Abstract
Atış E, Farklı Dozlarda Uygulanan Streptozotosinin Sıçan Pankreas Langerhans Adacık Volümüne Etkilerinin Stereolojik Metotlarla Araştırılması, Y.Y.Ü. Sağlık Bilimleri Enstitüsü Tıbbi Histoloji ve Embriyoloji Anabilim Dalı Yüksek Lisans Tezi, Van, 2010. Diyabetin neden olduğu komplikasyonların incelenmesi ve tedavi yaklaşımlarının belirlenmesinde deneysel diyabet modelleri önemli bir yer tutmaktadır. Bu çalışmanın amacı; sıçanlara farklı dozlarda uygulanan streptozotosin (STZ)'in pankreas langerhans adacık volümü morfometrisi üzerine etkilerini stereolojik yöntemler kullanarak araştırmaktır. Çalışmamızda 3?4 aylık, erişkin, ortalama 250g ağırlığında 20 adet erkek sıçan kullanıldı. Her grupta 5'er adet sıçan olmak üzere toplam 4 grup oluşturuldu. Deneyin ikinci ve onaltıncı günlerinde tüm deney gruplarının ağırlıkları ve kan glikoz değerleri ölçüldü. STZ 45mg ve 55mg intraperitonal olarak uygulanıp diyabet oluşturuldu. Toplam 17 gün olan deney süresinin sonunda kontrol ve denek gruplara ait tüm sıçanlar, ketalar 50 mg/kg anestezisi altında açılarak perfüzyon uygulandı. Pankreasların ışık mikroskobik incelemeler için uygun takipleri yapıldı. Hematoksilen - Eosin boyası uygulanan kesitteki Pankreas ve Langerhans adacıkları toplam volümü, hacim oranları, her bir grup için elde edildi. Işık mikroskobu kullanılarak fotoğrafları çekilen pankreas adacıklarının toplam alanlarının oranları toplam hacimle çarpılarak hesaplama yolu ile ölçüldü. Elde edilen veriler istatistiksel olarak değerlendirildi. İstatiksel analizler için Kruskal Wallis, Tukey çoklu testleri kullanıldı. Tüm gruplarda Pankreas volümünde herhangi bir değişiklik gözlenmedi (P>0.05). Her iki diyabet grubunda Langerhans adacıkları volümünde azalmanın anlamlı olduğu tespit edildi (P< 0.05).Anahtar Kelimeler: Diyabet, streptozotosin, sıçan, farklı doz, stereoloji.

Experimental diabetes models play important roles in determining the complications caused by diabetes and treatment approaches. The purpose of this study was to investigate the effects of different doses of streptozotocin (STZ) by stereological methods on pancretic langerhans islet volume of rats. Twenty adult male rats aging 3-4 months and weighing approximately 250 g were used in the study. Rats were divided into 4 groups, each containing 5 animals. Second and sixteenth days of experiment, weights and blood glucose levels were measured in groups. 45 mg and 55 mg STZ were injected intraperitoneally to produce diabetes. At the end of the experimental period of 17 days, perfusion was performed under ketalar anesthesia (50 mg/kg) in all rats of the control and treatment groups. Appropriate follow-up of pancreas for the light microscopic examinations were performed. The total volume and volume ratios of pancreas and the islets of Langerhans were obtained for each group. The total area ratios of pancreas islets photographed by light microscopy were measured by multiplying with total volume. The obtained data were evaluated by statistically. Kruskal Wallis and Tukey's multiple tests were used. There was no change (P>0.05) in the volume of pancreas in all groups. Significant decreases (P< 0.05) were found in the volume of Langerhans islets in both diabetic groups.Key Words: Diabetes, streptozotocin, rats, different doses, stereology.
