Article Antioxidant and Hepatoprotective Effect of Oriental Hackberry (Celtis Tournefortii L.) Against Cci4 Injury in Rat Liver(Parlar Scientific Publications (p S P), 2019) Temiz, Mehmet Ali; Temur, Atilla; Surucu, Mustafa; Yaman, TuranThis study aimed to investigate the antioxidant and hepatoprotective effects of Celtis tournefortii leaf extract (Ct) against CCl4-induced liver damage in rats. A total of 32 Wistar albino rats were divided into four equal groups (n = 8): control, CCl4 group, CCl4 + Celtis group (CCl4 + Ct), and Celtis group (Ct). Superoxide dismutase (SOD), glutathione peroxidase (GPx), and catalase (CAT) activities, glutathione (GSH) level, malondialdehyde (MDA) content, total antioxidant status (TAS), and total oxidant status (TOS) were analyzed in the liver tissues. Liver enzymes were assayed in the serum. The liver histopathology was also evaluated. The levels of alanine aminotransferase and lactate dehydrogenase significantly decreased in the CCl4 + Ct group in comparison with those in the CCl4 group. SOD and GPx activities and TAS increased significantly with CCl4 + Ct administration when compared with the administration of CCl4 alone. The oxidative stress parameters, such as MDA and TOS, showed a reduction when Ct treatment was applied to the CCl4-induced group. The results revealed that Celtis remarkably attenuated the degenerative and necrotic changes caused by CCl4 in the liver tissues. Consequently, the Ct may augment the antioxidant status and moderated the severity of CCl4-induced liver damage. The therapeutic use of Celtis tournefortii might be effective in the treatment of liver injuries.Article Antioxidant and Hepatoprotective Effects of Vitamin E and Melatonin Against Copper-Induced Toxicity in Rats(Pharmacotherapy Group, 2018) Temiz, Mehmet Ali; Temur, Atilla; Oguz, Elif KavalPurpose: To determine the preventive effects of vitamin E and melatonin against acute copper toxicity in rats. Methods: A total of 28 Wistar albino rats were randomly divided into four equal groups: control, copper sulfate, copper sulfate + vitamin E, and copper sulfate + melatonin. The following were analyzed in all groups: serum enzymes; activity of the antioxidant enzymes superoxide dismutase (SOD), catalase (CAT) and glutathione peroxidase (GPx), protein carbonyl (PC) content. Liver histopathology was also evaluated. Results: PC levels decreased in rats administered copper sulfate/vitamin E and copper sulfate/melatonin. The activities of SOD, GPx and CAT increased significantly with melatonin and vitamin E administration when compared with administration of copper sulfate alone. Histopathologic features revealed severe degeneration and necrotic changes in rats administered copper sulfate alone. These changes were observed to a lesser extent in the copper sulfate + melatonin group and only minimally in the copper sulfate + vitamin E group. Conclusion: Vitamin E and melatonin are protective against liver damage caused by copper. Vitamin E, and melatonin may strengthen the antioxidant defense system by inhibiting protein oxidation and enhancing the activity of antioxidant enzymes.Master Thesis Antioxidant and Liver Protective Effects of Carob Seed (Ceratonia Siliqua L.) on the Rats Induced Oxidative Stress by Ethyl Alcohol(2011) Temiz, Mehmet Ali; Çelik, İsmail; Temur, AtillaBu çalışmada, etil alkol ile oluşturulan oksidatif stresli sıçanlarda keçiboynuzu çekirdeği'nin (Ceratonia siliqua L.) karaciğer koruyucu ve antioksidan etkisinin belirlenmesi amaçlanmıştır.Yirmi dört Wistar albino sıçan altışar gruplar halinde dört gruba ayrıldı (kontrol, %20 alkol, %15 çekirdek ve %15 çekirdek+%20 alkol). 50 günlük muamele sonunda, keçiboynuzu çekirdeği katkılı yemin karaciğer koruyucu ve antioksidan rolü için; karaciğer harabiyeti serum enzimlerinden aspartat aminotransferaz (AST), alanin aminotransferaz (ALT), laktat dehidrogenaz (LDH), albumin (ALB) ve total protein (TP) enzim seviyeleri, beyin, böbrek, dalak, eritrosit, kalp ve karaciğer dokularında antioksidan savunma unsurlarından süperoksid dismutaz (SOD), glutatyon peroksidaz (GSH-Px), glutatyon-S-transferaz (GST), glutatyon redüktaz (GR) enzim aktiviteleri ile redükte glutatyon (GSH) seviyeleri ve malondialdehit (MDA) içerikleri spektrofotometrik yöntemle tespit edilerek değerlendirilmiştir.Bulgularda, alkol grubunun serum AST, ALT, LDH enzim seviyeleri artmış, çekirdek+alkol grubunda ALT ve LDH seviyeleri azalmıştır. Kalp dokusu haricindeki dokuların MDA değerleri alkol grubunda artmış, çekirdek+alkol grubunda düşmüştür. Zeytin, özellikle Akdeniz diyetinde ve son zamanlarda da herkesin ilgisini çeken geniş metabolik etkileriyle çok kullanılan bir besin kaynağı haline gelmiştir. Zeytin ve yaprağı polifenolik bileşikler bakımından zengindir. Polifenolik bileşikler arasında da oleuropein en bol bulunan ve en aktif olanıdır. Diğer önemli aktif bileşikler ise hidroksitirozol, tirozol ve verbaskozittir. Oleuropein antioksidan, antidiyabetik ve antimikrobiyal etkilere sahiptir. Bu çalışma; çözücü farkının zeytin yaprağındaki polifenolik bileşiklerin miktarlarına ve toplam fenolik madde miktarına etkisinin araştırılması amacıyla planlanmıştır. Saf su, metanol/su ve metanol solventleri kullanılarak zeytin yaprağı ekstrakte edilmiştir. Elde edilen ekstraktta, içerik tayini kromatografik analiz kullanılarak araştırılmıştır. Ayrıca toplam fenolik madde miktarı Folin-Ciocalteu metodu ile spektrofotometrik yöntem kullanılarak tespit edilmiştir. Kromatografik analizler sonucunda metanolik solventlerin oleuropeini zeytin yaprağından daha etkin bir şekilde ekstrakte ettiği bulunmuştur. Solvent olarak su kullanıldığında ise hidroksitirozol, tirozol ve verbaskozitin daha iyi ekstrakte edildiği tespit edilmiştir. Ekstrakte edilen toplam fenolik madde miktarı bakımından solventler karşılaştırıldığında sırasıyla metanol, metanol/su ve suda en yüksek miktarlar bulunmuştur. Sonuç olarak, yüksek hidrofilik karakterlerinden dolayı polifenolik bileşiklerin, polaritesi fazla olan metanolik solventlerde daha verimli ekstrakte edilebileceği belirlenmiştir.Article The Effect of Bioactive Compounds of Prunus Spinosa Leaves and Flowers Mixture on The Antioxidant Capacity in Streptozotocin-Induced Diabetic Rats(Publ House Bulgarian Acad Sci, 2022) Temiz, Mehmet Ali; Okumus, EmineDiabetes is a serious and chronic public health problem worldwide. Hyper-glycemia leads to the production of free radicals thus impairing oxidant/anti-oxidant balance and causing various complications in diabetes. Herbs can pro-vide natural remedies against various diseases in traditional medicine. Phyto-chemicals especially phenolic compounds have some biological properties such as antioxidant and antidiabetic effects. Plant parts of Prunus spinosa are used in traditional medicine against various disorders. This study evaluated the mixture of Prunus spinosa leaves and flowers extract (PSE) on the antioxidant capacity in diabetes. The phenolic profile of PSE was determined by HPLC method. Antioxidant capacity was evaluated with radical scavenging activities as well as total phenolic and flavonoid content. Seven random groups (n = 8) of rats were constituted to evaluate the antioxidant capacity - control, dia-betic, PSE25, PSE50, insulin, metformin, and acarbose. Total oxidant status (TOS) and total antioxidant status (TAS) were analyzed in the liver tissues. Chlorogenic acid and ellagic acid were found to be the main compound in the mixture of P. spinosa leaves and flowers extract. Antioxidant capacity of PSE exhibited effective radical scavenging activity. Although oxidative stress seri-ously increased in diabetes, PSE25 and PSE50 supplementation significantly recuperated liver antioxidant status. In conclusion, PSE markedly has antioxi-dant capacity through its high phenolic compounds, thus PSE treatment may have potential antioxidant effects for health benefits in diabetes.Master Thesis The Effect of Celtis Tournefortii (hackberry) on Total Antioxidant and Oxidant Status Against Ccl4-Induced Hepatotoxicity(2018) Sürücü, Nuri Mustafa; Temür, Atilla; Temiz, Mehmet AliBu çalışma, CCI4 ile oluşturulan karaciğer hasarına karşı Celtis tournefortii (Ct)(Doğu Çitlembiği)'nin iyileştirici ve koruyucu etkilerinin araştırılması amacıyla planlanmıştır. Çalışmada 32 adet Wistar-albino türü sıçan kullanıldı. Sıçanlar kontrol grubu, CCI4, CCI4+Ct ve Ct olmak üzere 4 gruba ayrıldı. Celtis tournefortii yaprak ekstraktının 28 günlük muamele sonrası; aspartat aminotransferaz (AST), alanin aminotransferaz (ALT), laktat dehidrogenaz (LDH) ve alkalen fosfataz (ALP) enzim seviyeleri, karaciğer dokusu süperoksit dizmutaz (SOD), glutatyon peroksidaz (GPx), katalaz (KAT) enzim aktiviteleri ile glutatyon (GSH) ve malondialdehit (MDA) içerikleriyle birlikte total antioksidan seviye (TAS) ve total oksidan seviye (TOS) spektrofotometrik yöntemle tespit edilerek değerlendirilmiştir. Elde edilen bulgular CCI4 grubunda serum AST, ALT, ALP ve LDH enzim seviyelerinin yükseldiğini, CCI4+Ct grubunda ise düştüğünü göstermiştir. Karaciğer dokusunda MDA, TOS ve OSI düzeyleri istatistiki olarak artmakla birlikte, CCI4+Ct grubunda azaldı. SOD, GPx, KAT, GSH ve TAS düzeyleri CCI4 grubunda anlamlı olarak azalırken, CCI4+Ct grubunda anlamlı olarak artmıştır. Gruplardaki anitoksidan enzim seviyelerindeki bu değişiklik ve iyileşmenin nedeni doğu çitlembiği yaprağının karaciğer koruyucu etkisi ve antioksidan rolünden kaynaklanmaktadır.Article The Effect of Olive Leaf Extract on Digestive Enzyme Inhibition and Insulin Production in Streptozotocin-Induced Diabetic Rats(Ankara Univ Press, 2019) Temiz, Mehmet Ali; Temur, AtillaOlive leaf has natural bioactive compounds, mainly oleuropein, that are widely considered to have potentially beneficial effects on health. This study aimed to evaluate the effects of olive leaf extract (OLE) on the inhibition of carbohydrate digestive enzymes, and immunohistochemical study of insulin in the pancreas of in vivo streptozotocin-induced diabetic rats. Blood glucose levels, insulin, glycated hemoglobin (HbA(1c)), alpha-amylase and alpha-glucosidase activities, and an immunohistochemical study were performed at the end of the experiment. In the OLE treated group, blood glucose levels and HbA(1c) significantly decreased while insulin levels increased. Besides this, OLE treated group showed remarkable inhibitory activities on alpha-amylase and alpha-glucosidase compared with the Acarbose treated group. It was observed that OLE exhibited partial positive immunoreaction for insulin in beta-cells through immunohistochemical analysis. Considering that OLE is more tolerable for digestion system compared to acarbose, it may be a better fitoformulation for antidiabetic medications. OLE could offer an additional beneficial effect for the treatment of diabetes.Article Effects of Olive Leaf (Olea Europaea L.) Extract on Some Biochemical and Haematological Parameters in Diabetic Rats(Kastamonu Univ, Orman Fak, 2018) Temur, Atilla; Temiz, Mehmet AliThis study was aimed to investigate the possible protective effects of different amount of olive leaf extracts and infusion on some biochemical and haematological parameters of streptozocin-induced experimental diabetic rats. For this purpose, 56 Wistar albino rats were randomly divided into 7 groups (n=8). Biochemical and haematological analyses were performed at the end of the 21-day trial. It was observed that HGB, HCT, MCV were positively affected (p<0,05), while RBCs were not statistically significant affected in all OLE-applied groups. In diabetes+OLE-50 and OLE-100 groups, WBCs significantly decreased compared to the that of control group (p<0,05), while statistically significant change was not observed in the OLE-25 group. The cholesterol values of the diabetes+OLE-25 group were significantly lower than those of the control group (p<0,05), while the increase in the diabetes+OLE-100 group was significant at the statistical level (p<0,05). The increase in diabetes+OLE-50 group was not significant. In the diabetes+OLE-25, diabetes+OLE-50 and diabetes+OLE-100 groups, triglyceride decreased significantly (p <0,05) in comparison to diabetic group. In the infused diabetic group, RBC, HGB, HCT, MCV, MPV, PDW and EOS were significantly increased in the positive direction (p <0,05). Infusion was found to have negative effect on triglyceride; LDL, Cl, Na. HDL, Mg and P were found to be affected positively. It was determined that the most effective dose on cholesterol and triglycerides in OLE-administered groups is OLE-25 mg/kg. Infusion was found to have positive effect on HDL, while LDL was affected negatively. On the other hand, the decrease in cholesterol was found to be not statistically significant, while the decrease in triglyceride was significant.Article Hazelnut By-Products as a Valuable Resource: Lipid Peroxidation Inhibition Effect, Bioaccessibility and Antidiabetic Properties(Elsevier, 2025) Okumus, Emine; Temiz, Mehmet AliThe aim of this study is to determine the chemical and molecular properties of hazelnut by-products, to measure the bioaccessibility values of phenolic compounds found in the structure of these wastes and to reveal their antidiabetic and lipid peroxidation (LPO) inhibition effects. The highest moisture and protein content were found in skin samples with 6.33% and 7.70%, respectively. The predominant fatty acid in hazelnut skin is oleic acid, which constitutes 78.65% of the total fatty acid composition. The highest antioxidant activity value was measured in the shell with an IC50 value of 19.08 mg/mL. The sample with the highest content of gallic acid and protocatechuic acid content was the skin extract. The dominant phenolic component in the shell and husk samples was kaempferol, while in the skin sample it was rutin. Shell bioaccessibility was found to be much higher for o-coumaric acid (45.83%), rutin (49.25%) and kaempferol (46.32%) components. The bioaccessibility of the shell sample in total phenolic concentration was measured as 45.87%. The highest LPO inhibition value belonged to the shell sample with an IC50 of 4.50 mg/mL, while the lowest value belonged to the husk sample with an IC50 value of 25.03 mg/mL. The highest antidiabetic effect was observed in the shell sample with values of 3.55 mg/ mL and 25.44 mg/mL for alpha-amylase and alpha-glucosidase inhibition, respectively. The highest recovery belonged to 4-Hydroxybenzoic acid in the skin sample. As a result, it was concluded that hazelnut by-products, especially the shell, exhibit high antioxidant effects, have antidiabetic and LPO inhibition properties thanks to their bioactive components and high bioaccessibility values. Thus, they have valuable usage opportunities in the fields of food, pharmacology and medicine.Master Thesis İnvestigation of the Protective Effect of Oriental Hackberry (celtis Tournefortii) on Copper Toxicity in Rats(2018) Akgeyik, Muhammed Yusuf; Temür, Atilla; Temiz, Mehmet AliBu çalışmada, Bakırsülfat ile karaciğerlerinde toksisite oluşturulmuş sıçanlarda Doğu Çitlenbiğinin(Celtis tournefortii) koruyucu etkisine bakıldı. Otuz iki adet Wistar albino sıçanlar toplam dört gruba ayrıldı (n=8) (Kontrol, Bakırsülfat, Bakırsülfat+Celtis, Celtis). Kontrol grubundaki her bir fareye 28 gün boyunca 1 ml serum fizyolojik verildi. Bakırsülfat ve Bakırsülfat+Celtis grubundaki farelere ise 28 gün boyunca Ct ekstraktı bunun yanında Pazartesi ve Cuma günleri Bakırsülfat çözeltisi verildi. Celtis grubuna ise 28 gün boyunca Ct ekstraktı verildi. Aspartat Aminotransferaz (AST), Alanin Aminotransferaz (ALT), Laktat Dehidrojenaz (LDH), ve Alkalen Fosfataz (ALP) seviyeleri belirlendi. Ayrıca karaciğer dokusunda malondialdehit (MDA) içeriği, glutatyon (GSH) miktar tayini yapıldı. Yine Antioksidan antioksidan savunma sistemi parametrelerinden Superoksit Dismutaz (SOD), Glutatyon Peroksidaz (GPx) ve Katalaz (KAT) aktiviteleri ölçüldü. Bununla birlikte Total Antioksidan Seviyesi (TAS) ve Total Oksidan Seviyesi (TOS) de belirlendi. Elde edilen bulgulara göre BS grubunda AST, ALT, LDH ve ALP seviyeleri yükselmesine rağmen, BS+Ct grubunda düştü. Karaciğer dokusunda MDA ve TOS, BS grubunda istatistiki olarak artmakla birlikte BS+Ct grubunda azaldı. GSH ve TAS, BS grubunda anlamlı olarak azalırken BS+Ct grubunda anlamlı olarak artmıştır. Karaciğer dokusunda bulunan antioksidan enzimler SOD, KAT ve GPx aktiviteleri BS grubunda anlamlı olarak azalırken BS+Ct grubunda anlamlı olarak artmıştır. Elde edilen bu sonuçlara göre Ct bitkisinin terapötik olarak bakır zehirlenmelerine karşı koruyucu etkisi olabilir.Article Kısa Süreli Hiperglisemide Prunus Spinosa Tarafından Bağışıklık Sistemi ve Hemogram Parametrelerinin Modülasyonu(2023) Temiz, Mehmet Ali; Okumus, EmineDiabetes mellitus (DM), prevalansı ve ilişkili komplikasyonları nedeniyle acil küresel dikkat gerektiren, kan glukoz düzeyi ile ilişkili kronik hastalıklardan biridir. DM, bağışıklık sistemini baskılayarak diyabette çeşitli komplikasyonların gelişiminde önemli rol oynayan oksidatif strese yol açar. Prunus spinosa, yüksek antioksidan aktivitesi sayesinde geçmişten günümüze birçok hastalığın tedavisinde kullanılan bir bitkidir. Bu nedenle, bu çalışma kısa süreli diyabetik durumda P. spinosa yaprak ve çiçek karışımının bağışıklık sistemi üzerindeki etkisini araştırmayı amaçlamaktadır. Çalışmada, diyabetik sıçanların karaciğer dokularında P. spinosa’nın adenozin deaminaz (ADA), ksantin oksidaz (XO) ve miyeloperoksidaz (MPO) aktiviteleri ve hemogram parametreleri üzerine etkilerini belirlemek amacıyla biri kontrol, diğerleri altı diyabetik grup olmak üzere 7 gruba ayrılan 56 Wistar albino erkek sıçan kullanıldı. Bu gruplardan ikisine farklı konsantrasyonlarda (25 ve 50 mg/kg canlı ağırlık) bitki ekstraktı verildi ve sonuçlar insülin, metformin ve akarboz grupları ile karşılaştırıldı. Sonuçlar, uygulanan her iki dozun diyabetin neden olduğu değişen hematolojik parametreler üzerinde modüle edici bir etkiye sahip olduğunu gösterdi. Tedavi grupları, diyabetik gruba kıyasla ADA, XO ve MPO aktivitelerini önemli ölçüde azalttı. PSE50’nin etkilerinin diğer tüm tedavi gruplarından daha etkili olduğu bulundu. Bitkinin diyabetteki bu etkileri, terapötik immün düzenleyici potansiyeline bağlı olabilir. Sonuç olarak P. spinosa, diyabet üzerinde bir adjuvan olarak değerli bir kaynak olabilir.Article Olive (Olea Europaea L.) Leaf Extract Boosts Antioxidant Status and Attenuates Hepatic and Pancreatic Changes in Diabetic Rats(Centre Advancement Applied Sciences, 2019) Temiz, Mehmet Ali; Temur, AtillaThe purpose of this study was to evaluate the antioxidant, oxidative DNA damage and hepatoprotective effects of olive (Olea europaea L.) leaf extract (OLE) on experimental diabetic rats. These effects of OLE were studied using streptozocin-induced diabetic rats. Antioxidant enzyme activities, oxidative DNA damage analysis and protein oxidation were evaluated spectrophotometrically methods. In addition, histological examination of the liver and pancreas was performed routinely with paraffin embedding and staining method. Protein carbonyl level and 8-hydroxy- 2' - deoxyguanosine (8-OHdG) concentration decreased in OLE administrated group compared to diabetic group. Further, OLE treatment significantly increased superoxide dismutase and glutathione peroxidase activities. It was observed that OLE remarkably attenuated degenerative and necrotic changes in liver and pancreatic tissues. Consequently, it appears that OLE strengthens the antioxidant defense system, protects against oxidative DNA damage, and has a hepatoprotective effect on diabetic rats.Article Protective Effect of Celtis Tournefortii Against Copper-Induced Toxicity in Rat Liver(veterinarni A Farmaceuticka Univerzita Brno, 2021) Temiz, Mehmet Ali; Temur, Atilla; Akgeyik, Yusuf; Uyar, AhmetThis study aimed to investigate the antioxidant and hepatoprotective effects of Celtis tournefortii fruit extract (Ct) against copper-induced liver damage in rats. Thirty-two Wistar Albino rats were divided into four equal groups (n = 8): Control, Copper (Cu), Copper + C. tournefortii (Cu+Ct), and C. tournefortii (Ct). Superoxide dismutase (SOD), glutathione peroxidase (GPx), and catalase (CAT) activities, glutathione (GSH) concentration, malondialdehyde (MDA), total antioxidant status (TAS) and total oxidant status (TOS) were analysed in the liver tissues. Liver histopathology was also evaluated. Alanine aminotransferase and lactate dehydrogenase significantly decreased in the Cu+Ct group compared with the Cu group. Oxidative stress parameters MDA and TOS significantly increased with copper administration, whereas they decreased with C. tournefortii co-treatment compared to Cu group. GSH concentration and TAS showed significant decreases with copper administration. Celtis tournefortii co-supplementation with copper significantly enhanced antioxidant parameters such as TAS, SOD, and GPx. Celtis tournefortii remarkably attenuated degenerative and necrotic changes caused by the oral exposure of copper in the liver tissue of rats. Celtis tournefortii may provide amelioration of the antioxidant status and moderation of severity of liver damage against copper toxicity. The therapeutic use of C. tournefortii may exhibit protective effects in liver injury treatment.
