An Inrestigations on Prenatal Effects of Diclofenac Sodium of the Pregnant Rat of Morphomety of Uterus: a Streological Study

Abstract
Diklofenak sodyum, analjezik, antiinflamatuar ve antipiretik etkili bir ilaçtır. Gebelikte kullanılması halinde ise gebeliğin ve doğumun uzamasına, özellikle plasenta bariyerinden geçerek fetüse etki etmektedir. Uterus armut şeklinde bir organ olup, bir gövde (korpus), asağıda uterus kavitesinin daraldığı internal orifis ile iç ağızdan aşağıya doğru uzanan silindirik bir yapı olan serviksten olusur. Çalışmamızın amacı gebe sıçanlara uygulanan diklofenak sodyumun doğum sonrası 4 haftalık yavru uterus morfometrisi üzerine etkilerinin stereolojik yöntemlerle araştırılmasıdır. İlk olarak, bir gece boyunca sıçanlar çiftleşmeye bırakıldı. Çiftleşmenin ertesi günü vaginal plak gözlenen dişi sıçanlar, gebeliğin sıfırıncı günü olarak kabul edildi. Hayvanlar normal ışık ve karanlık siklusu ile 21±2 ºC (derecede) normal diyetle takibe alındı. Denek ve sham grubuna enjeksiyon gebeliğin beşinci gününde başlandı, hergün 15gün süreyle 1mg/kg diklofenak sodyum ve serum fizyolojik 1cc olmak üzere uygulandı. Postnatal 28. günde (3-4 hafta arası) yavruların erkek ve dişileri ayrılarak farklı kafese alındı. Çalışmamızda denek, sham ve kontrol gruplarında altışar adet dişi vistar albino cinsi toplam 18 adet sıçan kullanılmıştır. Gruplar 4 haftalık olunca derin anestezi altında perfüzyona alınarak tuba uterina ve overler beraber çıkarıldı, sonrasında uteruslar overlerden ayrıldı ve rutin histolojik takip metotlarından sonra parafine gömüldü. Dokulardan belirli aralıklarla 5 mikron kalınlığında kesitler alındı. İlk kesit rastgele seçildi, her 100. kesit çifti alındı. Kesitler, Masson trikrom ile boyandı ve Cavalieri prensibinin modifiye metodu kullanıldı. Epitel, mukoza, muskularis, seroza ve total doku volümleri noktalı alan cetveliyle ölçüldü. Stereolojik inceleme sonucu, gebelikte uygulanan diklofenak sodyumun postnatal 4 haftalık deneklerle karşılaştırıldığında, kontrol ve sham grublarında sıçan uteruslarında; tunika mukoza, tunika muskularis ve total doku volümlerine istatistiksel anlamda etkili olmadığı (P>0.05), ancak denek ve kontrol grublarının epitel ve seroza tabakaları volümlerinin sham grubuna göre azaldığı ve istatistikî açıdan anlamlı bulunduğu (P<0.05) tespit edildi.
The diclofenak sodium is analjezik, antiinflamatuar ve antipiretik a drug. During pregnancy, diclofenac sodium extends the pregnancy and the birth and since it can cross the placenta barrier it affects the fetus. The uterus is pear-shaped organ, a body (corpus), below: shrinking uterine cavity with the internal orifice extending down the inner mouth of the cervix consists of a cylindrical structure.The aim of our study was to investigate the effects of prenatally applied diclofenac sodium on the morphometry of the 4 week young uterus by stereological methods. First, the rats wre left overnight to mate. The next day when vaginal plaques were observed was considered as the day `0? of the pregnancy. Animals were kept in a room with normal ligt and dark cycles (21±2ºC) and fed with normal diet. The injections in the sham (1cc SF) and the experimenatal group (1cc; 1mg/kg) were started on the fifty day of pregnancy and applied for 15 days. On the postnatal 28th day (between 3-4 weeks) the male and female offsping were separated into different cages. In our study, 18 female Wistar albino rats were used in the experimental, sham and control groups each having 6 rats. When the animals are 4 weeks young they were perfused under deep anesthesia and the uterus was removed in to and embedded in parafin after routine histological procedures. Tissue sections of about 5 microns thick were taken at regular intervals. The first section was chosen at random and every 100th section pairs were taken. Sections were stained with masson trichrome. Modified method of Cavalieri?s principle was used for stereological analysis. Epithelium, mukoza, muscularis, seroza and total tissue volumes were measured with pointed field scale. As a result it was found that prenatally applied diclofenak sodium did not cause a statistically significant change in the 4 week young rat uterus tunica muscularis, tunica mucosa and total tissues volume compared to the control animals (P>0.05). But the volume of tunica epithelium and tunica serosa of the experiment group was decreased than those of sham. This difference between each group istatistically significant (P<0.05).
