The Lethal Dose Levels of Volatile Oil Extract of Petroselinum Crispum Seeds and the Investigation of Antiinflammatory Effect on Laboratory Animals

Abstract
Bu tez çalışmasında Petroselinum crispum (PC) tohumu uçucu yağ özütünün gaz kromatografik yöntemle tanısı ve analizi, median letal dozu (LD50) ve antienflamatuvar aktivite yönünden incelenmesi amaçlandı. LD50 için 36 adet fare, antienflamatuvar etki çalışması için 48 adet rat kullanıldı. PC tohumu uçucu yağının fitokiyasal analizinde ?-pinene %43.31, ß-pinene % 39.9, ß-phellandrene % 10.18, 1-Allyl-2-3-4-5-tetra methoxy-benzene %5.47, myricticin % 4.89, apiole %2.71, sabinene %1.18, elemicin %0.40 oranında saptandı. LD50 (ip) dozu 0,563 ml/kg olarak bulundu. Antienflamatuvar etki deneylerinde PC tohumu uçucu yağı uygulanan grup ile SF (serum fizyolojik), etil alkol ve indometazin uygulanan diğer her bir grup karşılaştırıldı.Antienflamatuvar etki deneyinde, PC tohumları 0,060 ml/kg dozlarında yüksek düzeyde, 0,015 ml/kg ve 0,030 ml/kg dozlarında ise indometazin'e göre zayıf bir etki gösterdi.PC uçucu yağının median efektif dozu (ED50) yaklaşık 0.029 ml/kg olarak bulunduAnahtar sözcükler: Petroselinum crispum tohumu, uçucu yağ, antienflamatuvar aktivite, letal doz, fare, rat.
The aim of this thesis was do gas chromatographic analysis of extract of volatile oil of Petroselinum crispum (PC) seeds and to investigate its median lethal dose (LD50) and possible antiinflammatory effects. For this aim, 36 mice were used in LD50 study and 48 rats were used for antiinflammatory effect in study. The phytochemical analysis of PC seeds volaite oil showed that PC seeds containing % 43.31 of ?-pinene, % 39.9 of ß-pinene, % 10.18 of ß-phellandrene, % 5.47 of 1-Allyl-2-3-4-5-tetra methoxy-benzene, % 4.89 of myricticin, % 2.75 of apiole, % 1.18 of sabinene and % 0.40 of elemicin.The LD50 (ip) dose was found to be 0,563 ml/kg. In the antiinflammatory experiments, the group exposured to PC was compared with the each of groups exposured to SF(serum physiologic), ethyl alcohol and indomethasine.
