Isolation and Diagnosis of Pseudomanas Savastanoi Pv. Savastanoi, and Determination of the Prevalence and Insidance of Olive Knot Disease Caused by It at Mardin Province

Abstract

Bu çalışmada; zeytinin yetiştiriciliğinin çok eski tarihlere dayandığı ve yerel çeşitleri barındıran Mardin ilinde Pseudomonas savastonoi pv. savastanoi'nin sebep olduğu Zeytin Dal Kanseri hastalığının şiddeti, oranı ve yaygınlığının belirlenmesi, ayrıca patojenin izolasyonu, biyokimyasal ve moleküler tanılanmasının yapılmasını kapsamaktadır. Mardin ili Nusaybin, Derik, Kızıltepe, Artuklu, Midyat, Savur ve Yeşilli ilçelerinde yapılan sörvey çalışmalarından elde edilen izolatların biyokimyasal tanılamasında Levan oluşumu, Oksidaz testi, IAA testi kullanılmıştır. İzolatların maleküler tanılaması indol asetitk asit bölgesine spesifik IAAF/F primerleri ile 16s rRNA bölgelerinin dizi analizleri ile yapıldı. İki yaşındaki Zeytin fideleri ile de patojenisite çalışmaları gerçekleştirilildi. Çalışma kapsamında 47 bahçe incelendi ve elde edilen izolatlardan 28'inin Ps pv. savastanoi olduğu tespit edildi. Midyat ilçesinde hastalık tespit edilemedi. Derik ilçesinde hastalığın yaygınlığının %86.66 düzeyinde olduğu belirlendi. Hastalık oranınında yine Derik ilçesi 3.54 ile ilk sırayı alırken, bunu takiben 3.38 ile Nusaybin ilçesi geldi. Hastalığın il geneli yaygınlığının %57.34, hastalık oranının ise %1.49 olduğu tespit edildi. Yetiştirilme bölgesi (ilçe) ile Hastalık şiddeti arasında bir ilişki olduğu tespit edildi. Ayrıca hastalık şiddetinin zeytin çeşitlerine ve pestisit uygulamasına göre değişiklik gösterdiği, fakat gübreleme ve sulama metodunun etkisinin olmadığı tespit edildi.

The aim of this study is to determine the severity, incidence, and prevalence of olive knot caused by Pseudomonas savastonoi pv savastanoi in the province of Mardin, where olive cultivation dates back to ancient times and has local varieties. It is also the isolation, biochemical and molecular diagnosis of the pathogen. The levan formation, Oxidase test, Gelatin test, and IAA test were used for biochemical identification of isolates obtained from survey studies in Mardin province Nusaybin, Derik, Kızıltepe, Artuklu, Midyat, Savur, and Yeşilli districts. Molecular identification of the isolates was performed with IAAF/F primers specific to the Indole Acetyltic Acid region and sequence analyses of the 16s rDNA regions. Pathogenicity studies were also carried out with two-year-old olive seedlings. Within the scope of the study, 47 orchards were examined and 28 of the obtained isolates were Ps pv savastanoi found to be. No disease has been detected in the Midyat district. It was determined that the prevalence of the disease in Derik district was 86.66%. In the rate of disease, Derik took first place with 3.54, followed by Nusaybin with 3.38. It was determined that the province-wide prevalence of the disease was 57.34% and the disease rate was 1.49%. It has been determined that there is a relationship between the cultivation area (district) and the severity of the disease. In addition, it was stated that the severity of the disease varied according to the olive varieties and pesticide application, but the fertilization and irrigation method had no effect.
