Article Activity, Acute and Sub-Acute Toxicity and Safety Assesment of the Hydroalcholic Root Extract of Diplotaenia Turcica(Agricultural Research Communication Centre, 2018) Ozdek, U.; Basbugan, Y.; Yildirim, S.; Boga, M.; Firat, M.; Deger, Y.Diplotaenia turcica is widely utilized in conventional treatment in the east of Turkey. Due to the insufficient data on the safety profile, the acute and sub acute toxicity of Diplotaenia turcica was determined. Furthermore, total phenolic content, flavonoid content and antioxidant activity of the extract were determined. The acute toxicity of hydroalcoholic root extract of Diplotaenia turcica (HREDT) was evaluated in mice after administration of single oral dose at the 5000 mg/kg (acute model) and rat after 28 days orally administration at the dose range of 250, 500 and 1000 mg/kg (subacute model). According to result, the LD50 value of HREDT was found to be greater than 5000 ing/kg. In sub acute toxicity study, no statistically significant differences were observed in the values of hematological and pathological parameters in comparison with control group. It was determined that the glucose, triglyceride, cholesterol and LDL levels exerted a significant effect depending on the HREDT doses. The results from the present study found out that HREDT did not produce any toxic effects or deaths in animals for both single and chronic administration. 'kara Erik' Seed Extract in Streptozotocin Diabetic Rats(Scibulcom Ltd., 2017) Yildirim, B.A.; Kordali, S.; Yildirim, S.; Yildirim, F.; Ercisli, S.In this study was aimed to investigate effects on the levels of liver and renal tissue CAT, MDA, SOD, GPx and GSH activities of Vitis vinifera L. cv. 'Kara Erik' seed extract on diabetic rats. A total of 28 male rats were randomly divided to four groups containing of 7 rats per group. Control Group: Received only vehicle 0.5% Carboxymethyl cellulose (CMC), STZ-DM (Streptozotosin-Diabetus mellitus) Group: Streptozotocin 60 mg/kg/IP single dose, VVE (Vitis vinifera L. seed extract) Group: Vitis vinifera seed extract 100 mg/kg/20days/po, STZ-DM+VVE Group: STZ as 60 mg/kg/IP single dose + VVE100 mg/kg/20days/po. At the end of these processes, the animals were sacrificed, liver and renal tissue were excised by dissection and evaluated. Catalase (CAT), Malondialdehyde (MDA), Superoxide dismutase (SOD), Glutathione peroxidase (GPx) and Glutathione (GSH) were measured in the liver and renal homogenisates. VVE was shown that the activity of CAT, SOD and GSH activities increased and decreased levels of MDA in diabetic rats. As a result, the results showed that oral administration of VVE extract reduced the levels of lIPid peroxides and improved the antioxidant activity in liver and renal tissues in rats treated with V. vinifera seed extract as compared with the diabetic and control rats. The purpose of this study was to present findings of congenital colo-vesico-urethral fistula and atresia coli, rectal agenesis and atresia ani clinically, radiographically, macroscopically and histopathologically. According to the clinical examination; anus did not form, animal was dehydrated and not able to get to its feet and had abdominal tension. Besides, few amounts of unclear urine and meconium were present in preputium. By taking into consideration these findings, it was decided to make surgical operation on suspicion of fistula presence between gastrointestinal system and urinary bladder. The animal died during operation. Subsequent to necropsy, colon was swollen and observed at the cranial of pelvis ending bluntly. The blunt end of colon adhered to both urinary bladder and urethra. In order to diagnosis anomaly, samples were obtained from the region where fistulas were present. According to histopathological examination, fistula formation of colon separately to both urethra and urinary bladder was observed. Observing congenital of colo-vesico-urethral fistula, atresia coli, rectal agenesis and atresia ani all in one only in materials rare and may present a good sample for congenital anomaly cases. © 2017, Chartered Inst. of Building Services Engineers. The material of this study consisted of 30 Turkish Van Cats of different ages and genders with Feline Infectious Peritonitis that were definitely diagnosed by post-mortem examinations and immunohistochemistry. The control group consisted of 6 healthy Turkish Van Cats of different ages and genders that were brought for routine clinical examination. Cats in the study group had clinical findings such as loss of appetite, weight loss, high fever, persistent fever, jaundice, dehy-dration, vomiting, respiratory system symptoms, anemia, nervous findings, uveitis, and ascites. These cats were mon-itored and following the death, post-mortem examinations were performed and cases with a definitive diagnosis were included in the study. Among the cats consisting study group, while 25 had the dry form of the disease, 5 had wet form. According to the hematological results, there was a statistically significant reduction in platelet counts. The biochemi-cal results showed statistically significant alterations that creatinine, aspartate aminotransferase, alkaline phosphatase, creatine kinase myocardial band, homocysteine, and nitric oxide concentrations were higher than the control group. Besides albumin concentrations were lower and the albumin/globulin ratio was 0.53. As a result; this is the first detailed study in Turkish Van Cats with Feline Infectious Peritonitis that evaluated clinical, hematological, biochemical, and pathological findings. Furthermore, serum homocysteine and nitric oxide levels were evaluated for the first time in cats with vasculitis which is the most important complication of the disease. It is concluded that the evaluation of serum homocysteine and nitric oxide concentrations in Feline Infectious Peritonitis may assist the antemortem diagnosis of e disease.Conference Object Comparison of the Oxidant-Antioxidant Parameters and Sialic Acid Levels in Renal Transplant Patients and Peritoneal Dialysis Patients(Elsevier Science inc, 2011) Emre, H.; Keles, M.; Yildirim, S.; Uyanik, A.; Kara, F.; Tamer, F.; Aydinli, B.Objective. Several studies have shown that in end-stage renal failure, increased oxidative stress and reduced antioxidant system functions may play an important role in inflammation, atherosclerosis, and many other complications. Paraoxonase (PON) and arylesterase (ARE) are antioxidant and malondialdehyde (MDA) an oxidant agent. Increased sialic acid (SyA) levels in uremic patients is an acute-phase response showing tissue and organ damage. The aim of this study was to compare PON, ARE, MDA, and SyA among continuous ambulatory peritoneal dialysis (CAPD) and renal transplant patients. Methods. Twenty-six CAPD patients including 7 women and 19 men of overall mean age 40.5 +/- 10.66 years were included in this study. There were 28 preemptive transplant patients, namely 6 women and 22 men of overall mean age 36.5 +/- 10.97 years. CAPD was the initial sole replacement therapy. Serum PON, ATE, MDA, and SyA levels were measured at 13-15 months of treatment. Results. Higher PON (P = .015) and ARE (P = .002) and lower MDA (P = .002) and SyA (P = .000) values were determined among transplanted compared with CAPD patients. Conclusions. In the transplant group, antioxidant parameters were higher and oxidant parameters lower with less activation of SyA than in the CAPD group. We observed that renal transplantation had a constructive effect on the oxidant-antioxidant system and consequent tissue destruction, which were impaired in uremic subjects. This study showed that renal transplantation is a more appropriate replacement treatment than CAPD.Article Dabigatran Versus Warfarin in Atrial Fibrillation: Multicenter Experience in Turkey(Sage Publications inc, 2016) Aslan, Onur; Yaylali, Y. T.; Yildirim, S.; Yurtdas, M.; Senol, H.; Ugur-Yildiz, M.; Ozdemir, M.Safety issues have been raised about dabigatran. We aimed to investigate the occurrence of safety outcomes in patients who had atrial fibrillation and a risk of stroke. We analyzed 439 patients prescribed dabigatran (n = 220) or warfarin (n = 219). Ischemic stroke occurred in 15 (6.8%) patients in the warfarin group versus 5 (5.2%) patients in the 110-mg group versus 1 (0.8%) patient in the 150-mg dabigatran group (P = .015). Intracranial hemorrhage occurred in 6 (2.7%) patients in the warfarin group versus 3 (2.4%) patients in the 150-mg dabigatran group (P = .104). Death from any cause occurred in 10 (4.6%) patients in the warfarin group versus 1 (1.0%) patient in the 110-mg dabigatran group (P = .005). Dabigatran was associated with less ischemic stroke and death from any cause than warfarin. Dabigatran may be a better option for stroke prophylaxis, where recommended monitoring with warfarin is suboptimal.Article Diagnostic Importance of Syndecan-1(cd138) in Chronic Endometritis of Dogs(Ataturk Universitesi, 2021) Yildirim, S.; İLhan, F.; Dörtbudak, M.B.; Cetin, N.In this study, the role of Syndecan-1 in the diagnosis of chronic endometritis, which is an important health problem for dogs and mostly causes infertility, was investigated. In addition, comparisons of Syndecan-1 and HE staining methods in the diagnosis of chronic endometritis were made. Generally, the prognosis is not good because the diagnosis is difficult in chronic endometritis, where no clinical symptoms can be observed. For this, early diagnosis with a reliable method in chronic forms of endometritis is very important. The presence of plasmocytes in the histopathological examination, which is one of the various diagnostic methods of endometritis, is a distinctive feature for the chronic form of the inflammatory character. Plasmocytes, which can be detected by histopathological staining methods can also be detected by immunohistochemical staining of the Syndecan-1 receptors that specific to plasmocytes at the same time. For this study, a sufficient number of female dog uterus was obtained in and around Van province. Hematoxylin-eosin (HE) staining of uterine tissues for histopathology and Syndecan-1 staining for immunohistochemical examination were performed. In the examinations made, 18 of 57 uterus samples with HE staining; With Syndecan-1, 21 of them were diagnosed with chronic endometritis. In the result of working; With Syndecan-1 staining, it was observed that plasmocytes could be selected more easily and clearly than HE. In addition, it was determined that Syndecan-1 is a reliable and practical method in the diagnosis of chronic endometritis with this study, which was performed for the first time in dogs. © 2021 Ataturk Universitesi. R.Article The Effect of Platelet Rich Plasma on Wound Healing in the Repair of Experimental Penile Fractures(Agricultural Research Communication Centre, 2022) Duz, E.; Beger, B.; Saglam, K.; Yildirim, S.Background: Penile fractures are caused by the rupture of the tunica albuginea and the underlying layers of corpus cavernosum as a result of the exposure of the erect penis to blunt trauma. Although conservative treatment methods have been recommended in former studies, recent studies favor early surgical options. Platelet-rich plasma (PRP) is a blood product rich in cytokines and several growth factors, obtained from the patient's own blood commonly used to stimulate healing in acute and chronic wound treatments. The efficiency of homologous PRP treatment in rats with penile fractures have been assessed in terms of clinical manifestation and histopathological findings in the present study. Methods: Thirty-five rats were randomly divided into five groups, each containing seven rats as follows: 1. Conservative group: Following the formation of penile fracture, medical interventions were not initiated on fracture area and the wound was left for secondary healing. 2. Bandage group: Following the formation of penile fracture, self-adherent elastic bandage (3 M Coban) was applied to the fracture area for three days. 3. Primary repair group: Following the formation of a penile fracture, the lacerated area was repaired with 7/0 vicryl primary sutures. 4. Primary repair + Homologous Platelet Rich Plasma group: Before the operation, 10 cc of blood was taken from one rat under general anesthesia by intracardiac puncture and PRP material was prepared from this blood. Homologous PRP was topically administered on the wound in the fracture area and then the penile fracture was repaired with 7/0 vicryl as simple separate stitches. 5. Non-operative control group: Surgical procedures were not applied to this group; it was used as the control group. Result: The effect of the PRP treatments on the surgical repair options in the rats with penile fractures was investigated. The present findings revealed that following the repair with conservative treatments, the fracture format persisted and various complications like infection, oedema, excessive fibrosis and hyperemia were encountered. On the other hand, the best healing was achieved in primary repair + PRP group in terms of clinical manifestation and histopathological findings.Article Effect of Pomegranate (Punica Granatum L.) Juice on Kidney, Liver, Heart and Testis Histopathological Changes, and the Tissues Lipid Peroxidation and Antioxidant Status in Lead Acetate-Treated Rats(C M B Assoc, 2017) Aksu, D. S.; Saglam, Y. S.; Yildirim, S.; Aksu, T.Pomegranate juice (PJ) contains relevant amounts of active biological compounds which alleviate the detrimental effects of chronic heavy metal exposure. This study investigated the protective potential of PJ against lead-induced oxidative stress. A total of forty adult male Sprague Dawley rats were divided into four experimental groups. The animals were fed a standard pellet diet and tap water ad libitum. The rats were divided into four groups (n=10 for each group): control, lead asetat (2000 ppm), low-treated PJ- a daily dose of 2.000 ppm lead plus 30 mu l pomegranate juice (included 1.050 mu mol total polyphenols, gallic acid equivalent), and high-treated PJ- a daily dose of 2.000 ppm lead plus 60 mu l pomegranate juice (included 2.100 mu mol total polyphenols, gallic acid equivalent). The treatments were delivered for 5 weeks. After the treatment period, the tissues samples (kidney, liver, heart and testis) were collected. Tissue lead (Pb) and mineral amounts (copper, zinc, and iron), tissues lipid peroxidation level and antioxidant status, and tissues histopathological changes were determined. The results showed that the highest rate lead loading was in the kidney and the testis. Pomegranate juice was decreased the lead levels of soft tissues examined; increased Zn amounts in tissues of which the lead accumulation was higher (kidney and the testis); decreased the copper, zinc and the iron levels of the liver and heart tissues, without creating a weakness in antioxidant capacity of these tissues, restricted the oxidative stress by decreasing lipid peroxidation, improved both of the activities of antioxidant enzymes such as superoxide dismutase (SOD) and catalaz (CAT), and the level of glutathione (GSH) in all the tissues examined in lead-treated groups. As histopathological findings, the cellular damage induced by lead in the tissues of the kidney, liver and the heart were observed to have been partially prevented by PJ treatment. The protective effect of PJ was more pronounced in the testis compared to those others.Article Effects of Butylated Hydroxytoluene on Blood Liver Enzymes and Liver Glutathione and Glutathione-Dependent Enzymes in Rats(Trakia University, 2018) Mean, S.; Değer, Y.; Yildirim, S.This study was aimed to detect the effect of butylated hydroxytoluene (BHT) on the liver glutathione level and glutathione-dependent enzyme activities in female Wistar albino rats. BHT was administered by oral gavage at a dose of 250 mg/kg (Group I) and 500 mg/kg (Group II) for 28 days, 1000 mg/kg (Group III) and 1500 mg/kg (Group IV) for 4 days. The serum ALT, AST and LDH activities were measured on an autoanalyzer, and liver gluthathione (GSH), gluthathione peroxidase (GPx), gluthathione S-transferase (GST), and gluthathione reductase (GR) activities were analysed with commercial ELISA kits. The ALT activity was significantly higher in Groups III and IV (P<0.05 and P<0.001, respectively) compared to the control group. Blood AST and LDH activities were significantly increased in Group IV (P<0.05). The GSH, GPx, GST and GR in the liver tissue were determined to be statistically low in Groups II, III and IV (P<0.001) in comparison with control group. In microscopic examination, BHT caused histopathological changes in the rat liver tissue in Groups II, III and IV depending on the dose and duration of exposure. It can be concluded that BHT plays a role in producing liver damage in rats with depressed hepatic antioxidant defense. The hepatotoxic response seemed to be dose-and time-dependent. © 2018, Trakia University. All rights reserved.Article Effects of Chronically Exposure To Flor and 7,12-Dimethylbenzanthracene (Dmba) on Spermatogenesis and Testicular Histopathology in Rats(Ataturk Universitesi, 2018) Yildirim, S.; Belhan, S.; Uyar, H.; Huyut, Z.; Oto, G.; Sağlam, Y.S.This study aimed to investigate the effects of both sodium fluoride (NaF) and polycyclic aromatic hydrocarbon (PAH) derivative such as 7,12-dimethylbenz[a]anthracene (DMBA) singly and in combination on spermatogenesis and testis histopathology in rats. Forty male Wistar albino rats weighing 250–300 g used. On the 90th day of the experiment, intracardiac blood samples and testes were taken. While there was no change in FSH and LH levels in the groups, testosterone levels were significantly decreased in the other groups according to the control group (P<0.001). It was decreased the ratio of motility of sperm, sperm density and, increased the abnormal sperm rate by chronic fluorosis and DMBA toxicity in rats significantly. In histopathological examinations; A decrease in the number of spermatocytes and spermatozoa in the tubulus wall of the testes of rats in the NaF (30 ppm) group was observed. When testicular tissues of rats in the DMBA (10 mg/kg) group were examined, atrophy in tubulus, edema in intertubular spaces and a decrease in the number of spermatocytes in the tubulus wall were found. When testes tissues of rats in the group of NaF (30 ppm) + DMBA (10 mg/kg) were examined, atrophy in seminiferous tubules, severe degenerative and necrotic changes in Spermatogoniums on the walls of tubules and thinning in tubule walls were observed. There is not found spermatozoon in the tubul lumens. Severe edema in the intertubular spaces, dilatation and hyperemia in the veins found in these regions. As a result, it was observed that NaF and DMBA had a negative effect on testicular tissue and fertility singly, and in combination, this effect was further exacerbated. © 2018 Ataturk Universitesi.All Rights Reserved.Article The Effects of Curcumin Nanoparticles on an Experimental Diabetes Model: Reproductive Hormones, Lipid Profile and Immunohistochemical and Immunofluorescence Markers(B W K Publishing Solutions & verlag, 2024) Belhan, S.; Huyut, Z.; Yildirim, S.; Algul, S.Diabetes affects many organs and causes serious problems for reproductive health. There is still no effective treatment but a range of new strategies are being used to treat and to reduce the possible complications of diabetes. One of the most promising of them relies on the use of nanoparticles. We have evaluated the effects of curcumin and curcumin nanoparticles on blood glucose, lipid profile, reproductive hormones and histopathological, immunohistochemical and immunofluorescent parameters in experimental diabetic conditions. We created eight study groups (control group, diabetes group, curcumin group, Mg-curcumin nanoparticle group, diabetes+curcumin group, diabetes+Mg-curcumin nano- particle group, encapsulated Mg-curcumin nanoparticle group, diabetes+encapsulated Mg-curcumin nano- particle group), allocating rats to the groups at random. Triglyceride, cholesterol and LDL levels were high in the diabetes group, NOP10 and PI3K levels were intense and reproductive hormone levels were low. Curcumin and curcumin nanoformulations administered to the diabetes groups led to significant improvements in these parameters. The testosterone level, which was low in the diabetes group, increased significantly after the application of encapsulated Mg-curcumin. Curcumin nanoformulations were more effective than curcumin at countering the diabetes-impaired lipid profile and improving the levels of reproductive hormones, the histopathological findings and PI3K and NOP10 expression.Article Histopathological and Immunohistochemical Study of the Effect of Sildenafil Citrate, Vitamin A, Vitamin C and Vitamin E on Wound Healing in Alloxan-Induced Diabetic Rats(Univ West indies Faculty Medical Sciences, 2021) Tas, A.; Karasu, A.; Yener, Z.; Yildirim, S.; Atasoy, N.; Duz, E.; Yavuz, E.Objective: To examine the impact of sildenafil citrate, vitamin A (Vit A), vitamin C (Vt C) and vitamin E (Vit E) on wound healing in a rat model of diabetes (n = 100). Methods: Diabetes was induced in the rats by intraperitoneal (i.p.) administration of alloxan. After anaesthesia, a standard wound was created on the back of each rat using a 10-mm sterile punch. The rats were randomly divided into 10 groups (n = 10 in each), as follows: normal saline, glibenclamide, insulin, sildenafil, Vit A, Vit C, Vit E, Vit A + sildenafil, Vit C + sildenafil, Vit E + sildenafil daily for 15 days. The rats were sacrificed after being anaesthetized 3, 7 and 15 days later Wounded skin tissue samples were collected for histopathological and immunohistochemical analyses. Results: On the 7th day, epithelial regeneration was completed in groups 8 and 9. Angiogenesis was insufficient in group 2. In terms of connective tissue proliferation, partially matured connective tissue was observed in group 4. On the 15th day of the study, groups 8, 9 and 10 had mature connective tissue. However; group 1 still had exudate-containing neutrophils. Immunohistochemically, on the 3rd day, the level of inducible nitric oxide synthase (iNOS) reactivity was intense in macrophages and neutrophils surrounding the wound edges in groups 4, 8, 9 and 10. The level of iNOS reactivity was moderate in group 6 and less distinct in groups 1, 2, 3, 5 and 7. Conclusion: Sildenafil citrate, together with Vit A and Vit C, is beneficial in wound healing of diabetic rats.
