Definition of Genetic Structure With Method Pcr on Sheeps With Hidatidozis in the Province Van
Abstract
E. granulasus; yüksek derecede genetik farklılık gösteren kistik ekinokokkus'a sebep olan bir ajandır. Suşların belirlenmesi; her endemik alanda önleyici ve hastalığın kontrol altına alınması için yeni stratejilerin belirlenmesinde önemlidir. Van'ın farklı bölgelerinde yetiştirilerek kesim için mezbahaya getirilen koyunlardan toplanan 53 hidatik kist izolatı, polimeraz zincir reaksiyonu (PCR) ve nested polimeraz zincir reaksiyonu (Nested-PCR) teknikleriyle analizi yapıldı. 53 koyundan DNA ekstrakte edilerek, DNASIS kullanılarak primerler, internal trankript spacer 1 (ITS1) bölgesinden dizayn edildi. Bunları takiben 30 E. granulosus nükleotid dizisi gen bankasından elde edildi. Koyun kökenli genomik DNA'nın ITS1 bölgesinin moleküler analizinin sonuçları, morfolojik bulgularla uyumlu olduğu gözlemlendi. Bulgular; yeni ve E. granulosus'a özgü olan bu primerlerin, ekinokokkus üzerine yapılan moleküler çalışmalarda amplifiye edilen DNA'da uygulanabilir olduğunu göstermektedir. Bu izolatlar, ülkemizde kistik ekinokokkosis'in insan ve hayvan sağlığı için bir risk faktörü olarak rol alabileceğini göstermektedir.
Echinocuccus granulosus is an agent causing cyttic echinococcosis with a high degree of genetic diversity. Identification of strains are important for determination of new strategies to control and prevent the diseases at each endemic region. 53 hydatic cyst isolates collected from sheep brought to the slaughterhouses to slaughter reared at different areas of Van were analyzed by polymerase chain reaction (PCR) and nested polimerase chain reaction (nested-PCR) techniques. After extracting from DNA 53 sheep primers were designed by using DNASIS at Internal Transkript Spacer 1(ITS1).30 E.granulosus nucleotide sequence was obtained from gene bank. The moleculer analysis results of the ITS1 region of genomic DNA with sheep origin was consistent with morfological findings. The results indiated that this new and specific E.granulosus primers can be applied at amplified DNA for molecular studies on echinococcus. These isolates showed that cystic echinococcosis can act as a risk factor for human and animal health in our country.
