Gökkuşağı Alabalıklarından Elde Edilen Lactococcus garvieae İzolatlarının Moleküler Karakterizasyonu

Abstract
Bu çalışma, Van ilinde faaliyet gösteren altı farklı gökkuşağı alabalığı (Oncorhynchus mykiss) üretim çiftliğinden izole edilen Lactococcus garvieae suşlarının mikrobiyolojik ve moleküler düzeyde karakterizasyonunu amaçlamaktadır. Seçilen çiftlikler bölgeye balık temin eden ve anaç balık üretimi yapan, yıllık periyodik üretim gerçekleştiren önemli üretim merkezleridir. Çalışmada, her çiftlikten klinik sem 6 adet balık örneği alınmış, böbrek dokularından TSA besiyerine ekimler yapılmıştır. Üreyen kolonilerden morfolojik ve biyokimyasal analizler (Gram boyama, katalaz, oksidaz, hareketlilik) gerçekleştirilmiş ve şüpheli kolonilerden DNA izolasyonu yapılmıştır. Real-Time PCR ile 16S rRNA bölgesi üzerinden L. garvieae tespiti sağlanmıştır. Pozitif örneklerde antibiyotik direnç genleri (TetA, FloR) ile virulans genleri (Hemoliz, Adhesive Pav) analiz edilmiştir. Sonuçlar, çiftlikler arasında genetik çeşitlilik farkları bulunduğunu ve özellikle antibiyotik direnç genlerinin yaygın olduğunu göstermiştir. Bu durum, bölgesel antibiyotik kullanım baskısına ve patojenitenin yayılımına işaret etmektedir. Çalışmanın çıktıları doğrultusunda, çiftlik bazlı filtrasyon sistemlerinin kurulması ve bakteriyel fauna bilgilerinin anonim olarak paylaşılması gibi öneriler sunulmuştur.
This study aims to characterize Lactococcus garvieae strains isolated from six rainbow trout (Oncorhynchus mykiss) farms in Van province, Turkey, using microbiological and molecular techniques. The selected farms are significant for regional aquaculture, conducting annual production cycles from larvae to market-size fish and serving as broodstock sources. From each farm, six symptomatic fish were sampled. Kidney tissues were cultured on TSA medium, and the resulting colonies were subjected to Gram staining, catalase, oxidase, and motility tests. Colonies showing similar characteristics to L. garvieae underwent DNA extraction and Real-Time PCR targeting the 16S rRNA gene. Positive isolates were further examined for antibiotic resistance genes (TetA, FloR) and virulence genes (Hemolysin, Adhesive Pav). The results indicated substantial genetic variability among farms, with widespread detection of resistance genes, highlighting the selective pressure from antibiotic usage. Based on these findings, recommendations included implementing filtration systems tailored to each facility and promoting anonymous sharing of bacterial disease profiles to improve regional fish health management.
