Nvestigation of Total Sulphydryl and Protein Carbonyl Levels in Spina Bifida Aperta Patients

Abstract

Bu çalışmada, spina bifida aperta bulunan çocuklarda ve kontrol grubunda serum protein karbonil ve serum total sülfihidril düzeylerinin araştırılması ve bu parametrelerin spina bifida aperta patogenezindeki rolünün araştırılması amaçlandı. Yöntem: Çalışmaya, spina bifida aperta tanısı ile takip edilen 45 hasta ve bu hastalığa sahip olmayan 47 kontrol dâhil edildi. Seçilen hastalardan, ailenin onamı alındıktan sonra kan örneği alınarak serum protein karbonil ve serum total sülfihidril düzeyleri çalışıldı. Ayrıca hastalarda, NTD tipi, lezyonun bulunduğu seviye, klinik bulgular ve hidrosefali cerrahisi ihtiyacıyla birlikte, serum B12 vitamini, folat ve kalsiyum düzeylerine ilişkin bilgiler toplandı. Bulgular: Serum total sülfihidril düzeyi spina bifida aperta hastalarında 0.528±0.195 nmol/mg protein olarak saptanırken kontrol grubunda 1,105±0.102 nmol/mg protein olarak belirlendi. Ayrıca, hasta grubunda serum protein karbonil düzeyi 0,428±0,009 µmol/ml iken kontrol grubunda 0,216±0,009 µmol/ml idi. Her iki parametre açısından hasta ve kontrol grupları arasında anlamlı fark bulunduğu belirlendi (p<0,001). Meningomyelosel, meningosel, myeloşizis hastaları arasında her iki parametre açısından istatistiksel olarak anlamlı fark bulunmuyordu. Sonuç: Oksidatif stresin iyi birer belirteci oldukları bilinen protein karbonil ve serum total sülfihidril düzeyleri ile yapılan bu çalışmada, spina bifida aperta hastalarında oksidatif stresin kontrol grubuna göre anlamlı olarak yüksek olduğu belirlendi.

In this study, it was aimed to investigate serum protein carbonyl and serum total sulfhydryl levels in children with spina bifida aperta and control group and to investigate the role of these parameters in the pathogenesis of spina bifida aperta. Method: Forty-five patients followed up with the diagnosis of spina bifida aperta and 47 controls who did not have this disease were included in the study. After obtaining consent from the families of the selected patients, blood samples were taken, and serum protein carbonyl and serum total sulfhydryl levels were studied. In addition, information on NTD type, level of lesion, clinical findings and need for hydrocephalus surgery, as well as serum vitamin B12, folate, and calcium levels were collected. Results: The serum total sulfhydryl level was 0.528±0.195 nmol/mg protein in patients with spina bifida aperta, while it was 1.105±0.102 nmol/mg protein in the control group. In addition, while the serum protein carbonyl level was 0.428±0.009 µmol/ml in the patient group, it was 0.216±0.009 µmol/ml in the control group. It was determined that there was a significant difference between the patient and control groups in terms of both parameters (p<0.001). There was no statistically significant difference between meningocele, meningocele, and myeloschisis patients in terms of both parameters. Conclusion: In this study conducted with protein carbonyl and serum total sulfhydryl levels, which are known to be good markers of oxidative stress, it was determined that oxidative stress was significantly higher in spina bifida aperta patients compared to the control group.
