Abstract
Çiftçi Yegin S. Deneysel Diyabet Oluşturulan Ratlarda Likopen Uygulamasının Lipoprotein, Paraoksonaz ve Sitokinler Üzerine Etkisi.Y.Y.Ü. Sağlık Bilimleri Enstitüsü, DoktoraTezi, Van, 2012. Bu araştırma,deneysel diyabet oluşturularak diyabetin tanı ve tedavisinde kullanılması öngörülen lipoprotein, paraoksonaz ve sitokinler üzerine likopen uygulamasının etkisini araştırmak amacıyla yapılmıştır. Bu çalışmada 7-8 haftalık, 250-300 gr ağırlıkları arasında 28 adet erkek Wistar-Albino rat kullanıldı ve rastgele her biri 7 rattan oluşan kontrol grubu, diyabet grubu, diyabet-likopen grubu ve likopen grubu olmak üzere dört gruba ayrıldı. Deneysel diyabet oluşturmak için 45 mg/kg düzeyinde soğuk sitrat tamponunda çözdürülmüş streptozotosin (STZ) intraperitonel yolla uygulandı. Likopen ise mısır özü yağında çözdürülerek 10 mg/kg/gün olarak likopen ve diyabet oluşturulmuş diyabet-likopen grubuna gavaj ile ağız yoluyla uygulandı. 1 aylık deneme süresi sonrasında eter anestezi altında ratların kalplerinden jelli tüplere kan örnekleri alındı. Alınan kan örnekleri santrifüj edilerek serum elde edildi. Serum örneklerinden HbA1c, Paraoksonaz aktivitesi, lipoprotein düzeyleri ve sitokinler tayin edildi. Analiz sonuçlarına göre, HbA1c düzeyi kontrol grubuna göre diyabet grubunda en yüksek seviyeye ulaşırken, diyabet-likopen grubunda kontrole yakın, likopen grubunda ise kontrolden biraz daha yüksek olduğu tespit edildi (P<0,001). Paraoksonaz aktivitesi kontrol grubuna göre, diyabetli grupta en düşük seviye gözlenirken, diyabet-likopen grubunda biraz yükseldiği, likopen grubunda ise önemli derecede bir yükselme oluştuğu saptandı (P<0,001). Trigliserit düzeyinde ise kontrol grubuna göre, bütün gruplarda anlamlı bir yükselme meydana geldi (P?0.001). Kolesterol düzeyinde kontrol grubuna göre diğer gruplarda bir düşüş gözlendi (P<0,05). VLDL düzeyinde kontrol grubuna göre, diğer gruplarda anlamlı bir yükselme gözlendi (P<0,05). HDL (P<0,001) ve LDL (P<0,05) düzeyinde de kontrol grubuna göre, diğer gruplarda anlamlı bir düşüş gözlendi. Tnf-?, IL-2, IL-6 ve IL-10 düzeylerinde ise gruplar arasında farklılık tespit edilmedi (P>0,05).
The Effects of Lycopene on Lipoprotein, Paraoxonase, Cytokine in Experimental Diabetic Rats,Yuzuncu Yıl University, Institute of Health Sciences, Ph.D Thesis in Department of Biochemistry, Van, 2012.This study was conducted with the purpose of researching the effect of lycopene application on lipoprotein, paraoxonase and cytokine that are projected to be used in the diagnosis and treatment of diabetes by making an experimental diabetes. In this study, 28 male Wistar-Albino rats that are 7-8 weeks with 250-300 gr. weights were used and they were randomly seperated into four groups each consisting of 7 rats: a control group, a diabetes group, a diabetes-lycopene group and a lycopene group. To make up an experimental diabetes, the streptozocin intraperitoneal (STZ) solved in the cold citrate tampon at the 45 mg/kg level was applied. Lycopene, solved in the corn oil, was applied orally with gavage to the diabetes-lycopene group as 10 mg/kg/day. At the end of one month trial period, under ether anaesthesia with jelly tubes blood samples were taken from rats? hearts. The blood samples were centrifuged and serum was obtained. From the serum samples, HbA1c, paraoxonase activity, lipoprotein levels and cytokines were determined. According to the analysis results, HbA1c level in the diabetes group was the highest with regard to the control group, in the diabetes-lycopene group it was close to the control group, in the lycopene group it was detected that the result was a little higher than control group (p<0,001). The paraoxonase activity with regard to the control group was at the lowest level in the diabetes group, it rose a little in the diabetes-lycopene group, in the lycopene group a significant rise was detected (p<0,001). At the triglyceride level, with regard to the control group, in all the groups a significant rise occurred (P?0,001). At the cholesterol level, with regard to the control group, a decline was observed in the other groups (P<0,05). At the VLDL level, with regard to the control group, a significant rise was observed in the other groups (P<0,05). At the HDL (P<0,001) and LDL (P<0,05) levels, with regard to the control group, a significant decline was observed in the other groups. At the Tnf-?, IL-2, IL-6 and IL-10 levels no difference was found (P>0,05).
