Abstract
Bu çalışmada 2012-2014 yılları arasında Diyarbakır İli Karacadağ yöresinde, farklı sürülerde bulunan 18 baş zom koyunundan alınan kan örnekleri kullanılmıştır. Harran Üniversitesi Ziraat Fakültesi Hayvansal Biyoteknoloji Labaratuvarı'nda izole edilen DNA örnekleri 8 adet RAPD primeri ile kullanılarak işlenmiş ve RAPD-PCR bantları elde edilmiştir. Agaroz- Jel fotoğrafları incelenmiş ve elde edilen bantlar ile bantların var (1) ve yok (0) olmasına göre oluşturulan veri matrisi, PopGen32 paket programı kullanılarak, değerlendirilmiştir. Çalışmada 300-3000 bç aralığında 899 adet bant elde edilmiştir. Kullanılan 8 adet primer ile toplamda 54 lokus tespit edilmiş olup, bu lokusların 15'i polimorfik 39'u monomorfik bulunmuş, polimorfizm oranı ise % 27.8 olarak hesaplanmıştır. OPA-19 primeri ile tüm lokuslar polimorfik bulunurken, OPE-02 ve OPE-06 primerlerinde ise polimorfizm tespit edilememiştir. Genetik uzaklık, en yüksek 3 – 6 (0.189) nolu örnekler arasında, en düşük ise 4 - 5 (0.019) nolu örnekler arasında tespit edilmiştir. Genetik benzerlik değeri en az 3 - 6 nolu örnekler arasında (0.811), en çok ise 4 - 5 nolu örnekler arasında (0.981) bulunmuştur. Ortalama heterozigotluk 0.1253 olarak bulunmuştur. Sonuç olarak RAPD-PCR yöntemiyle Zom koyunlarında polimorfizm, genetik benzerlik, genetik uzaklık ve heterozigotluk gibi bazı genetik parametreler hesaplanarak DNA seviyesinde tanımlayıcı ön bilgiler elde edilmiş, bireylerin heterojen bir yapıda olduğu ve bu genetik çeşitliliğin ıslah amaçlı kullanım potansiyeline sahip olduğu söylenebilir.
In this research, blood samples taken from 18 heads of Zom sheep in different flocks in Karacadağ province of Diyarbakır between 2012-2014 were used. DNA samples, isolated in the Animal Biotechnology Laboratory at Harran at the Faculty of Agriculture, University of Harran, were processed using 8 RAPD primers and RAPD-PCR fragments were obtained. Agarose-Gel photographs were examined, and the resulting tapes and the data matrix generated according to be existing tapes (1) or none existing tape (0) were evaluated by popgen32 package program. 899 fragments were obtained in the range of 300-3000 bp in the study. Of the 54 loci identified using 8 primers, 15 and 39 loci were found as polymorphic and monomorphic, respectively. Polymorphism rate was calculated as 27.8%. All loci were found as polymophic when using OPE-19 primers whereas no polymorphism was detected at any loci when using OPE-02 or OPE-06 primers. Genetic distance was determined as the highest (0.189) among the samples 3-6, and the smallest (0.019) between the samples 4 and 5. Average heterozygosity was found as 0.1253. Genetic polymorphism and some genetic parameters, such as polymorphism, genetic similarity and distance, and heterozygotes were determined by using the RAPD methods in Zom Sheep, and some descriptive information has been obtained at the DNA level. Therefore, it can be said that genetic diversity detected among the individuals has the potential to be used for animal improvement.
