Development of a Pvc Membrane Potentiometric Sensor for the Determination of Benzydamine Hydrochloride
Abstract
Benzidamin hidroklorür, anestezik ve analjezik özelliklere sahip, non-steroidal antiinflamatuar ilaç (NSAID) sınıfı bir ilaç etken maddesidir. Lokal olarak etki gösterir. Yumuşak doku zedelenmesine bağlı akut (primer) inflamasyonlarda, inflamasyonun lokal mekanizmalarını etkileyerek antiinflamatuvar, antiödem ve analjezik olarak, ağız ve boğazdaki inflamasyon sebebi ile ortaya çıkan semptomların tedavisinde kullanılır (Turnbull, 1995). Benzidamin, doz aşımı durumunda güçlü bir psikoaktif halüsinojendir. Benzidamin, bazı insanlarda ağızda batma, uyuşukluk, ani gelişen nefes darlığı, ciltte döküntü, yüzde ve/veya dilde şişme, aşırı duyarlılık reaksiyonları, alerjik reaksiyonlar, ağızda geçici his azalması ve yanma hissi, bulantı, kusma, öğürme, tahrişe bağlı cilt reaksiyonları, tükürük bezinde geçici şişme, ses tellerinde geçici kasılma, solunum yollarında geçici kasılma, döküntü ile birlikte görülen kaşıntı, ürtiker (döküntü), ışığa duyarlılık, ağızda meydana gelen döküntü gibi yan etkiler görülebilir. Bu nedenle benzidamin hidroklorürün tayini önemlidir. Bu çalışmada benzidamin hidroklorür tayini için doğrusal çalışma aralığı geniş, kısa cevap zamanına sahip olan benzidamin hidroklorür seçici PVC membran potansiyometrik sensör geliştirildi. Optimum membran bileşimi olarak %1 KTpClPB, %67 NPOE, %32 PVC bileşimi belirlendi. Bu sensör için doğrusal çalışma aralığı 1.0x10-2 mol L-1-1.0x10-8 mol L-1, 10 katlık konsantrasyon değişimindeki eğimi 56.1 mV; tayin sınırı 5.0x10-9 mol L-1;cevap zamanı 10 saniye olarak belirlendi. pH çalışma aralığı pH 3.18-7.58 olarak belirlendi. Geliştirilen sensör farmasötik preparatlarda başarıyla uygulandı.

It is used in acute (primary) inflammations due to soft tissue injury, as an anti-inflammatory, anti-edema and analgesic by affecting the local mechanisms of inflammation, and in the treatment of symptoms caused by inflammation in the mouth and throat (Turnbull, 1995). Benzydamine is a powerful psychoactive hallucinogen in case of overdose. In some people, benzydamine may be observed side effects such as stinging in the mouth, drowsiness, sudden shortness of breath, rash on the skin, swelling of the face and/or tongue, hypersensitivity reactions, allergic reactions, temporary decreased sensation and burning sensation in the mouth, nausea, vomiting, retching, skin reactions due to irritation, salivation, temporary swelling of the salivary gland, temporary contraction of the vocal cords, temporary contraction of the respiratory tract, itching accompanied by rash, urticaria (rash), sensitivity to light, and rash in the mouth Therefore, the determination of benzydamine hydrochloride is important. In this study, a benzydamine selective PVC membrane potentiometric sensor with a wide linear operating range and short response time was developed for the determination of benzydamine hydrochloride. The optimum membrane composition was determined as 1% KTpClPB, 67% NPOE, 32% PVC. For this sensor, linear range, slope, detection limit and response time were determined as 1.0x10-2 mol L-1 -1.0x10-8 mol L-1, 56.1 mV/decade, 5.0x10-9 mol L-1 and 10 seconds. The pH working range was determined as pH 3.18-7.58. The developed sensor has been successfully applied in pharmaceutical preparations.
