Isolation, Characterisation and Identification of Amycolatopsis and Streptomyces Spp. Isolated From Soil
Abstract
Çalı?manın amacı Van Gölü havzası Erçek Gölü çevresinde farklı lokalitelerdenalınan 30 toprak numunesinden Actinomycetales takımı içersinde bulunanAmycolatopsis ve Streptomyces cinslerine ait bakterilerini izole etmek, te?his etmek vekarakterize etmektir. Toprak numunelerinin bazı fiziko-kimyasal analizleri yapıldı.Selektif besi ortamı kullanılarak izole edilen ve Safla?tırılan 60 Amycolatopsis ve 60Streptomyces izolatlarının renk gruplandırılması yapıldıktan sonra nümerik taksonomikanalizi, ayrıca safla?tırılan tüm Streptomyces test izolatları bilgisayar yardımı ile fenetikveri içeren (?DENTAX) Bakteriyel te?his programı kullanılarak te?hisleri yapıldı.Moleküler karakterizasyon çalı?maları için renk gruplarını temsilen seçilen toplam 10Amycolatopsis ve 10 Streptomyces test izolatlarının tüm hücre protein profilleri SDSPAGE(Sodyum Dodesil Sülfat Poliakrilamid Jel Elektroforezi) ile belirlenerek nümerikanalizi yapıldı ve sonuçlar dendogram ?eklinde sunuldu.Toprak numunelerinin pH'ları genelde nötral değerde iken Organik maddeiçeriği %0,494 ila 14,944 arasında deği?ti, nem oranları ise %10'un altında ölçüldü.?zolasyon çalı?masında Amycolatopsis koloni sayımı 0,3x104 ile 12,4x104 cfu/gr kurutoprak arasında deği?irken Streptomyces koloni sayımı 2,4x104 ile 33,4x104 cfu/gr kurutoprak arasında deği?ti. Safla?tırılan Amycolatopsis test izolatları 11 Streptomyces testizolatları ise 12 renk grubuna ayrıldı. 60 Streptomyces test izolatlarından 42 tanesininte?hisi yapıldı. SDS-PAGE ve RAPD-PCR teknikleri kullanılarak yapılankarakterizasyon çalı?malarında test organizmaları aynı gruplarda yer aldı.Anahtar kelimeler: Amycolatopsis, Streptomyces, izolasyon, karakterisazyon, RAPD

Numeric taxonomic analyses have been performed aftercolour grouping of insulated, purified 60 Amycolatopsis and 60 Streptomyces isolates byusing selective nutrition environment, also the diagnoses of all the purifiedStreptomyces test isolates have been made with computer help by using Bacterialdiagnosis programme including fenetic data (IDENTAX). The numeric analyses oftotally 10 Amycolatopsis and 10 Streptomyces test isolates, that have been chosen forrepresenting colour groups for molecular characterization studies have been performed,by determining all the cell protein profiles of them with SDS-PAGE (Sodium DodesilSulfate Poliacrilamide Jell Electrophoresis), and the results have been presented asdendograms.When the pHs of the soil samples have been usually in neutral values, theorganic material content has changed between %0,494 and 14,944, and the humidity hasbeen measured under %10. In the insulation study, Amycolatopsis colony numbers havechanged 0,3x104 and 12,4x104 cfu/gr between dry soil and Streptomyces colonynumbers have changed 2,4x104and 33,4x104 cfu/gr between dry soil. PurifiedAmycolatopsis test isolates have been sorted into 11 colour groups and Streptomycestest isolates have been sorted into 12 colour groups. 42 of 60 Streptomyces test isolateshave been diagnosed.
