Article The Role of Glutation-S Mu1 and Teta1 Polymorphisms in Chronic Obstructive Pulmonary Disease(University of Ankara, 2021) Hayat, B.; Yavuz, M.S.; Sahin, E.; Dirican, O.; Sarialtin, S.Y.; Yilmaz, C.; Oguztüzün, S.Objective: The aim of this study the investigation of "null" alleles in GSTM1 and GSTT1 gene regions in the development of COPD disease. Material and Method: There are 36 patients with COPD and 14 control cases, who received the Ethics Committee permission from Polatli Duatepe State Hospital Chest Diseases Department. DNA isolations were made from blood samples from the end of 2019 and the control group. Deletions in GSTM1 and GSTT1 gene regions were examined by qPCR method in COPD patient and control groups. The results of the study were evaluated comparatively by distributing the gene dose according to the Hardy-Weinberg. Result and Discussion: When seen from 36 COPD patients after qPCR analysis, it was found that deletion expressions due to gene doses in all patient groups; 23 individuals (63.8%) in the (+/-) genotype for GST-M1, 13 individuals (26.2%) in the (-/-) genotype. For GST-T1, 14 (%) in the (+/+) genotype 38.8), while deletion was observed with the highest rate, 4 individuals (11.1%) in the (+/-) genotype and 18 individuals (50.1%) in the (-/-) genotype were found. For GST-M1, deletion was observed in 19 individuals (63.3%) in the genotype (+/-) in male individuals, while it was observed in 4 individuals (66.6%) with the same genotype in women. While deletion was not observed in 11 (36.6%) male patients, this rate was observed as 2 (33.4%) in women. In the GST-T1 gene region, there were 10 (33.3%) males in male patients and 4 (66.6%) individuals in female patients with deletion occurring and the frequency of the "null allele" was high (+/+). In the (+/-) genotype, 3 (10%) in males and 1 (16.7%) in females were found. In the genotype where deletion was not observed and the gene was conserved (-/-), 17 (56.7%) individuals were observed in male patients and 1 (16.7%) in female patients. In the case that the gene "null" allele status in the GST-M1 gene region is slightly higher than the GST-T1 gene communication, this situation is thought to be a factor in obstructive pulmonary disease. © 2021 University of Ankara. All rights reserved.Article Synthesis of Bis(Thiosemicarbazone) Derivatives: Definition, Crystal Structure, Biological Potential and Computational Analysis(Taylor & Francis Ltd, 2018) Ates, D.; Gulcan, M.; Gumus, S.; Sekerci, M.; Ozdemir, S.; Sahin, E.; Colak, N.(2-(2-(2-methoxyphenyl)hydrazono)cyclohexane-1,3-diylidene) bis(hydrazine carbothioamide) (L-1), 4-(2-(-2,-6-bis(2-carbamothioylhydrazono) cyclohexylidene) hydrazinyl)benzoic acid (L-2), and (2-(2-(4-bromophenyl) hydrazono) cyclohexane-1,3-diylidene) bis(hydrazinecarbothioamide) (L-3) bis(thiosemicarbazone) derivative compounds were synthesized by the condensation reaction of thiosemicarbazide and various ketone compounds. The structures of synthesized compounds were characterized by using FT-IR, H-1 and C-13-NMR spectra, elemental analysis and mass spectra. Also, the structure of L-1 has been determined by X-ray crystallographic analysis. Additionally, the antioxidant activities (free radical (DPPH) scavenging, iron chelating and reducing power) of compounds were evaluated. Especially, L-3 displayed good DPPH activity (84.13%) at 100mg/L. Moreover, pBR322 plasmid DNA cleavage activity was examined and all of the compounds were able to cleave the plasmid DNA. Finally, the ground state geometries of the thiosemicarbazone derivatives were optimized using Density Functional Theory applications at B3LYP/6-31G (d,p) level in order to obtain information about the 3D geometries and electronic structure.Article Synthesis, Characterization, Antioxidant Properties and Dft Calculation of Some New Pyrimidine Derivatives(Taylor and Francis Ltd., 2019) Akbas, E.; Ergan, E.; Sahin, E.; Ekin, S.; Cakir, M.; Karakus, Y.In this study, 5-benzoyl-4-(4-(methylthio)phenyl)-6-phenyl-1,2,3,4-tetrahydro-2-thioxo (1), oxo (2) and imino (3) pyrimidine derivatives were prepared via Multicomponent Cyclocondensation Reactions (MCRs). The compounds thiazolo[3,2-a]pyrimidin-3(5H)-one (4) and thiazin-4(6H)-one (5) were obtained via the reaction of compound 1 with bromoacetic acid and 3-bromopropionic acid, respectively. Structures were determined by using FT-IR, 1H/13C NMR and elemental analyses. Also the compounds 4 and 5 were analyzed by X-ray single crystal analysis. All compounds were investigated as corrosion inhibitors using density functional theory (DFT) at the level of B3LYP/6-31G (d, p). According to the calculations, the compound 3 appears to be a good inhibitor for corrosion. On the other hand, total antioxidant properties were measured in vitro by DPPH•, ABTS•+ test, hemolysis of phenylhydrazine erythrocytes and metal chelating effect. The results were compared with standard antioxidants such as trolox and α-tocopherol. These data revealed that compounds 1, 2 and 5 are more active with respect to 3 and 4 in scavenging the radicals. © 2018, © 2018 Taylor & Francis Group, LLC.Article The Use of an Ostial Stent Does Not Increase the Success Rate of Endoscopic Dacryocystorhinostomy(Cambridge Univ Press, 2018) Dinc, M. E.; Ulusoy, S.; Sahin, E.; Bozan, N.; Avincsal, M. O.; Tutar, B.; Uyar, Y.Objective. This prospective, controlled study assessed how placing a stent into a newly formed ostium affects ostial patency, success and complication rates in endoscopic dacryocystorhinostomy patients. Methods. In group 1 (40 eyes of 36 patients), both silicone tube intubation and tube stenting were performed. In group 2 (36 eyes of 34 patients), only silicone tube intubation was performed. Success, operative time and post-surgical complications were investigated two months post-operatively in each group. Results. The success rates were 92.5 per cent and 83.3 per cent for groups 1 and 2 respectively, but the difference was not statistically significant (p = 0294). The complication rates also differed between the two groups, but this was again insignificant. Conclusion. Compared with the use of a silicone tube alone, the addition of an ostial stent did not significantly increase the success rate of endoscopic dacryocystorhinostomy.
