Inveatigation of the Effects of Exogenous Ja, Naa and Aba on Maize Plant (Zea Mays L.) Exposed To Drought Stress
Abstract
ÖZET KURAKLIK STRESİNE MARUZ BIRAKILAN MISIR (Zea mays L.) BİTKİSİNE EKSOJEN OLARAK UYGULANAN NAFTALEN ASETİK ASİT (NAA), ABSİSİK ASİT (ABA) VE JASMONİK ASİT (JA) ' İN ETKİLERİNİN BELİRLENMESİ ÇEÇEN, Ömer Yüksek Lisans Tezi, Biyoloji Anabilim Dalı Tez Danışmanı: Doç. Dr. Peyami BATTAL Temmuz-2004, 78 sayfa Bu çalışmada, kuraklık stresine maruz bırakılan mısır (Zea mays L.) bitkisi yapraklarına eksojen olarak uygulanan Naftalen Asetik Asit (NAA), Absisik Asit(ABA) ve Jasmonic Asit (JA)' in bitkinin morfolojisine, kuru ağırlık oranlarına, stomalan üzerine, endojen hormon seviyelerine, proteinlere, klorofil ve karotenoid miktarlarına etkileri tespit edilmiştir. Hormon analizleri yüksek performanslı sıvı kromotografisi (HPLC), proteinler ise SDS-PAGE yardımı ile tayin edildi. Çalışma sonucunda ABA uygulanan bitkiler stres şartları altında iyi bir gelişim gösterdi. Ancak stres derecesinin artışı ile birlikte ABA etkisinin düştüğü tespit edildi. Stres şartlarına bağlı olarak değişik protein bantları belirlendi. NAA uygulanan ve değişik periyotlarda su verilen bitkilerde kuraklık stresinin şiddetine paralel olarak gelişimin azaldığı gözlendi. Su verilmeyen şartlarda ise NAA uygulamasının belirli kuraklık stresinde iyi gelişim gösterdiği tespit edildi. ABA ve NAA uygulanan bitkilerde, uygulanan hormon konsantrasyonu artışının gelişimi engellediği belirlendi. JA uygulanan bitkilerde ise diğer uygulamalardan farklı olarak bitkilerin bodurlaştığı ve kuraklık stresinin artışına paralel olarak JA uygulanan bitkilerin gelişimini tamamlayamadığı tespit edildi. Prof. Dr. Peyami BATTAL July-2004, 78 pages In the present study, the effects of exogenous JA, NAA and ABA on morphology, dry weight, stomata, endogenous phytohormones, chlorophyll and caretenoid levels and proteins of maize plant (Zea mays L.) exposed to drought stress were investigated. Quantitative phytohonnon analysis was performed by High Performance Liquid Chromatograph (HPLC) and protein profile was determined by Sodium Decodyl Sulphate-Poly Acrylamide Gel Electrophoresis (SDS-PAGE). According to the results of experimental analysis, exogenous ABA application improved the development of the plant under drought stress. However the effect of ABA on plant growth gradually decreased with increasing drought level. Different protein profiles were observed occured in different stress conditions. NAA application increased plant development in a short period under drought stress. NAA application under limited water utilisation, gradually reduced plant development depending on drought severity. Plant development suppressed with increasing NAA and ABA concentrations under drought stress. In contrast plant development was retained dwarf in all JA treatments. In conclusion, plants exhibited the highest tolerance to ABA application, whereas the lowest to JA application under drought stress.
