Determination of Serum Lipid Profile and Lipoprotein Levels of Sheep With Naturally Acute Babesiosis

Abstract
Babesia ovis ile doğal enfekte olan koyunlarda kandaki lipit profilini ve lipoprotein düzeylerindeki değişimi saptamak için yapılan bu çalışmada Van ili Erciş ilçesinde yetiştirilen 3-5 yaşları arasındaki Morkaraman ırkı koyunlar materyal olarak kullanıldı. Paraziter ve klinik muayaneler sonucu sağlıklı 15 ve babesia teşhisi kondu 30 koyun kontrol ve hasta grubu olarak seçildi. Tüm hayvanlardan usulüne uygun olarak alman kan örneklerinde lipit profili ve lipoprotein düzeyleri incelendi. Serum örneklerinde trigliserit, total kolesterol, LDL ve HDL ölçümü enzimatik kolorimetrik yöntem kullanılarak Roche INTEGRA-800 cihazında çalışıldı. VLDL düzeyleri formülle hesaplandı. Serbest yağ asitleri, trigliserit, fosfolipit, kolesterol ve kolesterol esterlerinin % miktarları ince tabaka kromatografisi kullanılarak ölçüldü. Kontrol grubunda trigliserit, total kolesterol, VLDL, LDL, HDL düzeyleri sırasıyla 16.65±3.66, 65.41±3.96, 3.53±0.99, 18.41ill.99, 41.60i9.27 mg/dl olarak bulunurken, hasta gruptaki değerler yine sırasıyla 34.51il5.81, 57.12il7.65, 6.86i3.08, 14.42i8.48, 34.40Ü 1.49 mg/dl olarak saptandı. Trigliserit, VLDL ortalamaları arasındaki farklılıklar istatistiksel olarak p<0.001, HDL de ise p<0.05 düzeyinde anlamlı bulundu. Lipit miktarlarının kromatografik olarak saptanması sonucunda kontrol grubunda serbest yağ asidi, trigliserit, fosfolipit, kolesterol, kolesterol esterleri yüzdeleri sırasıyla 1.93Ü.3, 8.16i2.17, 18.72i3.46, 15.36il.26, 55.67i3.92 hasta grupta bu değerler 2.29i2.12, 14.9U5.8, 16.65±2.41, 14.96i2.17, 51.16±5.5 olarak saptandı. Trigliseritte p<0.001, kolesterol esterleri değişimlerinde p<0.01, fosfolipitte ise p<0.05 düzeyinde istatiksel önem bulundu. Sonuç olarak, babesiosisli koyunlarda glikolizise bağlı olarak trigliseritlerin karaciğerde üretiminin artması, parazitin sebep olduğu eritrosit hücre harabiyeti ve karaciğer hasarına bağlı olarak hepatositler tarafından kolesterolün normal sentezinin sağlanamaması nedeniyle düştüğü, böylece yapısında kolesterol bulunan HDL, LDL gibi lipoproteinlerin yeterince sentezlenemediği, serum VLDL düzeyinin yükselmesinin trigliseritlerin ve VLDL kolesterolün karaciğerde yapımını stimüle eden yağ doku lipolizisi nedeniyle olabileceği kanatine varılmıştır. Babesialarm gelişmesi için lipitlere ihtiyaç duyduğu, koyun babesiosisinde lipid profilinin ve lipoprotein düzeylerinin hayvanlarda prognoz açısından önemli olduğu söylenebilinir.Anahtar sözcükler: Babesia ovis, HDL, kolesterol, LDL, trigliserit, VLDL
In this study, which was done to determine the lipid profile and the variations about the levels of lipoprotein in the blood of sheeps naturally infected with Babesia ovis, 3-5 year old Morkaraman breed sheeps raised in Erciş county of Van city were used as materials. As the result of parasitic and clinical inspections, 15 healthy and 30 sheep diagnosed with Babesia were selected as control and patients groups. The levels of lipid profile and lipoprotein levels were examined in blood samples which were collected appropriately. The measurement of triglyceride, total cholesterol, LDL and HDL were studied in Roche INTEGRA-800, by using enzymatic colorimetric method. VLDL levels were calculated by formula. The percentages of free fatty acids, triglyceride, phospholipids, cholesterol and cholesteryl esters were measured by using thin layer chromatography. Triglyceride, total cholesterol, VLDL, LDL, HDL levels in the control group were found as, 16.65±3.66, 65.41±3.96, 3.53±0.99, 18.41ill.99, 41.60±9,27 mg/dl respectively; whereas the values in the patients group were detected as 34.51±15.81, 57.12±17.65, 6.86i3.08, 14.42i8.48, 34.40ill.49 mg/dl respectively. The differences between averages of triglyceride, VLDL were statistically found meaningful at the level of p<0.01, however was found meaningful at the level p<0.05 in HDL. After the chromatographic determination of lipid amounts, the percentages of free fatty acids, triglyceride, phospholipids, cholesterol, cholesterol esters in the control group were determined respectively as 1.93Ü.3, 8.16±2.17, 18.72i3.46, 15.36il.26, 55.67i3.92; same values in patients group were detected respectively as 2.29i2.12, 14.9U5.8, 16.65±2.41, 14.96±2.17, 51.16±5.5.The statistical significance was found as p<0.01 for the variations of triglyceride and cholesterol esters, and as p<0.05 for phospholipids. As a result, for Babesia ovis infected sheep, it was concluded that, the triglycerides production in the liver increased depending on the glycolysis cholesterol decreased due to the red blood cell destruction depending on parasite and the failure to provide the normal synthesis of cholesterol depending on hepatocytes caused by liver damage, in this way cholesterol structured lipoproteins like HDL and LDL were not synthesized adequate, serum VLDL levels could increase because of adipose tissue lipolysis that stimulates the production of triglycerides and VLDL cholesterol in liver. It is said to be important that lipids are needed for the development of Babesias, lipid levels in sheep babesiosis and lipoprotein levels are important for animals in terms of prognosis.Key Words: Babesia ovis, HDL, Cholesterol, LDL, triglyceride, VLDL.
