Abstract
Ekonomik açıdan en önemli ürünlerden biri olan Patates dördüncü ana gıda grubu olarak kabul edilir. Patates yumuşak çürüklüğü, Pectobacterium ve Dickeya türleri gibi çeşitli bakteriyel patojenler tarafından oluşturulur. Dünya çapında patates yumrularını etkileyen başlıca yıkıcı hastalıklardan biridir. Bu çalışma, Irak, Erbil bölgesinde patates yumuşak çürüklük hastalığına neden olan bakteriyel patojenleri geleneksel ve moleküler yöntemlerle belirlemeyi amaçlamaktadır. Örnekler Irak-Erbil ilinin çeşitli lokasyonlarındaki depo ve tarlalarda toplanmıştır. İzole edilen aday patojen bakteri izolatları ile biyokimyasal ve fizyolojik testler (KOH, oksidaz, katalaz, aşırı duyarlılık reaksiyonu, patojenite testi ve CVP ortamı) testler yapılmıştır. DNA izolasyonu yapılan patojen bakterilerin 16S rRNA bölgeleri PCR çalışması ile çoğaltılarak dizi analizi çalışması yapılmıştır. Çalışma sonucunda Erbil ilinde baskın yumuşak çürüklük patojeninin Pectobacterium carotovorum subsp. caratovorum olduğu bunu takiben Dickeya dianticola ve Dickeya solani'nin geldiği belirlenmiştir.
The potato crop is one of the most economically important crops and is considered the fourth main food crop in the world. Potato soft rot is one of the major destructive diseases affecting potato tubers worldwide caused by several bacterial species such as Pectobacterium and Dickeya species. This study aims to identify the causal agent of potato soft rot disease using conventional methods and molecular approaches and then create a phylogenetic tree for close related strains of bacteria. After the samples were collected in storage and fields in various locations of Erbil province-Iraq, then the bacteria were isolated and cultured for different biochemical and physiological tests, including (KOH, oxidase, catalase, hypersensitivity reaction, pathogenicity test, and CVP medium). The positive results were obtained. Then as a molecular approach, DNA was extracted from the bacterial isolates, and PCR was carried out by using a specific primer to amplify the 16S rRNA gene of pathogenic bacteria. So far, this is the first study where Pectobacterium carotovorum, Dickeya dianticola, and Dickeya solani were identified by using PCR and sequencing approaches in Erbil province-Iraq.
