Molecular Characterizations of Some Virus Isolates Andinvestigation of Barley Yellow Dwarf Viruses (bydvs) in the Wheatproduction Areas in Şirnak Province by Multiplex Rt-Pcr

Abstract

Şırnak ili ve ilçelerinde buğday üretim alanlarında Arpa sarı cücelik virüslerini (BYDV-PAV, BYDV-MAV, BYDV-SGV, BYDV-RMV ve CYDV-RPV) belirlemek amacı ile 2020 yılında survey çalışmaları yapılmıştır. Survey çalışması kapsamında toplanan 411 örnekte virüs tespiti için multipleks RT-PCR ve RT-PCR yöntemleri kullanılmıştır. Bölgeden toplanan 411 buğday örneğine uygulanan PCR testleri sonucu sonrası toplamda 55 (% 13.38) tane pozitif örnek tespit edilmiş ve bu örnekler içerisinde 40 tane örnekte BYDV-PAV (% 9.73), 3 örnekte CYDV-RPV (% 0.73) virüsleri tespit edilirken 12 örnekte BYDV-PAV+CYDV-RPV (% 2.91) karışık infeksiyonları tespit edilmiştir. Tespit edilen virüslerin kılıf protein gen bölgesinin karakterizasyonu için herbir virüs türünden ikişer pozitif izolat seçilerek kılıf protein gen bölgeleri karakterize edilmiştir. BYDV-PAV Şırnak 1 izolatı OL685734 ve Şırnak 4 izolatı OL685736 erişim numaralarıyla, CYDV-RPV Şırnak 12 izolatı OL685735 ve Şırnak 33 izolatı OL685737 erişim numaralarıyla Gen bankası (NCBI)'na kaydı yapılmıştır. Şırnak ili ve ilçelerinde Arpa sarı cücelik virüslerinin tespiti için yapılan bu çalışmada BYDV-MAV, BYDV-SGV ve BYDV-RMV virüsleri görülmezken, BYDVPAV ve CYDV-RPV virüsleri tespit edilip burada ilk defa raporlanmıştır.

Survey studies were carried out in 2020 in order to determine Barley yellow dwarf viruses (BYDV-PAV, BYDV-MAV, BYDV-SGV, BYDV-RMV, and CYDV-RPV) in wheat production areas in Şırnak province and its districts. Multiplex RT-PCR and RTPCR methods were used for virus detection in 411 samples collected within the scope of the survey study. As a result of PCR tests applied to 411 wheat samples collected from the region, a total of 55 (13.38%) positive samples were detected. Among these samples, BYDV-PAV (9.73%) viruses were detected in 40 samples, CYDV-RPV (0.73%) viruses were detected in 3 samples, and BYDV-PAV+CYDV-RPV (2.91%) mixed infections were detected in 12 samples. For the characterization of the coat protein gene region of the detected viruses, two positive isolates from each virus species were selected and the complete coat protein gene regions were characterized. BYDV-PAV Şırnak 1 isolate was registered in Genbank (NCBI) with accession numbers OL685734 and Şırnak 4 isolate with OL685736. On the other hand, CYDV-RPV Şırnak 12 isolate were registered in Genbank (NCBI) with OL685735 and Şırnak 33 isolate OL685737. While BYDV-MAV, BYDV-SGV, and BYDV-RMV viruses were not observed in this study for the detection of Barley yellow dwarf viruses in Şırnak province and its districts, BYDV-PAV and CYDV-RPV viruses were detected and reported here for the first time.
