Investigation of the Effects of Proline Applications on Cadmium Toxicity in Curly Leaf Lettuce (lactuca Sativa Var. Crispa)
Abstract
Kıvırcık yaprak salata (Lactuca sativa var. Crispa) Caipira çeşidinin kullanıldığı bu çalışma ile prolin uygulamalarının bitkilerin kadmiyum stresine karşı toleransı artırıp artırmayacağı, kadmiyum stresinin bitkilerde metabolik olayları nasıl etkilediği, bitkilerin kadmiyum stresine karşı hangi tepkiler verdiğini ve prolin uygulamaları ile hangi uyum mekanizmaları geliştirdiğini anlamak amacıyla yapılmıştır. Çalışma kontrollü atmosferin (152 oC 13 saat gece- 222 oC 11 saat gündüz ve % 70nem) sağlandığı iklim odasında ve su kültüründe yapılmıştır. Fideler yeni kök oluşturup 4-5 yapraklı olduklarında, öncelikle bir grup fidenin yapraklarına stres uygulamadan 4 gün önce 5 mM, 10 mM, 15 mM prolin uygulaması püskürtülerek yapılmış ve sonrasında bitkiler 40 ppm kadmiyum stresine maruz bırakılmıştır. Diğer bir gruba ise stres uygulamadan hemen önce yine 5 mM, 10 mM, 15 mM dozlarında prolin uygulaması ile birlikte kadmiyum uygulaması yapılmıştır. Ayrıca bir grup bitkiye ise sadece kadmiyum uygulaması yapılmıştır. Yani toplamda kontrol (0 ppm kadmiyum), sadece Kadmiyum (40 ppm), stresten 4 gün önce prolin (5 mM, 10 mM, 15 mM) + kadmiyum, stresle birlikte prolin (5 mM, 10 mM, 15 mM) + kadmiyum olmak üzere sekiz farklı uygulama yapılmıştır. Elde edilen veriler doğrultusunda, Cd stresinin yol açtığı zararlı etkilerin azaltılmasında stresle birlikte 5 mM prolin ve stres öncesi 10 mM prolin dozunun nispeten olumlu etki yapabilecek yardımcı uygulama olabileceği düşünülmüştür. Bu dozda dışsal prolin uygulamalarının Cd stresi altındaki kıvırcık yaprak salata bitkisindeki büyüme parametreleri üzerine negatif etkilerinin tolere edilmesinde etkili olduğu ve dışsal prolin uygulama dozu ve zamanının önemli olduğu belirlenmiştir.
In this study, in which the curly leaf lettuce (Lactuca sativa var. crispa) Caipira cultivar was used, it was aimed to understand whether proline applications would increase the tolerance of plants to cadmium stress, how cadmium stress affected metabolic events of plants, which responses plants gave to cadmium stress and which adaptation mechanisms developed with proline applications. The study was carried out in a climate room and hydroponic culture, where a controlled atmosphere (152 oC 13 hours at night- 222 oC 11 hours during the day and 70% humidity) was provided. When the seedlings formed new roots and had 4-5 leaves, first of all, 5 mM, 10 mM, 15 mM proline application was sprayed on the leaves of a group of seedlings 4 days before applying stress, and then the plants were exposed to 40 ppm cadmium stress. To another group, just before applying stress, cadmium was applied together with proline application at 5 mM, 10 mM, 15 mM doses. In addition, only cadmium was applied to a group of plants. In the applications, eight different applications were made as Control, Cadmium only (40 ppm), proline (5 mM, 10 mM, 15 mM) + cadmium 4 days before the stress, and proline (5 mM, 10 mM, 15 mM) + cadmium with stress. In line with the data obtained; In reducing the harmful effects caused by Cd stress, it was thought that the 5 mM proline dose together with stress and proline 10 mM dose before the stress, could be an adjunct application that could have a relatively positive effect. It was determined that exogenous proline applications at this dose were effective in tolerating the negative effects on growth parameters of curly leaf lettuce plant under Cd stress and the exogenous proline application dose and time were important.
