Determination of in Vitro Plant Regeneration and Antibacterial Activity of Centaurea Depressa Bieb.

Abstract
Centaurea depressa Bieb. Türkiye florasında önemli bir tıbbi aromatik ve süs bitkisi olmasına rağmen, mikroçoğaltımı üzerinde çalışmalar yok denecek kadar azdır. Bu çalışmada, in vitro koşullarda çimlendirilmiş C. depressa tohumlarından gelişen in vitro bitkiciklerin; hipokotil, gövde, yaprak, kotiledon, boğum ve sap boğum organlarından mikroçoğaltım gerçekleştirilmiş ve bitki ekstraktlarının antibakteriyel aktiviteleri belirlenmiştir. Eksplantlar 0.20 mg/l BAP (6-Benzil amino pürin); 0.10, 0.15, 0.30, 0.45 ve 0.60 mg/l NAA (Naftalen asetik asit) içeren MS (Murashige ve Skoog) ortamda kültüre alınmıştır. Çalışma sonucunda 0.20 mg/l BAP ve 0.15 mg/l NAA içeren MS ortamında kotiledonlardan rejenerasyon elde edilmiş ve sürgünler farklı IBA (İndol-3-bütirik asit) konsantrasyonları içeren ortamlarda köklendirilerek dış koşullara alıştırılmıştır. Ek olarak, farklı 2,4-D (2,4-Diklorofenoksi asetik asit) konsantrasyonları içeren MS ortamında yaprak, aksiller tomurcuk ve kotiledon eksplantlarından somatik embriyogenez elde edilmiştir. En iyi somatik embriyogenez 0.6 mg/l 2,4-D içeren MS ortamında kaydedilmiştir. Antibakteriyel aktivite çalışmaları sonucunda ise C. depressa'nın gövde ve yaprak ekstraktlarının Staphylococcus aureus ATTCC25923, gram (+) Escherichia coli ATCC8739 ve Pseudomonas aeruginosa ATCC25823 gram (-) bakterilerine karşı belirgin düzeyde etkili olduğu gözlenmiştir.
Although, Centaurea depressa Bieb. is an important medicinal and ornamental plant in the flora of Turkey, there is few studies about its micropropagation. In current study, micropropagation was performed in the hypocotyl, stem, leaf, cotyledon, node of the in vitro plantlets grown from C. depressa seeds germinated in vitro, and antibacterial activities of plant extracts were determined. Explants were cultured in MS (Murashige and Skoog) medium with 0.20 mg/l BAP (6-Benzyl amino purine) containing 0.10, 0.15, 0.30, 0.45 and 0.60 mg/l NAA (Naphthalene acetic acid). As a result of the study, regeneration was obtained from cotyledons in MS medium contained 0.20 mg/l BAP and 0.15 mg/l NAA, and the shoots were rooted in the medium included different IBA (Indole-3-butyric acid) concentrations and acclimated to external conditions. In addition, somatic embryogenesis was obtained from explants of leaf, axillary bud and cotyledon in MS (Murashige ve Skoog) medium contained different concentrations of 2,4-D (2,4-Dichlorophenoxyacetic acid). The best somatic embryogenesis was recorded in MS medium contained 0.6 mg/l of 2,4-D. As a result of antibacterial activity studies, it was observed that the stem and leaf extracts of C. depressa were considerably effective against Staphylococcus aureus ATTCC25923, gram (+) Escherichia coli ATCC8739 and Pseudomonas aeruginosa ATCC25823 gram (-) bacteria.
