Babesiosisli Köpeklerde Tiyol-disülfit Dengesinin, Total Oksidan-antioksidan ve İskemi Modifiye Albümin Seviyelerinin Araştırılması

Abstract
Bu çalışma babesiosisli köpeklerde oksidatif stresin değerlendirilmesinde total antioksidan kapasite (TAK), total oksidan kapasite (TOK), oksidatif stres indeksi (OSİ), tiyol/disülfit denge (TDD), albümin (ALB) ve iskemi modifiye albümin (İMA) parametrelerinin tanısal etkinliklerini ortaya koymak için planlandı. Çalışmada klinik ve parazitolojik (ELISA) olarak babesiosis tanısı konulan 7 adet köpek babesiosisli grubu, 7 adet sağlıklı köpekte kontrol grubunu oluşturdu. Her iki gruptan kan örnekleri alındı. Kan serumlarında TAK ve TOK seviyeleri ticari kit kullanılarak, total tiyol (TT) ve nativ tiyol (NT) seviyeleri metot kullanılarak ELISA cihazında ölçüldü. İMA seviyesi metot kullanılarak spektrofotometre cihazında ölçüldü. ALB seviyesi ticari kit kullanılarak modüler oto analizör cihazında ölçüldü. OSİ ve disulfit (Ds) seviyeleri formül kullanılarak hesaplandı. Kontrol grubu ile karşılaştırıldığında babesiosisli grupta istatistiki olarak anlamlı olacak şekilde TAK ve ALB seviyelerinin azaldığı, bununla birlikte TOK ve OSİ seviyelerinin arttığı tespit edildi (p<0,05). ). NT, TT, Ds ve İMA seviyeleri ile NT/TT, Ds/TT ve Ds/NT oranları açısından gruplar arasında fark olmadığı belirlendi (p˃0,05). TAK ile OSİ arasında ve Ds/NT ile NT, TT ve NT/TT arasında anlamlı negatif korelasyon olduğu bulundu (sırasıyla p<0,05; p<0,001). NT ile TT ve NT/TT arasında ve TT ile NT/TT arasında anlamlı pozitif korelasyon olduğu belirlendi (p<0,001). TAK, TOK, OSİ ve ALB'nin ROC eğrisi altında kalan alanları sırasıyla 0,918, 0,796, 0,980 ve 1 olarak tespit edildi (p<0,05). Elde edilen sonuçlar babesiosisli köpeklerde artan oksidatif stresi değerlendirmede TAK, TOK, OSİ ve ALB'nin tanısal performansa sahip olduğunu göstermektedir.
This study aimed to investigate the diagnostic value of total antioxidant capacity (TAC), total oxidant capacity (TOC), oxidative stress index (OSI), thiol/disulfide homeostasis (TDH), ischemia modified albumin (IMA) and albumin (ALB) levels in evaluating oxidative stress in dogs with babesiosis. The babesiosis group included the 7 dogs clinically and parasitologically (ELISA) diagnosed with babesiosis while 7 healthy dogs were assigned as the control group. Blood samples were taken from both groups. In the blood serums, commercial kit was used to determine the levels TAC with TOC and metod was used to determine levels total thiol (TT) with native thiol (NT) in ELISA device. IMA level was measured in spectrophotometer device by using of the method. ALB level was measured on modular auto analyzer device using commercial kit. OSI and disulfide (Ds) levels were calculated using the formula. Compared with the control group, it was detected that TAC and ALB levels were significantly decreased, at the same time TOC and OSI levels were significantly increased in the babesiosis group (p<0,05). It was determined that there was no difference between the groups in terms of NT, TT, Ds and IMA levels and NT/TT, Ds/TT and Ds/NT ratios (p˃0,05). A significant negative correlation was found between TAC with OSI and between Ds/NT with NT, TT and NT/TT (respectively P<0,05; p<0,001). It was determined that there was a significant positive correlation between NT with TT and NT/TT and between TT with NT/TT. The areas under the ROC curve of TAC, OSİ and ALB were determined as 0,918, 0,796, 0,980 and 1 respectively (P<0,05). The results show that TAC, TOC, OSI, and ALB have diagnostic performance in assessing increased oxidative stress in dogs with babesiosis.
