Article Analysis of Genetic Diverity Among Different Melon Accessions Using Peroxidase Gene Markers(Parlar Scientific Publications (p S P), 2020) Kocak, Metin; Turan, Sibel; Erdinc, Ceknas; Yildiz, MehtapPlant peroxidases encoded by a broad multi gene family have important roles in many physiological events such as salt tolerance, auxin metabolism, seeding, protection against pathogens and oxidative stress. In this study the genetic diversity and population structure of 45 different melon (Cucumis melo L.) genotypes from a variety of regions in Turkey and different countries were investigated using peroxidase gene polymorphism markers. 12 polymorphic primers were used to obtain 97 scorable bands for the 45 melon genotypes. Of these bands, only 62.9% were determined to be polymorphic. The number of bands per primer ranged from 6 to 11 and the average number of bands was 8.08. The lowest and the highest polymorphism information contents (PIC) were 0.49 (Pox 12 FaRa) and 0.99 (Pox 13), respectively. The mean PIC value was 0.75. The lowest genetic similarity was found between CU-365 (conomon) and CU-247 (0.143), on the other hand, the highest genetic similarity was observed between CU-360 (cantalopensis) and CU-389 (cantalopensis) (0.875). Using the STRUCTURE computer program, all melon accessions were determined to separate into two subpopulations. These results support the usability of the PDX gene marker system in genetic diversity studies. Also, the data obtained are considered to be a guide for other studies to be performed on melons in the future.Master Thesis Determination of Virus Resistance With Mechanic Inoculation and Molecular Methods in Some Melon Accession Collected From Van Lake Basin(2018) Turan, Sibel; Erdinç, ÇeknasBu çalışmada, Van Gölü Havzasından toplanan 22 adet kavun genotipi ile 3 adet dayanıklı genotip ve 2 adet ticari çeşitte hem mekanik inokulasyon yöntemi kullanılarak genotip ve çeşitlerin ZYMV (Zucchini Yellow Mosaic Virus)'ye karşı dayanıklılığını belirlemek hem de bazı SCAR ve CAPS markör yöntemleri ile kavun genotip ve çeşitlerinde ZYMV (Zucchini Yellow Mosaic Virus), CMV (Cucumber Mosaic Virus) ve WMV (Watermelon Mosaic Virus) türlerine dayanıklılık belirteçlerinin varlığının araştırılması amaçlanmıştır. Mekanik inokulasyon sonrasında genotiplerde morfolojik gözlemler yapılarak 0-5 skalasına göre hastalık şiddeti belirlenmiştir. Çalışmanın ikinci aşamasında bu genotiplerde 3 adet SCAR ve 4 adet CAPS primeri olmak üzere 7 adet primer ile tarama yapılarak ZYMV, CMV ve WMV'ne karşı dayanıklılık ile ilgili belirteçlerin varlığı tespit edilmeye çalışılmıştr. SCOPE14541 primerinde 21 genotipte bant tespit edilirken, Creb-2 dayanıklılık genine bağlı olarak geliştirilen SCAPB051046 primerinin 25 genotipten yalnızca U6, U13 ve YYU6 genotiplerinde bant verdiği görülmüştür. ZYMV ve WMV' ye karşı dayanıklılık genlerine bağlı olarak geliştirilmiş olan VirSq-F19 primeri YYU47 genotipi dışında tüm genotiplerde bant vermiştir. Zym dayanıklılık geni ile ilişkili 4 CAPS primerinden sadece ikisinden sonuç elde edilmiştir. dCAPS-G99A ve CAPS-T86C primerleri ile istenilen gen bölgesi çoğaltılmış fakat kesim enzimleri (Dra I/MnI I) için istenilen bant aralıklarında kesme işlemi gerçekleşmemiştir.Article Effects of Hybrid Drying on Kinetics, Energy Analysis and Bioactive Properties of Sour Black Mulberry (Morus Nigra L.)(inst Tecnologia Parana, 2023) Cetin, Necati; Turan, Sibel; Gurcan, Kahraman; Turkay, Seyda Nur; Duman, Suheda; Karaman, KevserDue to the short harvest season and their sensitivity to storage, the preservation of fresh mulberry fruits is a very important process. Drying is a method used to preserve mulberry fruits in the long term. In this study, response surface methodology (RSM) was applied for the optimization of hybrid drying conditions of two different sour black mulberries. The linear and interaction effects of independent parameters such as temperature (50, 60 and 70 & DEG;C) and microwave power (100, 200 and 300 W) variables were determined on mulberries. Bioactive properties and energy aspects were monitored as influenced by drying conditions. According to the results increase in microwave power provided a significant decrement in the specific energy consumption (SEC) and the total anthocyanin content (TAC), while increase in the energy efficiency (& eta;en) and total phenolic content (TPC) for both genotypes. In all cases, statistical values showed that all drying curves of black mulberry were best described by the Logistic model. Multiple response optimization was carried out for studied parameters and it was concluded that maximum antiradical activity (ARA), TPC, TAC, & eta;en and minimum drying time (DT) and SEC values would be at 300 W-50 oC (desirability=0.842) and 300 W-66.5 oC (desirability=0.744), for Morus nigra 1 (MN1) and Morus nigra 2 (MN2), respectively. According to the finding, the greatest TPC, ARA, TAC, DT, SEC and & eta;en were determined as 20.10 mg GAE/g, 86.00%, 456 mg/kg, 330 min, 18.59 kWh/kg and 9.04% for MN1, and 18.08 mg GAE/g, 83.92%, 835.81 mg/kg, 330 min, 16.16 kWh/kg and 10.40 % for MN2, respectively.Article The First Report About Genetic Diversity Analysis Among Endemic Wild Rhubarb (Rheum Ribes L.) Wild rhubarb (Rheum ribes L.) is a wild vegetable grows especially in the eastern region of Turkey and is an endemic species. In this study, genetic relationships among 80 wild rhubarb genotypes collected from some regions of Lake Van Basin, which are in the distribution area, were tried to be determined by iPBS marker system. At the same time, a commercial variety of R. rhabarbarum, which is a cultivated species, was used as control. PCR studies were conducted with 23 iPBS primers to determine genetic relationships, and a total of 340 scorable bands were obtained. 100% polymorphism rate was obtained from all primers studied. While the average PIC value was found to be 0.90, the highest value was found to be 0.97 from the primer # 2220. It was determined that the genotypes were divided into 3 basic groups in the dendogram created with UPGMA based on Jaccard similarity coefficient.Article Genetic Characterization of Rheum Ribes (Wild Rhubarb) Genotypes in Lake Van Basin of Turkey Through Issr and Ssr Markers(Friends Science Publishers, 2019) Ekincialp, Aytekin; Erdinc, Ceknas; Turan, Sibel; Cakmakci, Ozlem; Nadeem, Muhammad Azhar; Baloch, Faheem Shehzad; Sensoy, SuatRheum ribes L. (wild rhubarb) is one of the less known plants to the world and the only species from the Rheum genus present in Turkey. In this study, one R. rhabarbarum (as check genotype) and 80 R. ribes genotypes were collected from different geographical locations of Turkey for the investigation of diversity and genetic structure using ISSR (Inter Simple Sequence Repeat) and SSR (Simple Sequence Repeats) markers. SSR markers reflected higher (100%) polymorphism as compared to the ISSR marker. However, ISSR markers produced higher average Polymorphism Information Content (PIC) value (0.805) than the SSR markers (0.724). A Similar range of (PIC) values with ISSR markers was found greater (0.935-0.395) as compared to the range of SSR makers (0.88-0.47). Using Jaccard similarity index, genetic distance was measured for both markers and average genetic distance was found to be higher with ISSR markers as compared to the SSR markers. Neighbor-joining analysis clustered genotypes into 3 groups for both marker systems. During this study some distinct genotypes like R. rhabarbarum, YYUERC19, YYUERCO9 and YYUNIER65 were investigated that can be used as candidate parents for the development of R. ribes L. varieties. Structure analysis grouped the genotypes according to altitude by clustering genotypes having at more than 2000 m in one group and genotypes less than 2000 m altitude in another group. Genetic variations observed in this study can be applied to investigate various traits of interest for the R. ribes L. breeding. (C) 2019 Friends Science Publishers
