Article Antidiabetic Effect of Fig Seed Oil in Rats With Diabetes Induced by Streptozotocin(Japan Oil Chemists Soc, 2024) Mert, Handan; Mert, Nihan; Cibuk, Salih; Yildirim, Serkan; Mert, NihatThe anti -diabetic effect of Ficus carica (Fig) seed oil was investigated. 4 groups with 6 rats in each group were used in the experiment as control, diabetes (45 mg/kg streptozotocin), fig seed oil (FSO) (6 mL/ kg/day/rat by gavage) and diabetes+FSO groups. Glucose, urea, creatinine, ALT, AST, GSH, AOPP and MDA analyses were done. Pancreatic tissues were examined histopathologically. When fig seed oil was given to the diabetic group, the blood glucose level decreased. In the diabetes+FSO group, serum urea, creatinine, AOPP, MDA levels and ALT and AST activities decreased statistically significantly compared to the diabetes group, while GSH levels increased significantly, histopathological, immunohistochemical, and immunofluorescent improvements were observed. It has been shown for the first time that FSO has positive effects on blood glucose level and pancreatic health. It can be said that the protective effect of fig seed oil on tissues may be due to its antioxidant activity.Article Antioxidant Properties of Ferulago Angulata and Its Hepatoprotective Effect Against N-Nitrosodimethylamine Oxidative Stress in Rats(Taylor & Francis Ltd, 2017) Kiziltas, Hatice; Ekin, Suat; Bayramoglu, Mahire; Akbas, Esvet; Oto, Gokhan; Yildirim, Serkan; Ozgokce, FevziContext: Ferulago angulata (Schlecht.) Boiss. (Apiaceae) (FASB) is used to treat liver diseases and has been used both as food and therapeutics by many cultures for thousands of years because of the natural antioxidant compounds. Objective: This study determines antioxidant properties of FASB flowers, the levels of minerals and vitamins, and also, evaluates the hepatoprotective effect of flowers against N-nitrosodimethylamine (NDMA) induced on liver tissue by assessing antioxidant enzymes and histopathological parameters in Wistar albino rats. Materials and methods: In the study, the rats were divided into six groups of ten. Control, untreated animals were given 0.9% NaCl. Rats were intraperitoneally given NDMA (10 mg/kg) for the first 7 days. FASB methanol extract (150 and 300 mg/kg) was administered orally for 21 days. Results: a-Tocopherol, retinol, ascorbic acid, total antioxidant activity, phenolic and flavonoid contents of FASB were 0.70 +/- 0.13, 0.29 +/- 0.03 mu g/g, 139.32 +/- 7.06 mu g/100 g, 171.61 +/- 6.05 mM ascorbic acid/g, 90.47 +/- 4.11 mg GA/g and 37.39 +/- 2.85 mg QE/g. DPPH and hydroxyl radical scavenging activity was obtained IC 50 67.34 +/- 4.14 and 64.87 +/- 4.68 mu g/mL, respectively. Discussion and conclusion: The results of the study indicated that FASB flowers contain high levels of vitamins, minerals, total antioxidant activity, phenolics and flavonoids. Due to the positive effect on significant changes in antioxidant enzymes of liver tissue and histopathological examination, it is thought that the plant could be used as a hepatoprotective.Article Bazı Fosfodiesteraz 5 İnhibitörlerinin Ovariektomize Sıçanların Karaciğer Dokusunda Oksidatif Stres, Vegf, Bmp 2 ve 9 Üzerine Etkileri(2023) Alp, Hamit Hakan; Huyut, Zübeyir; Cihan, Murat; Şekeroğlu, Mehmet Ramazan; Alyar, Gülşah; Yildirim, Serkan; Akbay, Halil İbrahimAmaç: Osteoporoz önemli bir sağlık sorunudur ve henüz etkili bir tedavisi yoktur. Fosfodiesteraz 5 inhibitörleri osteoporoz tedavisi için umut verici ajanlardır. Bu çalışmada, fosfodiesteraz 5 inhibitörlerinin (vardenafil, tadalafil ve udenafil) ovariektomi ile osteoporoz oluşturulan sıçanların karaciğer dokusunda kemik morfojenik protein-2 ve 9 (BMP-2 ve 9), vasküler endotelyal büyüme faktörü (VEGF) ve oksidatif stres belirteçleri (malondialdehit ve CoQ10) üzerindeki etkilerini belirlemeyi amaçladık. Gereç ve Yöntem: 50 Albino wistar dişi sıçan her grupta 10 sıçan olacak şekilde rastgele 5 gruba ayrıldı. Gruplar sırasıyla sham-operated, ovariectomise (OVEX), OVEX + Tadalafil, OVEX + udenafil ve OVEX + vardenafil idi. VEGF, BMP-2 ve 9 seviyeleri ELISA kitleri ile ölçülmüştür. MDA ve CoQ10 seviyelerini tespit etmek için yüksek basınçlı sıvı kromatografi yöntemi kullanılmıştır. Bulgular: PDE-5 inhibitörleri uygulanan gruplarda VEGF, BMP-2 ve 9 seviyeleri sham ve OVEX gruplarına göre anlamlı derecede yüksekti. OVEX+vardenafil ve OVEX+udenafil grupları arasında VEGF, BMP-2 ve 9 düzeyleri açısından anlamlı bir fark bulunmamıştır. PDE5 inhibitörü uygulanan gruplarda MDA ve CoQ10 düzeyleri OVEX grubuna göre anlamlı derecede düşüktü. Histolojik ve immünohistokimyasal sonuçlar incelendiğinde, PDE-5 inhibitörü gruplarında anjiyogenezin anlamlı derecede yüksek olduğu görüldü. Sonuç: Sonuç olarak, bu inhibitörlerin karaciğer dokusunda VEGF, BMP-2 ve 9 ekspresyonunu indükleyerek kemik mineralizasyonu ve yeniden şekillenmesi üzerinde olumlu etkileri olabileceğini söyleyebiliriz.Article Beneficial Influence of Topical Extra Virgin Olive Oil Application on an Experimental Model of Penile Fracture in Rats(Sage Publications inc, 2015) Gunes, Mustafa; Ozkol, Halil; Pirincci, Necip; Gecit, Ilhan; Bilici, Salim; Yildirim, SerkanPenile fracture (PF) is known as a traumatic rupture of the tunica albuginea of corpus cavernosum. In this study, we aimed to investigate the healing influence of topical extra virgin olive oil (EVOO) on PF through evaluating levels of some oxidative stress biomarkers for the first time. Histopathological evaluation was also realized. A total of 18 male Sprague-Dawley albino rats were divided into three groups of six rats each as control group, in PF (alone) group, and PF + EVOO group. Experimental PF was formed via incising from the proximal dorsal side of the penis in the rats of all groups except control. While in PF (alone) group, fracture was formed and the incision was primarily closed, in PF + EVOO group in addition to foregoing processes, EVOO was also administrated topically twice a day for 3 weeks. At the end of the experiment, all rats were killed and penectomy was carried out. While malondialdehyde, myeloperoxidase, lipid hyroperoxide, and total oxidant status significantly (p < 0.05) increased, reduced glutathione and total free sulfhydryl groups markedly (p < 0.05) decreased in PF (alone) group when compared with PF + EVOO group. Levels of these parameters were reversed to nearly normal values by topical EVOO application. Protection by EVOO is further substantiated via the improved histological findings in PF + EVOO group as against degenerative changes in the rats of PF (alone) group. Our data revealed that EVOO has protective effect in penile cavernosal tissue through probably its antioxidant, free radical defusing, anti-inflammatory, and antimicrobial effects.Article Bir Güvercinin (Oriental Roller) Göz Kapağında Tüberküloz Olgusu(2021) Eser, Gizem; Saglam, Yavuz Selım; Yildirim, Serkan; Alkan, IsmaılKanatlı tüberkülozu Mycobacterium avium spp. tarafından oluşturulan pek çok evcil ve yabani kanatlılarda rastlanılan kronik seyirli bir hastalıktır. Bu çalışmanın materyalini Van Yüzüncü Yıl Üniversitesi Cerrahi kliniğine getirilen gözkapağında bir kitle (1,5 x 1,5 x 0,5 cm. ölçülerinde ve 0,88 gr ağırlığında) bulunan üç yaşında Oriental Roller ırkı bir güvercin oluşturdu. Göz kapağı ve çevresinde bulunan bu kitle anestezi altında cerrahi operasyonla total olarak alındı. Alınan kitle sitolojik ve histopatolojik olarak incelendi. Sitolojik incelemede çok sayıda langhans dev hücresi gözlendi. Histopatolojik muayenede ise; merkezinde nekroz, çevresinde çok çekirdekli langhans dev hücreleri, mononükleer hücre infiltrasyonları ve en dıştan fibröz kapsül ile çevrili tüberküllerin varlığı gözlendi. Alınan kesitlerin Ziehl-Neelsen boyama ile boyanmasında, nekrotik kitle içerisinde yoğun asit-fast basili tespit edilerek tüberküloz tanısı konuldu. Tanı sonucu hayvan sahibinin isteği üzerine ötenazi yapıldı. Nekropside iç organlarda herhangi bir patolojik bulguya rastlanmadı. Bu olgu Van yöresinde güvercinlerde bildirilen ilk avian tüberküloz vakasıdır. Van ili göçmen kuşların önemli göç yolları ve uğrak bölgelerinden biri olduğu için bölgede hastalığın görülmesi evcil ve yabani hayvanlar arasında hastalığın yayılmasında önemli bir risk faktörüdür. Bu nedenle konuyla ilgili daha fazla çalışmaların yapılması önerilmektedir.Article Bir Montofon Melezi Süt İneğinde Kutanöz Aktinobasilloz Olgusu(2018) Karasu, Abdullah; Kayıkcı, Caner; Yildirim, Serkan; Kuşcu, Yağmur; Sancak, TunahanBu çalışmada, montofon melezi bir inekte gözlenen kutanöz aktinobasilloz olgusunun klinik ve histopatolojik bulgularıile sağaltım sonuçlarının değerlendirilmesi amaçlandı. Anamnez bilgisinde, hayvanın sol açlık çukurluğu ile arkaekstremitelerin medial yüzlerinde değişik boyutlarda kitleler şekillendiği ve giderek büyüdüğü ifade edildi. Yapılan klinikmuayenede, sol açlık çukurluğunda yumurta büyüklüğünde bir adet, sol arka ekstremitenin medial yüzünde cevizbüyüklüğünde 2 adet ve sağ arka ekstremitenin medial yüzünde fındık büyüklüğünde 5 adet kutanöz granülomatöz kitlelergözlendi. Sol açlık çukurluğunda ve sol arka ekstremitede yer alan kitleler total olarak ekstirpe edildi ve sağ arka bacağınmedial yüzeyindeki kitlelere cerrahi müdahale yapılmadı. Ekstirpe edilen kitlelerin histopatolojik inceleme sonucunda kronikpiyogranülomatöz yangının sebebi olarak Actinobacillus-benzeri bakteri olduğu tespit edildi. Postoperatif oral sodyumiyodür ile birlikte parenteral prokain penisilin ve dihidrostreptomisin sülfat kombinasyonu uygulandı. Sonuç olarakActinobacillus lignieresii tarafından oluşturulan pyogranülamatöz kutanöz lezyonlar sığırlarda oldukça nadir şekillendiği içinderide şekillenen pyogranülamatöz lezyonların ayırıcı tanısında aktinobasillozun göz ardı edilmemesinin gerektiğidüşünülmektedir. Ayrıca lezyonların tedavisinde, cerrahi yöntemin yanı sıra sodyum iyodür ile birlikte uzun süre antibiyotikuygulaması ile tedavide başarılı sonuçlar alınabileceği kanısına varıldı.Article Can Zaprinast and Avanafil Induce the Levels of Angiogenesis, Bone Morphogenic Protein 2, 4 and 7 in Kidney of Ovariectomised Rats(Taylor & Francis Ltd, 2022) Huyut, Zubeyir; Bakan, Nuri; Yildirim, Serkan; Akbay, Halil Ibrahim; Huyut, Mehmet Tahir; Ahlatci, Adem; Ucar, BunyaminObjective: This study investigated effects of zaprinast and avanafil on angiogenesis, vascular endothelial growth factor (VEGF), bone morphogenic protein (BMP) 2, 4 and 7. Methods: Female rats were randomly divided into four groups (n = 6). Sham; abdomen was approximately 2 cm opened and closed. Ovariectomised (OVX); abdomen was opened 2 cm and the ovaries were cut. OVX + zaprinast and OVX + avanafil groups; after the same procedure with OVX, 10 mg/kg zaprinast and avanafil were orally administered for 2 month, respectively. Angiogenesis and the levels of VEGF, BMP2, 4 and 7 were determined. Results: VEGF, BMP2, 4 and 7 levels in OVX + zaprinast and especially OVX + avanafil groups were higher than the sham and OVX (p < .05). However, only VEGF and BMP2 levels in OVX + zaprinast group were significant according to sham (p < .05). Also, angiogenesis in OVX + zaprinast and OVX + avanafil groups was dominant according to sham and OVX (p < .05). Conclusions: Zaprinast and avanafil induced BMP2, 4 and 7 levels synergistically with increased VEGF and angiogenesis in renal tissue.Article Clinical Coenurosis (Coenurus Cerebralis) and Associated Pathological Findings in a Calf(Univ Agriculture, Fac veterinary Science, 2011) Ozkan, Cumali; Yildirim, Serkan; Kaya, AbdullahThis study aims to investigate clinical and pathological findings of a clinical Coenurus cerebralis case in a 10-month-old Simmental male calf. Clinical examination of the calf revealed incoordination, irregular gait, failure to hold the head straight, leftward head tilt, and circling. The animal was diagnosed with C. cerebralis and euthanazia was recommended. The autopsy demonstrated a cyst (9x7 cm) in the caudal of the left cerebral hemisphere within the cranium. The cyst caused compression over the ventral portion of the left cerebral hemisphere, while a marked perforation of 3-4 cm diameter was found on the sphenoid bone. Histopathologically, hyperemia and perivascular mononuclear cell infiltration were observed. In conclusion, we found it beneficial to present the clinical and pathological findings of this calf infected with C. cerebralis which is known to be a rare clinical entity among cattle. The purpose of this study was to present findings of congenital colo-vesico-urethral fistula and atresia coli, rectal agenesis and atresia ani clinically, radiographically, macroscopically and histopathologically. According to the clinical examination; anus did not form, animal was dehydrated and not able to get to its feet and had abdominal tension. Besides, few amounts of unclear urine and meconium were present in preputium. By taking into consideration these findings, it was decided to make surgical operation on suspicion of fistula presence between gastrointestinal system and urinary bladder. The animal died during operation. Subsequent to necropsy, colon was swollen and observed at the cranial of pelvis ending bluntly. The blunt end of colon adhered to both urinary bladder and urethra. In order to diagnosis anomaly, samples were obtained from the region where fistulas were present. According to histopathological examination, fistula formation of colon separately to both urethra and urinary bladder was observed. Observing congenital of colo-vesico-urethral fistula, atresia coli, rectal agenesis and atresia ani all in one only in materials rare and may present a good sample for congenital anomaly cases.Article Detection of Se, Vit. E, Se, vit. A, malondialdehyde (MDA), 8-hydroxy-2-deoxyguanosine (8-OHdG), and ubiquinone-10 (CoQ10) levels and histopathological changes in sheep with white muscle disease (WMD). A total of 30 sheep were used; 20 sheep with WMD were brought to our clinic for diagnosis and treatment at various times, and 10 healthy sheep were in the control group. The Se, vit. E, vit. A, MDA, 8-OHdG, and CoQ10 values of the healthy and WMD sheep were as follows: 0.917 +/- 0.037, 0.790 +/- 0.067; 1.190 +/- 0.011, 1.090 +/- 0.021; 5.400 +/- 0.275, 5.200 +/- 0.173; 1.602 +/- 0.264, 2.636 +/- 0.576; 0.656 +/- 0.197, 1.485 +/- 0.271; and 0.280 +/- 0.044, 1.753 +/- 0.551 respectively (p<0.05). According to histopathological and immunohistochemical findings in the WMD group, hyaline degeneration, Zenker's necrosis, and dystrophic calcification were observed in the muscle fibers. Immunohistochemically, 8-OHdG staining of the heart tissue determined a severe 8-OHdG expression in the WMD group. The findings of this study suggest that MDA, 8-OHdG, and CoQ10 values could be used as diagnostic and prognostic biomarkers in sheep affected with WMD.Article Determination of Selected Hormones and Biochemical Parameters in Turkish Van Cats With Tail Gland Hyperplasia(Polish Soc veterinary Sciences Editorial office, 2020) Ozkan, Cumali; Kayar, Abdullah; Kaya, Abdullah; Yildirim, Serkan; Iskefli, Onur; Kozat, Suleyman; Akgul, YakupThe present study was performed on Turkish Van cats with tail gland hyperplasia in order to determine changes in some hormone levels and biochemical parameters. Animal material consisted of 18 male Van cats of between 2-8 years of age, 12 with tail gland hyperplasia and 6 unaffected cats as the control group. Following routine clinical examination, hormone levels and biochemical parameters were measured by obtaining blood samples, and biopsy samples were obtained from the region and examined under a light microscope. When compared with the control group, testosterone levels were high although the difference between groups was not statistically significant. Similarly, no statistical significance was found between the groups in terms of other hormones. Among the hormonal parameters, the only remarkable increase was seen in the testosterone level. Among biochemical parameters, the alkaline phosphatase (ALP), aspartate amino transferase (AST), cholesterol and triglyceride levels were statistically different, while other biochemical parameters were not statistically significant. While AST and cholesterol results were increased, ALP and triglyceride levels were decreased. As a result, we conclude that the disease is an important problem which occurs extensively in mature male Van cats, particularly during and after mating periods. We also conclude that the disease might be associated with an increase in testosterone levels. Presenting this study is beneficial as there are limited reports about this disease in cats. This is the first study presenting tail gland hyperplasia with some hormonal levels and biochemical parameters in male Van cats.Article Do Avanafil and Zaprinast Exert Positive Effects on Bone Tissue Via the Nitric Oxide/Cyclic Guanosine Monophosphate/Protein Kinase-G Signaling Pathway in Rats With Ovariectomy-Induced Osteoporosis(Univ Sao Paulo, Conjunto Quimicas, 2021) Huyut, Zubeyir; Bakan, Nuri; Yildirim, SerkanPhosphodiesterase-5 inhibitors (PDE-5Is) exert positive effects on bone healing and mineralization by activation the nitric oxide/cyclic guanosine monophosphate/protein kinase-G (NO/cGMP/PKG) signaling pathway. In this study, the effects of zaprinast and avanafil, two PDE-5Is, on the NO signaling pathway, estrogen levels, selected bone formation and destruction marker levels, whole-body bone mineral density (WB-BMD), right femur trabecular bone thickness (RF-TBT) and epiphyseal bone width, angiogenesis in the bone-marrow, and selected oxidative stress parameter levels were investigated in rats with ovariectomy-induced osteoporosis. Twenty four adult rats (8 months old) were equally divided into four groups. The first group was the sham operated group. Groups 2, 3 and 4 included ovariectomized rats. At six months after ovariectomy, the 3rd and 4th groups were administered 10 mg/kg zaprinast and avanafil daily as a single dose for 60 days, respectively. Increases in the activity of the NO/cGMP/PKG signalling-pathway, C-terminal collagen peptide levels, angiogenesis in the bone marrow, RF-TBT, epiphy seal bone width and WB-BMD were observed compared to the ovariectomized positive control group (OVX), while the pyridinoline and deoxypyridinoline levels were decreased in the OVX+zaprinast and OVX+avanafil groups (p<0.05). The malondialdehyde, ubiquinone10/ubiquino110 and 8-hydroxy-2-deoxyguanosine/10(6) deoxyguanos ine levels were also increased in the ovariectomized groups compared to the sham group (p<0.05). Based on these results, the levels of bone atrophy and some markers of oxidative stress were increased due to acute estrogen deficiency induced by ovariectomy, but zaprinast and avanafil administration significantly prevented these changes.Article Effect of Acute and Chronic Fluoride Administration on Bone Histopathology, Bone Fluoride Accumulation, and Locomotor Activity in an Animal Model of Paleopathological Fluorosis(int Soc Fluoride Research, 2020) Turkekul, Ramazan; Arihan, Seda Karaoz; Yildirim, Serkan; Arihan, Okan; Oto, Gokhan; Ekin, Suat; Yildiz, DamlaFluorosis may be identified in skeletal materials from ancient civilizations by macroscopic signs in teeth and bone and hard tissue fluoride levels. In the present study, human teeth, femoral, and rib specimens from the Van Fortress excavation, in Turkey, were examined for the presence of fluorosis. In addition, an animal study in rats was conducted as a model of human fluorosis, by examining the effects of fluoride administration, in various doses and for different durations, on weight loss, locomotor activity, fluoride accumulation, and deformation in bone and teeth. Fifty-six adult male Wistar albino rats, weighing 150-200 g, were divided into 7 different groups of 8 rats. Four acute groups were treated with 0 (control), 5, 15, and 50 mg/L of fluoride in drinking water for 7 days and three chronic groups were treated with 5, 15, and 50 mg/L of fluoride for 90 days. The results of the analysis of the human samples from the Van Fortress excavation showed that none of the dental, femoral, or rib samples had a fluoride content that was significantly greater than that of the surrounding soil. The results of the rat study showed that no significant differences between the groups were found in body weight on days 1, 30, 60, and 90. The rotarod locomotor test showed a significant (p<0.05) dose- and time-dependent reduction in locomotor activity as a result of the fluoride administration in the 50 mg/L chronic fluoride group compared to the control, 5 mg/L acute fluoride, 15 mg/L acute fluoride, and 5 mg/L chronic fluoride groups. Significant fluoride accumulation was found in the femoral neck (cortical tissue), the femoral head (trabecular tissue), and in rib bone. Light microscopy showed a severe thinning of the epiphyseal growth plate and bone trabeculae in the femoral bone tissue. We concluded that femoral bone (cortical and trabecular parts) and ribs are good sites for assessing the effects of fluoride exposure in animal models of human fluorosis.Article Effect of Chronic Exposure To Sodium Fluoride and 7,12-Dimethylbenz[a]anthracene on Some Blood Parameters and Hepatic, Renal, and Cardiac Histopathology in Rats(int Soc Fluoride Research, 2018) Yildirim, Serkan; Ekin, Suat; Huyut, Zubeyir; Oto, Gokhan; Comba, Arzu; Uyar, Hasan; Cinar, D. AliThis study aimed to investigate the effects of both sodium fluoride (NaF) and 7,12-dimethylbenz[a]anthracene (DMBA), both separately and in combination, on some blood parameters and hepatic, renal, and cardiac histopathology in rats. Forty male Wistar albino rats, weighing 250-300 g, were randomly divided into one control and three experimental groups (i) a NaF group who received 15 ppm of NaF in their drinking water for 90 days, (ii) a DMBA group who received 10 mg DMBA/kg body weight/po/ weekly for 90 days, and (iii) a NaF+DMBA group who received 15 ppm NaF in their drinking water plus 10 mg DMBA/kg bw/po/weekly for 90 days. The animals in the groups were sacrificed at the end of the 90 days. The AST, ALT, LDH, CK, creatinine, troponin I, and MDA levels increased in the NaF, DMBA, and NaF+DMBA groups compared to the control group, while the WBC, K, Na, Cl, urea, SOD, GSH-Px, CAT, and GSH values showed a statistically significant decrease (p<0.05). In addition, the CK-MB significantly increased in the DMBA and NaF+DMBA groups compared to the control group (p<0.05). The histological structure of the liver, kidney, and heart tissues in the control group was normal. In the NaF and DMBA groups, degenerative and necrotic changes were detected. In the NaF+DMBA group: (i) the liver exhibited hydropic degeneration and coagulation necrosis in hepatocytes, severe dilation in the sinusoids, congestion in the central and portal regions, and mononuclear cell infiltration in the portal region; (ii) the kidneys displayed congestion in the glomerulus and interstitial vessels, interstitial nephritis, diffuse hydropic degeneration, and coagulation necrosis in the tubule epithelium; (iii) the heart showed myocardial hyperemia, severe mononuclear cell infiltration in interstitial tissue, hyaline degeneration, and Zenker's necrosis in myocardium As a result of these blood and oxidative stress parameters and histopathological findings, it was determined that NaF, DMBA, and NaF+DMBA induce toxicity in the liver, kidney, and heart tissues and thus play an important role in the physiopathology of toxicity.Article Effect of Chrysin on Methotrexate-Induced Testicular Damage in Rats(Wiley, 2019) Belhan, Saadet; Comakli, Selim; Kucukler, Sefa; Gulyuz, Fetih; Yildirim, Serkan; Yener, ZabitThis study was conducted on 28 male Wistar albino rats to determine the effects of chrysin on methotrexate-induced damage to testicular tissue. Rats were grouped into four groups of seven rats reach: Group 1 (n = 7) was the control group to which no drugs were administered; this group was only provided with food and water. Group 2 (n = 7) was administered 20 mg/kg of methotrexate once intraperitoneally. Group 3 (n = 7) was administered 50 mg/kg of chrysin for 7 days orally. Group 4 (n = 7) was administered 20 mg/kg of methotrexate once intraperitoneally, followed by oral administration of 50 mg/kg of chrysin for 7 days. At the end of the experiment, rats were anaesthetised, rat testes were removed, and spermatozoon was obtained from the cauda epididymis. It was determined that sperm count and motility, glutathione peroxidase, superoxide dismutase and catalase activities decreased in the methotrexate group, whereas malondialdehyde, tumour necrosis factor-alpha, interleukin-1 beta and nuclear kappa factor B expression levels increased. Furthermore, damage to tubulus seminiferus structures and affusion in germ cells was identified. In the methotrexate + chrysin administered group, sperm count improved, biochemical enzyme levels increased, and structural improvements were observed in testicular tubules. These findings demonstrated that chrysin plays a protective role in testicular damage in rats.Article The Effect of Diplotaenia Turcica Root Extract in Streptozotocin-Induced Diabetic Rats(Walter de Gruyter Gmbh, 2020) Ozdek, Ugur; Yildirim, Serkan; Deger, YeterBackground: Diplotaenia turcica has been used traditionally to diabetes treatment. In this study, the effects of D. turcica root extract (DT) on diabetes mellitus induced by streptozotocin (STZ) were investigated. Materials and methods: In this study, 78 male rats were used, rats were divided into 9 groups randomly. In diabetic groups, STZ was given a single dose of 45 mg/kg by intraperitoneally. DT (50,100 and 200 mg/kg) and glibenclamide (5 mg/kg) were given by orally. Blood and pancreas tissue samples were taken for biochemical and pathological tests. Results: It was found that glucose levels decreased, and insulin levels increased in the treatment groups compared with the diabetes group. In addition, only in 200 mg/kg DT dose group was found to decrease HbA1c levels. Pancreatic tissue analysis showed that MDA levels decreased and GSH levels and CAT, SOD, GSH-Px and GSH-R activities increased in diabetic rats treated with DT. Histopathological and immunohistochemical examinations of the pancreas showed significant improvements in the treatment with DT. Conclusion: These results clearly show the antioxidant property of DT. The rats were divided into 4 groups: control, evening primrose oil, fructose, fructose + evening primrose oil. Body weight, daily feed and water consumptions and systolic blood pressures of animals were measured. At the end of trial, blood samples were taken, livers were excised and histopathological examination was performed. Glucose, uric acid, triglyceride, T.cholesterol, LDL, HDL, VLDL, ALT, AST, ALP, LDH, adiponectin, insulin, IL-6, TNF-alpha, TAC, and TOS levels were analysed. Some analysed parameters and systolic blood pressure of fructose + evening primrose oil group decreased significantly compared to fructose group and adiponectin, TAC, and HDL levels were significantly increased. As conclusion, evening primrose oil can be considered as antioxidant agent by reducing oxidative stress, increasing adiponectin levels and insulin sensitivity, anti-inflammatory properties, exhibiting anti-atherogenic effect by regulating dyslipidemia and systolic blood pressure.Article The Effect of Fucoidan on Changes of Some Biochemical Parameters in Nephrotoxicity Induced by Gentamicin in Rats(Ankara Univ Press, 2018) Ataman, Nese; Mert, Handan; Yildirim, Serkan; Mert, NihatFucoidan is a polysaccharide with high viscosity and mucilage, which contains significant proportion of sulphate ester group and L-fucose. It is present in intercellular spaces of brown algae. This study aimed to investigate the effect of fucoidan on some biochemical parameters and kidney tissues in nephrotoxicity induced by GM in rats. The rats used in the study were randomly divided into 4 groups. Each group had 7 rats as control, fucoidan, GM and GM+fucoidan. Blood samples were taken after 24 hours from the end of experiment which lasted eight days. Creatinine, BUN, uric acid, glucose, triglycerides, total cholesterol, VLDL, HDL, total bilirubin levels and ALT, AST, ALP, LDH, CK, amylase activities were assayed by an autoanalyser. The kidney tissues were examined histopathologically. In GM+fucoidan group, creatinine (p<0.001), BUN (p<0.001), uric acid (p<0.05), triglycerides (p<0.05), VLDL (p<0.05), AST (p<0.001), ALP (p<0.05), LDH (p<0.001), CK (p<0.01) levels statistically decreased, however HDL (p<0.05) level increased when compared to GM group. As conclusion, it can be said that administering fucoidan during GM toxicity decreases the nephrotoxicity damage. Considering biochemical parameters and histopathological findings, fucoidan can be suggested as a protective agent for kidneys in nephrotoxicity induced rats.Article Effect of Low- and High-Dose Methotrexate on Wound Healing in Rats(Acta Cirurgica Brasileira, 2025) Karasu, Abdullah; Kuscu, Yagmur; Kayikci, Caner; Yildirim, Serkan; Kuscu, Oguzhan; Kiliclioglu, MetinPurpose: To investigate the effect of intraperitoneal treatment with low- and high-dose methotrexate (MTX) on wound healing in rats. Methods: The study sample consisted of 54 healthy rats. Under aseptic conditions, skin wounds were created with two circular fullthickness punch tools, 10 mm in diameter, one on the right and the other one on the left of the dorsal vertebral line. The rats were randomly assigned to one of three main treatment groups. On the 0th day (2 hours before wound creation), 7th day, and 14th day, the control group received 0.3-mL saline, the low-MTX group received 3 mg/kg MTX, and the high-MTX group received 30 mg/kg MTX, all administered intraperitoneally. The wounds were evaluated seven, 14, and 21 days after injury through morphometrical, biochemical, histopathological, and immunohistochemical analyses. Results: MTX dose-dependently decreased the degree of inflammation and angiogenesis, tissue hydroxyproline level, and HSP70 and tumor necrosis factor-alpha expression in the early phase of wound healing. It also suppressed epithelialization and collagen 1 expression throughout the wound-healing process. Conclusion: The wounds treated with high-dose of MTX had statistically delayed wound closure on days 7, 14 and 21 compared to the saline group, while wounds treated with low-dose of MTX only had statistically delayed wound closure on day 14. In addition, weight loss was observed in rats treated with high-dose MTX, which was thought to reflect its toxicity. The dose-dependent adverse effect of MTX on wound healing may be due to its antiproliferative, antifibrotic, anti-inflammatory, and antiangiogenic effects.Article The Effect of Pde5 Inhibitors on Bone and Oxidative Damage in Ovariectomy-Induced Osteoporosis(Sage Publications Ltd, 2017) Alp, Hamit H.; Huyut, Zubeyir; Yildirim, Serkan; Basbugan, Yildiray; Ediz, Levent; Sekeroglu, Mehmet R.Osteoporosis is a major public health problem associated with many factors, and it affects more than 50% of women over 50 years old. In the current study, our purpose was to investigate the effects of phosphodiestarase-5 inhibitors on osteoporosis via the nitric oxide/3',5'-cyclic guanosine monophosphate/protein kinase G signalling pathway. A total of 50 female albino Wistar rats were separated into five groups. The first group was appointed as the healthy control group with no ovariectomy. All animals in the other groups underwent a bilateral ovariectomy. Six months after the ovariectomy, vardenafil, udenafil and tadalafil were given to the third, fourth and fifth groups, respectively, but were not administered to the positive control group (10 mg/kg per day for two months). The bone mineral density values were determined using a densitometry apparatus for all groups pre- and post-ovariectomy as well as after treatment. The levels of nitric oxide, endothelial nitric oxidesynthase, asymmetric dimethylarginine, 3',5'-cyclic guanosine monophosphate, protein kinase G, phosphodiestarase-5, pyridinoline, deoxypyridinoline, carboxyterminal telopeptide fragments and plasma carboxy terminal propeptide of type I collagen were determined using an enzyme linked immunosorbent assay. The levels of malondialdehyde, 8-hydroxy-2-deoxy guanosine, deoxyguanosine and coenzyme Q10 were determined by a high-performance liquid chromatography assay. Additionally, the right femoral trabecular bone density and the epiphyseal plate were measured in all groups. Angiogenesis was histologically observed in the bone tissue. In addition, we determined that the inhibitors may have caused a positive impact on the increased bone mass density and reduction of bone resorption markers. We also observed the positive effects of these inhibitors on oxidative stress. In conclusion, these phosphodiestarase-5 inhibitors increase angiogenesis in bone tissue and improve the re-formation rate of bone in rats with osteoporosis. Impact statement The results in our study appear to establish the osteoporosis model and provide evidence of the positive effects of three separate PDE5 inhibitors (vardenafil, udenafil, and tadalafil). The positive effects of these PDE5 inhibitors are investigated and demonstrated by the bone mass density and bone resorption markers. These effects are associated with significant demonstrated antioxidant activities. Osteoporosis is a significant major public health problem especially in more aged populations. Advances in identifying and understanding new potential therapeutic modalities for this disease are significant. This study provides such an advance.
