Abstract
Depo kanlarında zamana bağlı olarak birtakım değisiklikler olduğu ve kan transfüzyonundan sonra eritrositlerin yasam sürelerinin kısaldığı bilinmektedir. Bu çalısmada kan torbalarına alınan kanlardaki eritrositlerde zamana bağlı olarak lipid peroksidasyonu ve antioksidan kapasitedeki değisiklikler, ayrıca melatonin ve propofol'ün bu parametrelere etkilerinin arastırılması amaçlandı. Bu amaçla sağlıklı 10 donörden CPD'li torbalara kan alındı ve her kan üç farklı pediatrik torbaya esit olarak bölündü. Birinci grup kanlar, kontrol grubu olarak incelendi. ?kinci grup kanlara 500 pq/ml melatonin, üçüncü grup kanlara 50 ?g/ml propofol ilave edildi. Kanlar çalısma süresi boyunca +4 0C' de bekletildi. 0, 7, 14, 21 ve 28. günlerde kan torbalarından alınan numunelerde eritrosit MDA, glutatyon, SOD, katalaz ve GSH-Px ölçümleri gerçeklestirildi. Ayrıca eritrositlerin in vitro oksidasyona duyarlılıkları test edildi. Kontrol grubunda zamana bağlı olarak eritrosit MDA seviyesi ve in vitro oksidasyona duyarlılık artarken, eritrosit glutatyon, GSH-Px, SOD seviyeleri azaldı (p<0.05). Melatonin eklenen grupta ise kontrollere göre eritrosit MDA artısı daha azalmısken, glutatyon, GSH-Px ve SOD seviyeleri korunmustu. Ayrıca in vitro oksidasyona karsı duyarlılık azalmıstı. Propofol eklenen grupta ise eritrosit glutatyon ve GSH-Px seviyeleri korunurken, katalaz aktivitesi zamana bağlı olarak azaldı (p<0.05). Sonuç olarak bu çalısma, depo kanlarına melatonin ilave edilmesinin eritrosit lipid peroksidasyonunu önleyebileceğini, eritrositlerin in vitro oksidasyona dayanıklılığını artırabileceğini ve eritrosit antioksidan kapasitesini koruyabileceğini göstermistir. Donated blood undergoes some changes during the storage that shortens the life-spans of erythrocytes after transfusion. We investigated the changes in lipid peroxidation and antioxidant capacity of erythrocytes in stored bloods by time. We also tested the effects of added melatonin and propofol on these parameters. Blood samples from 10 healthy donors were taken into three CPD-containing pediatric blood bags each. One of the bags used as the control, others were added with either melatonin 500 pg/ml or propofol 50 mg/ml. The bloods were stored at + 40C during the study. The samples were taken at the beginning and days 0th, 7th, 14th, 21th, 28th to study erythrocyte malondialdehyde (MDA), glutathione, superoxide dismutase (SOD), catalase and glutathione peroxidase (GSH-Px) levels. Erythrocytes were also tested for their sensitivity to oxidation in vitro. In control group, erythrocyte MDA levels and sensitivity to oxidation in vitro were increased whereas glutathione, GSH-Px and SOD levels were decreased (p<0,005). In melatonin group, MDA accumulation were less than controls and glutathione, GSHPx and SOD levels were preserved. In propofol group glutathione and GSH-Px levels were conserved but catalase activity decreased by time (p<0,005). In this study we showed that addition of melatonin to stored whole blood could prevent lipid peroxidation, increase the resistance of erythrocytes to oxidation and preserve antioxidant capacity of erythrocytes.
