Article Adverse Effects of Oxytetracycline and Enrofloxacin on the Fertility of Saanen Bucks(Springer Science and Business Media B.V., 2021) Yücel, U.M.; Koşal, V.; Kara, M.; Taşpınar, F.; Uslu, B.A.This study aimed to determine the adverse effects of oxytetracycline and enrofloxacin application on the fertility of Saanen bucks. For this purpose, twenty-four bucks were divided into three groups. Group I (control group) received only 5 ml of 0.9% NaCl for 7 days, group II was given a single dose of 20 mg/kg oxytetracycline and group III was given at a dose of 2.5 mg/kg per day for 7 days intramuscularly. Serum and semen samples were collected from the bucks at post-treatment 1, 3, 5, 7, and 9 days and examined spermatological parameters (quantity, motility, density, abnormal sperm ratio, and live-dead sperm ratio), serum testosterone levels (with ELISA) and sperm DNA parameters (with Comet assay). The results showed no change in sperm volume, abnormal sperm rate, and dead-live sperm ratio in group II and III following oxytetracycline and enrofloxacin administration. However, a decrease in sperm density, sperm motility, mass activity, and testosterone levels, and an increase in sperm DNA damage were detected. These spermatological parameters (density, motility, mass activity) and testosterone levels were less decreased and sperm DNA damage was less increased in group II than group III. The greater damage in group III may be attributed to the longer duration of enrofloxacin administration compared to oxytetracycline and the effect of enrofloxacin on DNA. The results obtained from this study suggest that usage of oxytetracycline and especially enrofloxacin should be restricted and antibiotics with fewer side effects on sperm should be preferred in Saanen bucks during the reproduction period. © 2021, The Author(s), under exclusive licence to Springer Nature B.V.Article The Effects of Gnrh Injection After Intravaginal Sponge, Cidr-G and Ear Implant Application in Coloured Mohair Goats During Early Anoestrus Season(KAFKAS UNIVERSITY, 2009) Uslu, B.A.; Gülyüz, F.In the present, in the Colored Mohair Goats, during anoestrus season, vaginal sponge, CIDR-G and ear implants used to stimulate oestrus and additionally GnRH injection. In this study 57 female and 12 male coloured mohair goats aged between 2-4 year old were used. The goats were divided randomly into three equal groups. To the 1st group (n=19), 30 mg Flourogestone acetate (FGA) containing sponge applied for intravaginally for 14 days. The day sponges removed, 300 IU eCG were applied. To the 2nd group (n=19), 0.3 g P4 containing implants (CIDR-G) were applied for intravaginally for 13 days. The day implants removed, 300 IU eCG were injected. To the 3th group (n=19), the day implants applied, 5 mg oestradiol valerate and 3 mg norgestomet were injected intramuscularly and 3 mg ear implant were applied during 10 days. The day implants removed, 300 IU eCG were injected. Goats in group I, II and III showing oestrus symptoms were divided into 2 subgroups. Twelve days after mating 1 ml GnRH (25 mg Lesireline acetate) (group IA, group IIA, group IIIA) and other groups 1 ml serum phsiologic (group IB, group IIB, group IIIB) were injected. Pregnancy ratios in group IA were 60%, group IB 25%, group IIA 80%, group IIB 25%, and group IIIA 33.33%. On the other hand, in group IIIB no pregnancy were observed. GnRH injected 10 goats in group IA, 6 of them 10 goats in group IIA, 8 of them, 9 goats in group III, 3 of them were observed to be pregnant. As a result, at early oestrus season stimulation of oestrus by three different method were effective in coloured mohair goats, vaginal sponges containing FGA and P4 containing CIDR-G cause to develope oestrus early. FGA containing vaginal sponge, P4 containing CIDR-G and norgestomet containing crestar ear implant application and 12 days these aplication, GnRH injection had a positive effect on pregnancy rate.Article The Effects of Levamisole and Albendazole on Spermatological Parameters, Testosterone Levels, and Sperm Dna Damage in Saanen Bucks(Springer Science and Business Media B.V., 2021) Yücel, U.M.; Koşal, V.; Taşpınar, F.; Uslu, B.A.This study aimed to investigate the effect of levamisole and albendazole on spermatological parameters, testosterone levels, and sperm DNA damage in Saanen bucks. For this purpose, twenty-four Saanen bucks were divided into three groups as control, levamisole, and albendazole administration group. The control group received only water (20 ml, oral), the levamisole group received 7.5 mg/kg of levamisole (2 oral tablets once daily for 2 days) + water (20 ml, oral), and the albendazole group received 7.5 mg/kg of albendazole (1 oral tablet) + water (20 ml, oral). Semen and blood samples were collected from all animals, both before drug application (day 0) and within a 2-day interval after drug application between day 1 (day of the treatment) and day 11. Spermatological parameters were evaluated immediately after collection. Testosterone levels were also measured from the blood samples with ELISA. Sperm DNA damage was determined with comet assay. The present research showed that especially albendazole administration decreased spermatological parameters and levamisole administration decreased testosterone levels. Significant sperm DNA damage was seen after both albendazole and levamisole administration. As a result, albendazole and levamisole administration should be used carefully on Saanen bucks, especially during the breeding season. © 2021, The Author(s), under exclusive licence to Springer Nature B.V.Article The Effects of Pregnancy, Early Lactation Weeks and the Number of Suckling Kittens on the Serum Mineral and Metabolite Profiles of Van Cats in Breeding Season(Veteriner Fakultesi Dergisi, 2012) Uslu, B.A.; Şendaǧ, S.; Uçar, Ö.; Gülyüz, F.; Taşal, I.; Çetin, N.; Wehrend, A.In Van cats, changes in serum mineral (Na-Sodium, K-Potassium, Ca-Calcium, P-Phosphorus, Mg-Magnesium, Cl-Chlorine and Fe-Iron) and metabolite concentrations (TG-Triglycerides, COL-Cholesterol, T.Prot.-Total protein and ALP-Alkaline phosphatase) were investigated before, during and after the pregnancy. A total of 22 mature cats (19 queens and 3 fertility-proven tomcats) were used as material. Thereafter, 13 queens showed oestrus while others (n=6) did not. Samples of venous blood were collected from anoestrous and oestrous queens upon hand-mating, on the 54th day. After parturition (n=7), the number of live kittens was recorded and additional samples were collected on the 5th, 12th and 19th days of lactation. Results were also considered for small (n=3) and large number (n=4) of suckling kittens (≤4 or >4 litters, resp.). For minerals, there was a significant (P<0.01) fluctuation in K levels during the prolonged lactation with large number of suckling kittens. However, significant (P<0.05) decrease was found for P value as the lactation period prolonged. Meanwhile, for metabolites, TG levels in previously-mated queens (becoming pregnant or not) were significantly (P<0.01) higher than those both in anoestrus and in lactation. Also, COL levels in pregnant queens were significantly (P<0.05) higher than those in all other occasions and further that there were significant decreases in COL levels both in the first and the second weeks of lactation. Collectively, present findings suggest that; a) the concentrations of K and P minerals and metabolites studied per se (TG, COL, T.Prot. and ALP) changed markedly during the late pregnancy and prolonged lactation, and b) declines in minerals (P) and metabolites (COL) become even greater when the number of suckling kittens increased in queen Van cats during breeding season.Article The Effects of Sildenafil Citrate and Vitamins A, C and E on Testicular Damage in Alloxan-Diabetic Rats(Medwell Journals, 2012) Uslu, B.A.; Ilhan, F.; Gulyuz, F.; Karasu, A.; Tas, A.; Sendag, S.; Wehrend, A.Sildenafil citrate is an active ingredient used successfully in the treatment of erection disorders caused by a variety of factors. The purpose of this study is to research the effects of Sildenafil citrate and vitamins A, C and E on testicular degeneration in rats with alloxan diabetes. The rats were divided into II groups; healthy control and diabetic groups. The diabetic rats were divided into X groups with ten animals in each group. Group I consisted of control diabetic rats that were given only distilled water, Group II was the diabetic group treated with glibenclamide; Group III was the diabetic group treated with insulin, Group IV was the diabetic group treated with Sildenafil citrate, Group V was the diabetic group treated with Vitamin A, Group VI was the diabetic group treated with vitamine, Group VII was the diabetic group treated with vitamin E, Group vni was the diabetic group treated with Sildenafil citrate and vitamin A, Group IX was the diabetic group treated with Sildenafil citrate and vitamin C and Group X was the diabetic group treated with Sildenafil citrate and vitamin E. Testis tissue samples were collected on the 3, 7 and 15th days of treatment in control and diabetic groups. In the histopathological examination of the testicles of diabetic rats, the seminiferous tubules and interstitium of testes in the control groups were normal and the complete spermatogenic cells in the seminiferous tubules were healthy and uniformly arranged. Severe degeneration and tubular atrophy was observed in the seminiferous tubules on the 3, 7 and 15th days in the group with induced alloxan diabetes given only distilled water (Group I). The lesions on day 3rd in the diabetic group given glibenclamide (Group II) were similar to those in the diabetic control group given only distilled water. All of the tubules had been affected by the 3rd day in group three which was given insulin. A small number of spermatogenic cells and giant cells were observed in some tubules. However, almost al of the degenerated tubules had healed on the 7 and 15th days. Even though, recovery on day 15th was very similar in Groups IV-X recovery was much more pronounced in Groups VIII-X beginning from day 3rd. © Medwell Journals, 2012.Article Induction of Oestrus With Single or Split Ecg Administrations in Turkish Van Cats During the Non-Breeding Season(Veteriner Fakultesi Dergisi, 2012) Uslu, B.A.; Gülyüz, F.; Uçar, O.Effects of equine chrionic gonadotrophin (eCG) upon reproductive traits of Van cats during the non-breeding season were investigated herein. In the non-breeding season (October), a total of 18 queens were divided equally into three experimental groups, as follows: one ml of 0.9% saline solution, as single placebo injection (intramuscularly, i.m.) was given to queens, as control (Group I, n=6), while single (250 IU, i.m.) or split dose of eCG (75 IU/daily for 5 d) was injected in Group II and III (n=6 each), respectively. Data from the rates of oestrus, days of mating, pregnancy, parturition, litter size, survival of kittens and lactation period were recorded. The oestrus rates (100% both) in Groups II and III were significantly (P<0.01) higher than those in controls (33.3±21.1%). Days of mating (9.0±1.0, 4.7±0.7 and 5.8±0.4 d) were significantly (P≤0.01) different between the groups (Group I, II and III, resp.). Pregnancy period (65.8±0.5 d) was significantly (P<0.05) longer in Group III ((65.8±1.0 d) than in Group II (62.4±1.0 d) and controls (62.5±0.5 d). The litter size tended (p=0.062) to be higher in Group II (4.6) than those in Group III (2.6) and controls (3.0). Finally, the lactation period (105.2±2.6 d) was significantly (P<0.05) longer in Group III than those in Group II (94.6±1.7 d) and controls (95.0±5.0 d). Findings suggest that; both single and split eCG injections were equally highly effective for induction of fertile oestrus in endemic Van cats in non-breeding season.
