Abstract
Demirol M, Tüberküloz tanısında Ehrlich-Ziehl-Neelsen ve Auramine-Rhodamine boyama yöntemlerinin karşılaştırılması, Y.Y.Ü. Sağlık Bilimleri Enstitüsü Tıbbi Mikrobiyoloji Anabilim Dalı, Yüksek Lisans Tezi, Van, 2010. Bu çalışmada, 2009-2010 yılları arasında Yüzüncü Yıl Üniversitesi Tıp Fakültesi Mikrobiyoloji Laboratuvarı'na direkt mikroskopi ve kültür işlemleri için gönderilen 197 balgam örneği ile Van Verem Savaş Dispanseri'ne başvuran hastalardan alınan 94 balgam örneği olmak üzere toplam 291 balgam örneği alınarak, Ehrlich-Ziehl-Neelsen (EZN) boyama yöntemi, Auramine-Rhodamine (A-R) boyama yöntemi, Bact/Alert 3D sıvı besiyeri kültür sistemi ve Löwenstein-Jensen katı besiyeri kültür yöntemi ile çalışılmış ve aside dirençli basilleri saptamada bu testlerin etkinliği araştırılmıştır. 291 balgam örneğinden 19 (% 6.5) tanesi kültür, 18 (% 6.1) tanesi Auramine-Rhodamine boyama yöntemi, 13 (% 4.9) tanesi ise EZN boyama yöntemi ile pozitif olarak saptanmıştır. Boyama yöntemlerinin her ikisinin de kültür yöntemi referans test alınarak yapılan karşılaştırılmasında, EZN boyama yöntemi için duyarlılık, özgüllük, pozitif prediktif değer ve negatif prediktif değer sırasıyla % 68.4, % 99.6, % 92.8 ve % 97.7 olarak bulunmuştur. Auramine-Rhodamine boyama yöntemi için duyarlılık, özgüllük, pozitif prediktif değer ve negatif prediktif değeri ise sırasıyla % 94.7, % 98, % 78.2 ve % 99.6 olarak bulunmuştur. Sonuç olarak her iki yöntemin birlikte kullanılmasının pozitif örneklerin hızlı ve doğru tespitinde avantaj sağlayacağı düşünülmektedir.

In order to evaluate the effectiveness to detect the acid-resistant rods.From these 291 sputum specimens; 19 (% 6.5) cultures, 18 (% 6.1) A-R stainings, 13 (% 4.9) EZN stainings were detected to be positive.The comparison of both staining techniques with the referance culturing method set forth that the sensitivity,specifity,positive predictive and negative predictive values are % 68.4, % 99.6, % 92.8 and % 97.7 for EZN respectively and % 94.7, % 98, % 78.2 and % 99.6 for AR.As a result, it was concluded to use both techniques together to achieve faster and more reliable results to detect positive specimens.
