicity of hydroalcoholic root extract of Diplotaenia turcica (HREDT) was evaluated in mice after administration of single oral dose at the 5000 mg/kg (acute model) and rat after 28 days orally administration at the dose range of 250, 500 and 1000 mg/kg (subacute model). According to result, the LD50 value of HREDT was found to be greater than 5000 ing/kg. In sub acute toxicity study, no statistically significant differences were observed in the values of hematological and pathological parameters in comparison with control group. It was determined that the glucose, triglyceride, cholesterol and LDL levels exerted a significant effect depending on the HREDT doses. The results from the present study found out that HREDT did not produce any toxic effects or deaths in animals for both single and chronic administration. Additionally, HREDT showed moderate antioxidant activities and exhibited relatively notable total phenolic content.Article The Impact of Chitosan on Oxidative Stress in Liver of Rats Loaded With Lead Acetate(Hellenic veterinary Medical Soc, 2022) Ozdek, U.; Toz, H.; Komuroglu, A. U.; Mis, L.; Deger, Y.This study was conducted to determine the impact of chitosan on lead (Pb)-induced hepatotoxicity. Thus, lead acetate was administered intraperitoneally (50 mg/kg for 5 days) and chitosan was given as 200 mg/kg via oral gavage for 28 days. When the trial was terminated, it was determined that aspartate aminotransferase (AST), alanine aminotransferase (ALT) and alkaline phosphatase (ALP) activities in serum as well as Pb, 8-hydroxy-deoxyguanosine (8-OHdG) and malondialdehyde (MDA) levels and catalase (CAT) activity in liver tissue significantly increased, however reduced glutathione (GSH) and ceruloplasmin (Cp) levels in liver and high density lipoproteins (HDL) in serum had a significant decrease in the Pb group when compared to the control group. The administration of chitosan significantly prevented Pb-induced changes in serum liver enzyme activities, 8-OHdG and MDA levels. In addition, chitosan showed a statistically insignificant effect on reduced GSH, Cp, HDL levels and CAT activity. Accordingly, administration of chitosan can strengthen the antioxidant defence system of liver tissue and may decrease oxidative stress.Article Synergistic Effects of Sinapic Acid and Ellagic Acid Ameliorate Streptozotocin-Induced Diabetic Nephropathy by Inhibiting Apoptosis, Dna Damage, and Structural Deterioration in Rats(Sage Publications Ltd, 2021) Altindag, F.; Ozdek, U.Introduction Diabetic nephropathy (DN), a global problem that threatens human health, is an important reason for chronic kidney disease and kidney failure. In our study, it was aimed to investigate the individual and combined effects of SA and EA in streptozotocin (STZ)-induced rats. Methods The groups are as follows: Control, untreated diabetic, diabetic treated with Sinapic acid (SA), diabetic treated with Ellagic acid (EA), diabetic treated with SA and EA, treated with SA, treated with EA, and treated with SA and EA. Total kidney volume, total glomerulus volume, total filtration space volume, caspase-3, and 8-OHdG immunoreactivity, Malondialdehyde (MDA), Glutathione (GSH), Catalase (CAT), serum urea, and creatinine levels were evaluated by stereological, immunohistochemical, and biochemical methods. Results The findings of the study showed that total kidney volume, total glomerulus volume, total filtration gap volume, caspase-3, and 8-OHdG immunoreactivity, MDA, serum urea, and creatinine levels significantly increased in the untreated diabetic group compared to the control group. Also, severe mesangial and glomerular enlargement, extracellular matrix accumulation, and glomerular and tubular basal membrane thickness were observed in the tubulointerstitial and glomerular of the diabetic rats. However, individual and combined treatments of SA and EA ameliorated these histological changes. Additionally, decreased GSH and CAT in the untreated diabetic group increased by SA and EA treatment. Conclusions The findings suggest that treatment of SA and EA prevent apoptosis and DNA damage and structural changes in STZ-induced DN. However, the combined treatment of SA and EA were more effective than their individual treatments in all parameters except serum urea and creatinine.Article Total and Lipid-Bound Serum Sialic Acid and Hematological-Biochemical Gas Changes in Dogs With Dirofilariosis(Hellenic veterinary Medical Soc, 2022) Kesici, H.; Deger, Y.; Oguz, B.; Ozdek, U.The measurement of serum sialic acid concentration is of importance to indicate damage in cells or tissues. Dirofilaria immitis causes cardiovascular, pulmonary, hepatic and kidney lesions. Therefore, present study was planned to investigate the potential changes in the levels of serum total sialic acid (TSA), lipid-bound sialic acid (LSA) and hematological-biochemical parameters in the dogs naturally infected by Dirofilaria immitis.The patient group included the 7 dogs clinically and parasitologically (Modified Knott and PCR) diagnosed with Dirofilaria immitis while 7 healthy dogs were assigned as the control group. The biochemical parameters, blood gas parameters and hematological parameters were tested in the blood samples taken properly according to the guidelines from all the study animals using automated biochemistry analyzer, blood gas analyser and automated hematology analyzers, respectively. Serum TSA and LSA levels were measured spectrophotometrically using Sydow and Katapodis methods, respectively. Compared with the healthy group, the dogs diagnosed with dirofilariosis were found to have significantly increased levels of serum cardiactroponin, triglycerides, VLDL, LDL, BUN, urea and creatinine and enzymatic activities of CK-MB, ALT, AST, ALP and LDH, and statistically significantly decreased levels of cholesterol and HDL (p<0.05). A significantly decreased level of RBC and a significantly increased level of WBC was determined in the dogs with dirofilariosis (p<0.05). The differential leucocyte count test of the dogs with dirofilariosis indicated statistically significantly increased eosinophil count (p<0.05). In the dogs with dirofilariosis, the decreases in the levels of pCO(2), HCO3 and BE in venous blood were statistically significant (p<0.05). The levels of serum TSA and LSA of the dogs with dirofilariosis were found statistically significantly higher than the healthy group (p<0.05). As a conclusion, statistically significant differences were identified between the dogs with dirofilariosis and healthy dogs in terms of sialic acid levels and certain biochemical, venous blood gas and hematological parameters. On the other hand, the present study is the first to investigate the serum sialic acid levels in the dogs with dirofilariosis.
