Hepatoprotective Effect of Dihydromyrcenol and Geranyl Formate in the Experimental Acute Liver Damage Induced by Carbon Tetrachloride
Abstract
Bu çalışmada Karbon tetraklorür (CCl4)'ün sıçan karaciğerinde yaptığı akut karaciğer hasarında hepatoprotektif etkisi bilinen Vitis vinifera L (VVL) bitkisinin ekstresinde bulunan Dihydromyrecenol (DM) ve Geranyl formate (GF)'nin hepatoprotektif etkileri araştırıldı.Çalışmada, Sprague-Dawley cinsi 54 adet sıçan kullanıldı. Sıçanlardan altışarlı dokuz adet grup oluşturuldu ve yedi günlük çalışma sonrası sakrifiye edildiler. Ardından DM ve GF'nin akut karaciğer hasarındaki etkileri, deneklerdeki kilo değişimi, serum Alanin aminotranseraz (ALT) ve Asparat aminotranseraz (AST) seviyeleri ve karaciğerdeki histopatolojik değişiklikler değerlendirilerek karşılaştırmalı olarak araştırıldı.Çalışmada CCl4'ün etkisine bağlı artan balon dejenerasyonu ve apoptotik hücre sayısının; DM ile anlamlı şekilde azaldığı ve GF'nin düşük dozları ile kısmen azaldığı ancak GF'nin yüksek dozu ile azalmadığı görüldü. Ayrıca ALT ve AST seviyelerindeki değişiklikler de bu bulgular ile benzerlik gösterdi.CCl4'e bağlı oluşturulan deneysel akut karaciğer hasarında, DM'nin hepatoprotektif etkisinin olduğu ve GF'nin ise düşük dozlarda kısmen hepatoprotektif etkisinin olduğu ancak yüksek dozlarda ise hepatoprotektif etkisinin olmadığı sonucuna varıldı.Anahtar Kelimeler: Akut karaciğer hasarı, Dihydromyrcenol, Geranyl formate, Hepatoprotektivite, Karbon tetraklorür, Vitis vinifera L.
In this study, the hepatoprotrective effects of Dihydromyrcenol (DM) and Geranyl formate (GF), which are substrates in the extract of Vitis vinifera L (VVL) plant, in the acute hepatic damage induced by Carbon tetrachloride (CCl4) in rats were investigated.54 Sprague-Dawley type rats were used in the study. The rats were classified to form nine groups, six rats in each, and they were sacrified after seven days study. Then, the effects of DM and GF on acute liver damage, body weight changes, serum Alanine aminotransferase (ALT) and Aspartate aminotransferase (AST) levels, and the histopathological changes in the liver were assessed and evaluated comparatively.In the study it was found that, baloon degeneration and apoptotic cells number which increase depending on CCl4 effect, decreased significantly by DM, and partially decreased with low doses of GF, but did not decrease with high doses of GF. Also the changes of serum ALT and AST levels were similar to these finging.It was concluded that DM has protective effect on experimental acute hepatic damage induced by CCl4, and GF has partial hepatoprotective effect in low doses, but it has not hepatoprotective effect in high doses.Key Words: Acute liver damage, Carbon tetrachloride, Dihydromyrcenol, Geranyl formate, Hepatoprotective, Vitis vinifera L.
