A Reserach on the Effects of Different Doses of Streptozotocin on the Prosthesis Stroma-Paranchyma Ratio Nerve of Rat by Sterological Methods
Abstract
Bu çalışma; farklı dozlarda uygulanan streptozotosin (STZ)'in sıçan prostat stroma ve parankim doku üzerine etkilerinin streolojik metodlarla araştırılmasıdır. Çalışmada 3-4 aylık, erişkin, ortalama 250 gr ağırlığında 20 adet erkek sıçan kullanıldı. Her grupta 5'er adet sıçan olmak üzere toplam 4 grup oluşturuldu. Deneyden önce grupların ağırlıkları ve kan glikoz değerleri ile deneyden sonra kan glikoz değerleri ve haftada bir olmak üzere dört hafta ağırlıkları ölçüldü. STZ 45mg/kg ve 65mg/kg intraperitonal olarak uygulanıp diyabet oluşturuldu. Toplam dört hafta olan deney süresinin sonunda kontrol ve denek gruplarına ait tüm denekler, ketalar 50 mg/kg anestezisi altında açılarak perfüzyon uygulandı. Prostatın ışık mikroskobik incelemeleri için uygun takipleri yapıldı. Hacim için masson trikrom boyası uygulanan kesitler, ışık mikroskobu kullanılarak görüntüleri kaydedildi. Prostat dokusunun total hacmi için Cavalieri Prensibi kullanılarak her bir grup için ayrı hesaplandı. Elde edilen veriler istatiksel analizler için Kruksal Wallis, Tukey çoklu testleri kullanıldı. Alınan kesitlerde stroma-parankim volüm oranı, bileşik alan ölçüm cetveliyle ölçüldü. Stereolojik inceleme sonucu Serum Fizyolojik grubu ve kontrol grubunda parankim doku %56 bağ doku %44, denek grubunda ise parankim doku %55 bağ doku ise %45 bulundu. Sonuç olarak farklı dozlarda uygulanan streptozotosinin denek ve kontrol grubunda sıçan prostat stroma-parankim oranına etkisinin istatistiksel açıdan anlamlı olduğu (P<0.05) gözlendi.Anahtar Kelimeler: Prostat, diyabet, sıçan, stereoloji, streptozotosin

Four groups were made which have five rats in each group. Before the experiment weights and blood groups of the rats,after the experiment blood glucose values and weights were measured in four week for once a week. Diabetes is made by intraperitonael application of STZ 45mg/kg and 65mg/kg. After four week which was the period of the experiment, all rats were perfused following kethalar anesthesia (50 mg/kg). Prosthesis was prepared for light microscopik investigations. Sectons of the prosthesis were stained with Masson tricrome stain for volüme and Hematoksilen-Eozin for counts and photographed by light microscope. Total volüme of the prosthesis tissue, rates of parankim and stroma cell were calculated by disector-cavalieri treated and principle of Cavalier and volüm fraction for each groups. Obtained values were evaluated by statistical methods which were Kruskal Wallis and Tucay multiple test. Stroma parenchyma volüme ratio was measured with combined field scale. At the end of stereological examination there were %56 parenchyma tissue and %44 connective tissue in the control and SF groups while there %55 parenchyma tissue and %45 connectice tissue in the in the experimental group. As a result it was found that effects of different doses of streptozotocin caused a statistically significant change in the old rat prosthesis stroma-parenchyma ratio compared to the control animals (P<0.05).Key word: Prosthesis, diabetes, rat, stereological, streptozotocin.
