Conference Object Changing of Iaa (indol-3 Acid) Content at Different Organs of Walnut (Juglans Regia L) After Grafting(international Society Horticultural Science, 1997) Sen, SM; Kazankaya, A; Testereci, H; Yoruk, IHIn this study, Walnut seedlings (Juglans regia L.) were grafted on dormancy period and placed into controlled conditions with 26 +/- 2 degrees C temperature and 60-70% humidity. IAA changes in shoot of graft, root and graft area were measured in 1, 14, 21, 28, 35, 42 and 49th days following grafting by high performance liquid chromatography (HPLC). IAA levels in the shoot of graft was maximum (251 ng/g) at the 21 st day, and minimum (59 ng/g) at the 49 th days. The levels of IAA of graft area were maximum (218 ng/g) at the 14 th day, and minimum (53 ng/g) at the 49th days. IAA concentration in roots were highest (213 ng/g) at the 14 th day later and minimum (42 ng/g) at the 49 th day. IAA levels showed rapid increase for 3 weeks in the shoot of graft and for 2 weeks in the root, and the graft area. IAA concentration had the decreasing and parallel tendency with each other after 21 th day.Article Separation of Retinol Acetate, Vitamin a Palmitate, Alpha- and Gamma-Tocopherol by Reverse-Phase High Performance Liquid Chromatography and Determination of These Vitamins in Van Lake Fish (Chalcalburnus Tarichi)(Scientific Technical Research Council Turkey, 1997) Testereci, H; Yoruk, IH; Akyuz, A; Sagmanligil, HThe resolution properties of several commercial columns for vitamin A and E standards by reverse phase chromatography methods have been examined, Hypersil MOS column with methanol-water (96:4) and acetonitrile-water (90:10) mobile phase, Shimadzu-ODS column with methanol-water (96:4) mobile phase have acceptable resoluion in separation of vitamin A esters. Shimadzu-ODS column/96% methanol and Shimadzu-C8/90% acetonitrile have the acceptable resolution ratio for vitamin E isomers. Methanol extracts of heart muscle and gill in Van lake fish (Chalcalburnus tarichi have been examined for vitamin A and E by high performance liquid chromatography. Avarage retinol acetate, vitamin A palmitate, alpha tocopherol, gamma tocopherol in heart muscle of Van Lake fish, have been found as 162+/-23 ng/g, 18+/-2.2 ng/g, 8.5+/-1.0 mu g/g, 505.7+/-94 ng/g, respectively. The vitamin levels in gills have been determined as retino[ acetate 231.5+/-38 ng/g, vitamin A palmitate 23.8+/-3.5 ng/g, alpha tocopherol 12.4+/-0.77 mu g/g, gamma tocopherol 350.1+/-97 ng/g. A significant differences have been found among the retinol acetate-vitamin A patmitate and alpha-gamma tocopherol values in heart muscles and gills (P<0.05). A negative correlation for retinol acetate between heart muscle and gill (r:-.86), vitamin A palmitate between heart muscle and gill (r:-.73) and between retinol acetate-alpla tocopherol in heart muscles tr:-.71) have been observed. A positiv correlation between retinol acetate and alpha tocopherol in gills (r:+.52) has been found. No correlation has seen between heart muscles and gills for gamma tocopherol values.Article A Study on Esterase Activity of Carbonic Anhydrase From Van Lake Fish (Calcalburnus Tarichi)(Tubitak Scientific & Technological Research Council Turkey, 1999) Testereci, H; Sekin, S; Ekin, SNo study has been reported on carbonic anhydrase enzyme of the fish (Calcalburnus tarichi) living in the carbonated Van Lake. Having partial purification of Carbonic anhydrase from gill and liver of Van Lake fish by ethanol-chloroform and acetone-chloroform, the esterase activity was performed by using p-nitrophenyl acetate as a substrate on spectrophotometer. The effects of the some kinetic parameters (km, Vo, Vmax, Na2CO3 activation, optimum pH etc.) have been examined When Lineweaver-Burk diagram of liver extracts (at 25 degrees C) is examined, ethanol has an apparent competitive inhibition compare to acetone extracts. So Km of the substrate (p-nitrophenyl acetate) is 0.95 mM is for ethanol extract and 0.54 mM of that is found for acetone extracts. In two extracts from gill (at 25 degrees C), ethanol seems to have a non-competitive inhibition comparing to acetone, Km for both extracts is 0.39 mM. Na+/CO32- ions induce esterase activity, Na+/CO3-2 ions are not competitive with the inhibitory effects of ethanol. Infect, inhibition of ethanol on gill esterase activity appears to be competitive which is understood by addition of Na+/CO3-2 Since carbonate ions and p-nitrophenyl acetate act on two different catalytic site of this enzyme, both have formed a cooperative effect. It is clear that carbonic anhydrase activity has not been affected till pH:7.4. Although pH above 8.0 enzyme activity has seem to increase, this increase is due to kinetic reaction between substrate (p-nitropheny acetate) and buffer at pH:8-11.5. So. the esteraseactivity of this enzyme at this high pH, can not be evaluated. for the esterase activity of this enzyme. The optimal and unaffected pH for the substrate is 7 for the esterase activity of carbonic anhydrase. "Esterase" activity could be useful for determining "anhydrase" activity of Carbonic anhydrase enzyme of Van lake fish. Stimulation of esterase activity of carbonic anhydrase has been shown by addition of CO32- ions. Carbonic anhydrase present in gill and liver of Van Lake fish has a significant regulatory role for internal pH balance.
