A Reserach on the Effects of Different Doses of Streptozotocin on Optic Nerve of Rat by Sterological Methods
Abstract
Optik sinir sağlıklı bir şekilde alınan uyarının beyne iletilmesi ve verilen cevabın da göze iletilmesinde sorumludur. Bu yüzden diyabette optik sinirin etkilenmesi ve bu işlevlerin yerine getirilememesi sonucu çok ciddi komplikasyonlara neden olduğu bildirilmektedir. Bu çalışma; farklı dozlarda uygulanan streptozotosin (STZ)'in sıçan optik sinirine etkilerini stereolojik metodlarla araştırmasıdır. Çalışmada 3?4 aylık, erişkin, ortalama 250g ağırlığında 20 adet erkek sıçan kullanıldı. Her grupta 5'er adet sıçan olmak üzere toplam 4 grup oluşturuldu. Deneyden önce grupların ağırlıkları ve kan glikoz değerleri ile deneyden sonra kan glikoz değerleri ve haftada bir olmak üzere dört hafta ağırlıkları ölçüldü. STZ 45mg/kg ve 65mg/kg intraperitonal olarak uygulanıp diyabet oluşturuldu. Toplam 4 hafta olan deney süresinin sonunda kontrol ve denek gruplarına ait tüm denekler, ketalar 50 mg/kg anestezisi altında açılarak perfüzyon uygulandı. Optik sinirin ışık mikroskobik incelemeleri için uygun takipleri yapıldı. Hacim için Masson trikrom ve sayım için crystal vıolet boyası uygulanan kesitler, Işık mikroskobu kullanılarak görüntüleri kaydedildi. Optik sinir dokusunun total hacmi için Cavalieri Prensibi, epinöryum oranları bileşen hacim oranları uygulaması, glia hücre sayıları için fiziksel Disektör-Cavalieri uygulaması kullanılarak her bir grup için ayrı hesaplandı. Elde edilen veriler istatiksel analizler için Kruskal Wallis, Tukey çoklu testleri kullanıldı. Kontrollere göre her iki diabet gruplarında optik sinir hacminde ve glia hücrelerinde anlamlı azalma gözlendi (P<0.05). Fakat epinöryum hacminde herhangi bir değişiklik gözlenmedi (P>0.05).
Optic nerve transmits the received stimulation to the brain in a healthy manner and is responsible for the transmission of the response to the eye. In diabetes, very serious complications occur due to affection of optic nerve and causes to failure of fulfilling the described function of optic nerve. This study aims to investigate the effects of different dose s of streptozocotin (STZ) by stereological methods. In this study, 20 male adult rats, 3-4 months of age weighing 250g were used. Four groups were made which have five rats in each group. Before the experiment weights and blood groups of the rats, after the experiment blood glucose values and weights were measured in four week for once a week. Diabetes is made by intraperitonael application of STZ 45mg/kg and 65mg/kg. After four week which was the period of the experiment, all rats were perfused following kethalar anesthesia (50 mg/kg). Optic nerve was prepared for light microscopic investigations. Sectons of the nerve were stained with Masson tricrome stain for voume and crystal vıolet for counts and photographed by light microscope. Total volume of the nerve tissue, rates of epineuria and number of glia cell were calculated by disector-cavalieri treated and principle of Cavalier and volume fraction for each groups. Obtained values were evaluated by statistical methods which were Kruskal Wallis and Tucay multiple test. There were significant decreases in the optic nerve volumes and glia cell in all groups (P<0.05) but not in the volume of the epineuria (P>0.05).
