Diagnosis, Distribution and Density of Eurygaster Spp. in Wheat Fields of Southeastern Anatolia Region by Morphologic and Molecular Characterization Methods
Abstract
Bu çalışmada, Güneydoğu Anadolu bölgesindeki buğday ekim alanlarında Eurygaster spp'nin morfolojik ve moleküler yöntemler kullanılarak tanımlanması, dağılımı ve popülasyon yoğunluğu belirlenmiştir. Bu bağlamda çalışma bölgesindeki tüm iller ve bu illere bağlı buğday üretiminin yoğun olduğu toplam 54 ilçeden 76 tarladan toplanan 309 adet örnek incelenmiştir. Morfolojik olarak erkek bireylerin genital bölge preparasyonu ile tür teşhisi yapılmıştır. Moleküler tür teşhisinde ise mitokondriyal DNA'nın COI gen bölgesi PZR ile çoğaltılarak diziler elde edilmiştir. Elde edilen bu diziler gen bankasında BLAST edilerek tür teşhisi tamamlanmıştır. Yapılan analizler sonucunda sünenin Güneydoğu Anadolu Bölgesi'ndeki tüm illerde yayılış gösterdiği görülmüştür. Bu iller arasında sünenin en fazla yoğunluk gösterdiği il Gaziantep iken en az yoğunluk ise Siirt ilinde kaydedilmiştir. Yapılan morfolojik ve moleküler teşhis sonucunda tüm örneklerin Eurygaster integriceps olduğu belirlenmiştir. Çalışmadan elde edilen sonuçlar bölgede süneye karşı yürütülecek Entegre Savaşım Yöntemleri programlarına temel bilgiler sunarken aynı zamanda elde edilen dizi sonuçları gen bankasında biyoinformatik alt yapıya katkı sağlamıştır.
In this study, the identification, distribution and population density of Eurygaster spp in wheat cultivation areas in the Southeastern Anatolia region were determined by using morphological and molecular methods. In this context, 309 samples collected from 76 fields from all provinces in the study region and 54 districts where wheat production is intense in these provinces were examined. Morphologically, the species was identified by the genital part preparation of male individuals. In molecular species identification, sequences were obtained by amplifying the COI gene region of mitochondrial DNA by PCR. Species identification was completed by BLASTing these obtained sequences in the genebank. As a result of the analyzes, it was observed that sunn pest spread in all provinces in the Southeastern Anatolia Region. Among these provinces, the province with the highest density of sunn pest was Gaziantep, while the lowest density was recorded in Siirt. As a result of the morphological and molecular diagnosis, it was determined that all samples were Eurygaster integriceps. While the results obtained from the study provided essential information for the Integrated Pest Management programs to be carried out in the region, the sequence results also contributed to the bioinformatics infrastructure in the gene bank.
