Investigation of the Effect of Ferulic Acid on Gentamicin Induced Nephrotoxicity in Rats

Abstract
Ferulik asit çeşitli meyve ve sebzelerde bulunan bir fenolik bir bileşiktir. Bu çalışma ile gentamisinle (GM) oluşturulan nefrotoksisite üzerine ferulik asitin etkisini incelemek amaçlanmıştır. Çalışmada kullanılan ratlar rastgele her biri 8 rattan oluşan 4 gruba ayrıldı: Kontrol, Ferulik Asit (FA), Gentamisin (GM), Gentamisin+ Ferulik asit (GM+FA). 8 günlük deneme süresinden 24 saat sonra kan örnekleri alındı, böbrekler histopatolojik değerlendirme için çıkarıldı. Ratların serum üre, kreatinin, ürik asit, LDH analizleri otoanalizörde, MDA, AOPP, GSH, SOD, CAT, IL-6, TNF-α analizleri ise ELISA'da yapıldı, alınan böbrek dokuları histopatolojik açıdan incelendi. GM+FA grubunda GM grubuna göre üre (p<0.001), kreatinin (p<0.001), MDA (p<0.01), AOPP (p<0.001), IL-6 (p<0.01), TNF-α (p<0.001) düzeylerinin istatistik olarak önemle azaldığı saptandı. Sonuç olarak, GM ile oluşturulan nefrotoksisitede, ferulik asit enflamasyonu azaltarak oksidatif stresin de azalmasına neden olmuştur. Bu çalışmada ferulikasitin antioksidan özelliğinden çok antienflamatuar özelliği ön plana çıkmıştır. Ferulik asitin nefrotoksik hasarı zayıflattığı, böbreği koruyucu özelliğinin bulunduğu söylenebilir.
Feruli cacid is a kind of phenolic compound that can be found in various fruits and vegetables. This study aims to investigate the effect of ferulic acid upon nefrotoxicity that is induced by gentamicin (GM). In this study, rats were separated into 4 groups such that each containing 8 randomly selected rats: Control group, Ferulic Acid (FA) group, Gentamicin (GM) group and Gentamicin+Ferulic Acid (GM+FA) group. Blood samples were collected after 24 hours following the 8-day trial period, and kidneys were taken out for histopathological evaluation. Serum urea, creatinine, uric acid and, LDH analyses were performed in auto analyser while MDA, AOPP, GSH, SOD, CAT, IL-6, TNF-α analyses were performed in ELISA, and kidney tissues were also examined histopathologically. Urea (p<0.001), creatinine (p<0.001), MDA (p<0.01), AOPP (p<0.001), IL-6 (p<0.01) and TNF-α (p<0.001) levels were found to be statistically and significantly lowered in GM+FA group when compared to GM group. As a result, ferulic acid has reduced the inflammation in nephrotoxicity induced by GM, causing decreased oxidative stress. In this study, anti inflammatory features of ferulic acid have come to the fore frontrather than the antioxidant features. We can say that ferulic acid reduces nefrotoxic damage and has protective properties for kidneys.
