Morphological and Histometrical Studies on Distribution and Heterogeneity of Mast Cells in Goose (Anser Anser) Digestive Tract
Abstract
Bu çalışma kaz sindirim kanalında bulunan mast hücrelerinin dağılımı ve heterojenitesini belirlemek amacıyla yapıldı. Çalışmada kullanılan doku örnekleri, Van' da faaliyet gösteren özel bir firmanın kanatlı mezbahasında (Ruhsat No: 577) usulune göre kesilen 6 adet kaz'dan (anser anser) temin edildi. Kazların sindirim kanalında bulunan özofagus, kursak, ön mide, taşlık, duodenum, jejunum, ileum, sekum ve kolondan uygun büyüklükte parçalar alındı. Alınan örnekler BLA (Basic Lead Acetate-Mota), Carnoy ve IFAA (İzotonik Formaldehit Asetic Asit) tespit solüsyonlarında tespit edildikten sonra rutin doku takibi yapılarak paraplast ile bloklandı. 6 μm kalınlığında alınan kesitler % 0,5' lik Toluidine blue ve Alcian blue/Safranin O kombine boyalarında boyandılar. Toluidine blue ile boyanan kesitlerde lamina propriya, submukoza, tunika muskularis + tunika serozada 40' lık objektif ile 1 mm2' lik alandaki mast hücre sayıları belirlendi. Tespitler arası fark istatistiksel olarak önemli bulunmamakla beraber sayısal olarak en fazla mast hücresine sekumda, en az mast hücresine ise taşlıkta rastlandı. Toluidine blue ile boyanan kesitlerde sayısal veriler için en uygun tespitin IFAA olduğu, granül sayısını belirlemek içinde en uygun tespitin BLA olduğu saptandı. Alcian blue/Safranin O kombine boyamasında ise tüm organlarda AB (+), SO (+) ve miks granüllü mast hücrelerine rastlandı.
This study was performed to determine mast cell distribution and heterogeneity in the digestive tract of the goose. The tissue samples were used in the study which were provided from 6 geese excised duly abattoir of winged of a special company in business in Van (Permit Number: 577). Sufficent amount of tissue pieces were taken from oesephagus, craw, proventriculus, gizzard, duodenum, jejunum, ileum,caeum and colon of digestive tract of goose. After the samples were fixed in BLA (Basic Lead Acetate-Mota), Carnoy, IFAA (Isotonic Formaldehyde acetic Acid) fixative solutions, they were follewed up in routine procedure until they were blocked with paraplast. The samples sections (6 μm) were stained with 0,5% toluidine blue and alcian blue-Safranin O. Number of mast cells determined by microscope objective (x40) in 1 mm² area of lamina propria, submucosa, tunica muscularis+tunica serosa in sections which were stained with toluidine blue. Although there is no statistically significant difference between results, the most mast cells were detected in caecum and the least mast cells were found in gizzard. The appropriate fixation technique to obtain numerical results for sections that were stained with toluidine blue was IFAA on the other hand BLA was the suitable stain for detection on granule numbers. AB (+), SO (+) and mix granules mast cells were obserwed in all organs with combined staining of Alcian blue and Safranin O.
