Effects of Omethoate, Pestisicides, on Various Tissues Antioxidant Defence Systems and Lipid Peroxidation Levels in Rana Ridibunda
Abstract
Bu çalışmada, laboratuar şartlarında omethoat adlı pestisite maruz bırakılan su kurbağalarının (Rana ridibunda) karaciğer, beyin, böbrek ve kalp dokularındaki, lipid peroksidasyon (MDA) ve glutatyon (GSH) düzeyi ile antioksidan enzim aktiviteleri süperoksit dismutaz (SOD), glutatyon peroksidaz (GSH-Px), glutatyon -S- transferaz (GST) aktivitesi üzerine etkilerinin incelenmesi amaçlanmıştır. Altmış su kurbağası (Rana ridibunda) üç deney grubuna ayrıldı. Çalışmalar sırasında kurbağalar omethoatın 10 ve 20 ppm'lik dozlarına 24, 48, 72 ve 96 saatlik sürelerle maruz bırakıldılar. Bu sürelerin sonunda kurbağalardan alınan dokuların GST, SOD ve GSH-Px aktiviteleri ile MDA ve GSH düzeylerinin dokulardaki analizleri, spektrofotometrik yöntemle belirlendi. Omethoate'ın farklı süre ve konsantrasyon etkisine maruz bırakılan kurbağalarda GSH düzeyi azalırken, MDA düzeyi artmış ve antioksidan savunma sistemi üzerine farklı etkilere neden olduğu gözlenmiştir.
The study was carried out to investigate of effects of omethoate, pesticide, on antioxidant enzymes activities superoxide dismutase (SOD), glutathione peroxidase (GSH-Px), glutathione -S- transferase (GST), lipid peroxidation (MDA) content and reduced glutathione (GSH) levels in liver, brain, kidney and heart tissues of frogs (Rana ridibunda) exposed to the chemical under laboratory conditions. 60 frogs (Rana ridibunda) were divided into three experimental groups. At the time of studies frogs were exposed to 10 and 20 ppm doses of omethoate for 24, 48, 72 and 96 hours. In the end of this time, the analysis on various tissues of MDA content, GSH levels and GST, SOD and GSH-Px activities of tissues excised from frogs were determined by means of spectrophotometer. The tissues of frogs exposed to different period and concentration of the chemical were affected. According to the resuls, while GSH level decreased whereas MDA level increased. Also, antioxidant defence system enzymes were fluctuated.
