Article Does Mdr1 Promoter Methylation Affect Temozolomide Resistance? a Clinical Study in Patients With Glioblastoma(Pamukkale University, 2022) Güner, E.; Bayatlı, E.; Kızıldoğan, A.K.; Gökmen, D.; Yüksek, V.; Taşpınar, F.; Uğur, H.Ç.Purpose: The multidrug resistance 1 (MDR1) gene expression and its epigenetic status may be an important factor in the chemotherapeutic resistance of glioblastoma (GB). The aim of this study was to analyze the effect of the MDR1 promoter methylation status on GB tumor tissue related to patient survival, chemotherapy resistance, and recurrence of the disease. Materials and methods: Thirty-six patients underwent surgery for GB at the Neurosurgery Department of Ankara University School of Medicine. The patients' clinical information and the MDR1 methylation status of the tumor tissues were compared to determine the effects on patient survival, chemotherapy resistance, and tumor recurrence. Results: Patients with MDR1 methylated GB had statistically significantly (p<0.001) shorter survival times. Early recurrence was detected in 25% of the patients with unmethylated tumor tissues and in 39.3% with hemimethylated tumor tissues. Conclusion: Instead of using the standard chemotherapeutics in all the patients with GB, tissue-specific medications must be chosen while taking into consideration the epigenomic characteristics and expression status of the tumor because of the genetic heterogeneity of GB. It is crucial to determine the prognostic, predictive, or therapeutic genetic factors for preventing NTD. The formation of the extracellular matrix (ECM) plays an essential role in migrating neural crest cells. Matrix metalloproteinases (MMPs) play a significant role in cell migration in ECM organization. The role of expressions and activation of MMP in NTD is unknown. This study aimed to investigate the roles of MMP-1,-2, and 9 gene expressions as biomarkers for NTD. Peripheral blood samples and NTD tissues were collected from 40 newborn babies diagnosed with NTD, which were also divided into subgroups based on pathology, and peripheral blood samples from only 20 healthy babies were taken for control. After total RNA isolation from blood and tissues, MMP-1,-2,-9 gene expressions were analyzed by Quantitative Real-Time PCR (RT-PCR). There was no difference between the control group and the NTD group in terms of MMP expressions in blood samples (p>0.05). A statistically significantly higher MMP-1 expression was found in Meningocele and Myeloschisis than in Encephalocele (p=0.014). A significant difference was found between the tissue and blood samples of the Meningomyelocele patient group regarding MMP-9 expression (p=0.019). There was no significant relationship between Ca2+, B12, and Folate levels, NTD, and MMP genes expressions (p>0.05). All rights reserved.Article Serum Protein Fraction Profile in Sheep Infected With Sarcoptes Ovis(Ataturk Universitesi, 2020) Çetin, S.; Usta, A.; Ekici, P.; Dede, S.; Yüksek, V.Scabies is an ectoparasitic infestation that is rapidly transmitted to sheep of any age. The present study was conducted to identify the significance of serum protein fractions in the diagnosis and treatment of animals infected by Sarcoptes ovis. The study group included 1-4 years old 18 Akkaraman sheeps with excessive itching and molting and infected with natural Sarcoptes ovis scabies and the control group included 1-4 years old 18 healthy sheeps, a total of 36 sheeps were included in the present study. Serum protein fractions were determined with the cellulose-acetate electrophoresis method in collected blood samples. There were no significant differences between total protein and α-2 globulin serum protein concentrations in the study and control groups. Although α-1, β-globulin and albumin concentrations were statistically higher in the patient group, gamma globulin levels decreased in the patient group (P<0.05). While the percentage (%) g protein did not differ based on α-2 globulin when compared to control, α-1, beta globulin, albumin and A/G ratio statistically increased in the patient group when compared to the control (P<0.05). There was a significant decrease in gamma globulins in the patient group when compared to the control (P<0.01). It was concluded that monitoring the serum protein parameters might be significant in the follow-up and treatment of scabies in sheep. © 2020 Ataturk Universitesi. All rights reserved.
