Commercial Clonal Propagation of Endemic and Endangered Bellevalia Specie Sunder in Vitro Conditions
Abstract
Türkiye endemik bitki tür ve taksonlarının yaklaşık yarısı ciddi şekilde yok olma riski altındadır. Bu nedenle, yerel germplazmı korumak çok önemlidir. Bu çalışmada dört adet endemik Bellevalia türünün 4 adet BAP ve NAA kombinasyonunu içeren MS, White ve Gamborg B5 besiyerlerinde 2 ve 3 ölçekli pul yaprak eksplantları kullanılarak başarılı bir şekilde muhafaza edilmesi ve neslinin tükenmesinin önüne geçmek amacıyla hızlı mikroçoğaltım yöntemi geliştirilmiştir. En iyi rejenerasyon BAP+NAA içeren MS ortamında izlenmiştir. Elde edilen soğanların köklenmesi için bir oksin işlemine gerek duyulmamıştır. Ancak her tür için optimum rejenerasyon hızının genotipe bağlı olduğu sonucuna varılmıştır. Elde edilen sonuçlar son zamanlarda keşfedilen ve nesli tükenmekte olan 4 adet Bellevalia türünün ilk olarak hızlı ve seri üretiminin önünü açmaktadır.
About half of the endemic plant species and taxa in Turkey are at serious risk of extinction. Therefore, it is very important to conserve the local germplasm. In this study, a rapid micropropagation method was developed for the successful preservation and extinction of four endemic Bellevalia species using 2-3 scale flake leaf explants on MS, White and Gamborg B5 media containing the combination of 4 BAP and NAA. The best regeneration was observed in MS medium containing BAP+NAA. An auxin treatment was not required for the rooting of the bulbs obtained. However, it was concluded that the optimum regeneration rate for each species depends on the genotype. The results obtained pave the way for the first rapid and mass production of 4 recently discovered and endangered Bellevalia species.
