Effects of Some Agricultural Chemicals on Antioxidant Enzymes Activity and Glutathione, Lipid Peroxidation Levels in Gill, Liver and Muscle Tissues of Fishes
Abstract
Bu çalısmada, zirai mücadele ilaçlarından Parathion Metil ve Diazinonun yasam ortamlarına eklenmek suretiyle 24,48 ve 72 saatlik uygulamalarına laboratuar sartlarında maruz bırakılan Gök kusagı alabalıklarının solungaç, karaciger ve kas dolularındaki lipid peroksidasyon (MDA) ve glutatyon (GSH) düzeyi ile antioksidan enzim aktiviteleri süperoksit dismutaz (SOD), glutatyon peroksidaz (GSH-Px), glutatyon -S- transferaz (GST), glutatyon redüktaz (GR) üzerine etkilerinin incelenmesi amaçlanmıstır. Yetmisbes alabalık (Oncorhynchcus mykiss) bes deney grubuna ayrıldı. Bu gruplardan biri kontrol digerleri ise Parathion metil ve Diazinonun 24, 48 ve 72 saatlik dozlarına maruz bırakılan balıklardan olusturuldu. Deney süresince balıklar yasam ortamlarına eklenmek suretiyle Parathion metil ve Diazinon'un 0.5 ve 1 ppm'lik dozlarına 24, 48 ve 72 saatlik sürelerle maruz bırakıldılar. Bu sürelerin sonunda solungaç, karaciger ve kas dokuları kesilerek çıkartıldı. MDA, GSH, GST, GR, SOD ve GSH-Px düzeylerinin dokulardaki analizleri, spektrofotometrik yöntemle belirlendi. Farklı düzeylerde söz konusu kimyasalların etkisine bırakılan balıklarda MDA düzeyi artarken, GSH düzeyinde azalmalara ve antioksidan enzim aktiviteleri düzeyleri üzerine farklı etkilere neden oldukları gözlendi. The study was carried out to investigate whether treatment of agricultural pesticides could affect on lipid peroxidation (MDA), reduced glutathione (GSH) levels and antioxidant enzymes activity, süperokside dismutase (SOD), glutathione peroksidase (GSH-Px), glutathione -S- transferase (GST), glutathione reductase (GR) in various tissues of fish under laboratory conditions. Fishes (Oncorhynchcus mykiss) were divided into five experimental groups; control and pesticides-exposed for 24, 48 and 72 hours. 0,5 ppm and 1 ppm of diazinon and parathion methyl were administered to fishes for 24, 48 and 72 hours. Gill, liver and muscle tissues were excised at the same time. MDA, GSH, GST, GR, SOD and GSH-Px levels were determined on both tissues samples by means of spectrophotometer. Fishes exposed to different levels of agricultural pesticides indicated that while MDA level increased significantly, glutathione levels decreased and caused the different effects on the level of antioxidant enzymes activity of various tissues in fish.
